The structures of three nine-residue peptide substrates that show differential kinetics of O-linked glycosylation catalyzed by distinct recombinant uridine diphosphate-N-acetylgalactosamine:polypeptide N-acetylgalactosaminyltransferases (GalNAc transferases) were investigated by NMR spectroscopy. A combined use of NMR data, molecular modeling techniques, and kinetic data may explain some structural features required for O-glycosylation of these substrates by two GalNAc transferases, GalNAc-T1 and GalNAc-T3. In the proposed model, the formation of an extended backbone structure at the threonine residue to be glycosylated is likely to enhance the O-glycosylation process. The segment of extended structure includes the reactive residue in a beta-like or an inverse gamma-turn conformation and flanking residues in a beta-strand conformation. The hydroxyl group of the threonine to be glycosylated is exposed to solvent, and both the amide proton and carbonyl oxygen of the peptide backbone are exposed to solvent. The exchange rate of the amide proton for the reactive threonine correlated well with substrate efficiency, leading us to hypothesize that this proton may serve as a donor for hydrogen bonding with the active site of the enzyme. The oxygens of the residue to be glycosylated and several flanking residues may also be involved in a set of hydrogen bonds with the GalNAc-T1 and -T3 transferases.  相似文献

The surface morphology and the optical property of sandblasted glasses and the photovoltaic property of amorphous-silicon solar cells fabricated on them were investigated. The results suggest that the cell performance is related to the surface micro roughness of glass substrate, especially the short period undulation and the surface fractal dimension. Consequently, the amorphous-silicon solar cell on the tempered glass substrate which had the short-circuit current of 14.26 mA/cm2 and the efficiency of 8.00% was obtained.  相似文献

We repaired the mitral valve in a patient with severe porcelain aorta. Significant mitral regurgitation developed in a 66-year-old woman with heavy calcification throughout the whole aorta. At operation, cardiopulmonary bypass was properly established by combined axillary and femoral arterial cannulations for sufficient systemic flow. Likewise, the combination of a superior mitral approach and profound hypothermic fibrillatory arrest in conjunction with low-flow cardiopulmonary bypass allowed us to repair the mitral valve successfully.  相似文献

The 5'-flanking region of human gamma-glutamylcysteine synthetase-heavy subunit (gamma-GCS-HS) was characterised by creating a series of chloramphenicol acetyl transferase (CAT) reporter deletion constructs. Analysis of various deleted CAT constructs revealed that a putative AP-1 consensus sequence is required to direct the constitutive and oxidant-mediated promoter activity. Gel mobility shift and mutation analysis of the sequence (-269 to -263 bp), showed binding of AP-1 is involved in the oxidant-mediated regulation of gamma-GCS-HS promoter activity.  相似文献

Effects of brain-derived neurotrophic factor (BDNF) and neurotrophin (NT)-3 on the expression of structural or synapse-associated proteins were examined in the developing rat cerebral cortex. Following ventricular administration of BDNF or NT-3 at embryonic day (E) 16, expression of microtubule-associated protein (MAP) 2 of 280 kDa was enhanced at E18 and/or E20, and threonine phosphorylation of MAP2 analogues of 120 and 66 kDa was modulated in different ways. NT-3 basically altered the distribution of MAP2 proteins at E20. These findings suggest that NT-3 and BDNF play a role in regulating production and phosphorylation of MAP2 analogues during development of the rat cerebral cortex.  相似文献

Investigation of genes, using data analysis and computer-based methods, has gained widespread attention in solving human cancer classification problem. DNA microarray gene expression datasets are readily utilized for this purpose. In this paper, we propose a feature selection method using improved regularized linear discriminant analysis technique to select important genes, crucial for human cancer classification problem. The experiment is conducted on several DNA microarray gene expression datasets and promising results are obtained when compared with several other existing feature selection methods.  相似文献

The La autoantigen (also known as SS-B), a cellular RNA binding protein, may shuttle between the nucleus and cytoplasm, but it is mainly located in the nucleus. La protein is redistributed to the cytoplasm after poliovirus infection. An in vitro translation study demonstrated that La protein stimulated the internal initiation of poliovirus translation. In the present study, a part of the La protein was shown to be cleaved in poliovirus-infected HeLa cells, and this cleavage appeared to be mediated by poliovirus-specific protease 3C (3Cpro). Truncated La protein (dl-La) was produced in vitro from recombinant La protein by cleavage with purified 3Cpro at only one Gln358-Gly359 peptide bond in the 408-amino-acid (aa) sequence of La protein. The dl-La expressed in L cells was detected in the cytoplasm. However, green fluorescence protein linked to the C-terminal 50-aa sequence of La protein was localized in the nucleus, suggesting that this C-terminal region contributes to the steady-state nuclear localization of the intact La protein in uninfected cells. The dl-La retained the enhancing activity of translation initiation driven by poliovirus RNA in rabbit reticulocyte lysates. These results suggest that La protein is cleaved by 3Cpro in the course of poliovirus infection and that the dl-La is redistributed to the cytoplasm. dl-La, as well as La protein, may play a role in stimulating the internal initiation of poliovirus translation in the cytoplasm.  相似文献

The ability of some bacteria to enhance the rate of cathodic oxygen reduction to water has been recently discovered, opening the way to an entirely renewable and environmentally friendly concept of biocathode. In this study we reveal that several mechanisms may induce catalytic effects by bacteria. These comprise mechanisms that are putatively beneficial to the bacteria as well as mechanisms which are merely side effects, including quinone autoxidation and direct O2 reduction by heme compounds. Here we showed that 1 μM of ACNQ is able to generate a significant catalytic wave for oxygen reduction, with onset at approximately 0 V vs. SHE. Similarly, adsorption of hemin on a carbon surface catalyses O2 reduction to H2O2 with an onset of +0.2 V vs. SHE. To evaluate the catalytic pathways of live cells on cathodic oxygen reduction, two species of electrochemically active bacteria were selected as pure cultures, namely Acinetobacter calcoaceticus and Shewanella putrefaciens. The former appears to exploit a self-excreted redox compound with redox characteristics matching those of pyrroloquinoline quinone (PQQ) for extracellular electron transfer. The latter appears to utilise outer membrane-bound redox compounds. Interaction of quinones and cytochromes with the membrane-bound electron transfer chain is yet to be proven.  相似文献

The tribological performance of halogen-free ionic liquids at steel–steel and diamond-like carbon (DLC)–DLC contacts was investigated. Hydrogenated amorphous carbon (a-C:H) and tetrahedral amorphous carbon (ta-C) were used as test specimens. Friction tests were carried out on steel–steel, a-C:H–a-C:H, and ta-C–ta-C contacts by using a reciprocating cylinder-on-disk tribotester lubricated with two different types of halogen-free ionic liquids: 1-ethyl-3-methylimidazolium dicyanamide ([BMIM][DCN]) and 1-butyl-3-methylimidazolium tricyanomethanide ([BMIM][TCC]). From the results of friction tests, the ta-C–ta-C tribopair lubricated with [BMIM][DCN] or [BMIM][TCC] exhibited an ultralow friction coefficient of 0.018–0.03. On the other hand, ultralow friction was not observed at the steel–steel and a-C:H–a-C:H contacts. Measurements obtained with a laser scanning microscope and an atomic force microscope (AFM) showed that a chemical reaction film, derived from the ionic liquid lubricant used, was formed on the steel surfaces. However, this chemical reaction film was not observed on either of the DLC surfaces. The AFM results showed that there were high-viscosity products on the ta-C surfaces, that the wear tracks on the ta-C surfaces exhibited low frictional properties, and that the ta-C surfaces were extremely smooth after the friction tests. Based on these results, it was concluded that an ionic liquid–derived adsorbed film formed on the ta-C surface and resulted in the ultralow friction when lubricated with a halogen-free ionic liquid.  相似文献

A practical synthesis yielding P‐perfluoroalkylated phosphines from triarylphosphines and perfluoroalkyl iodides has been developed. The photoinduced reaction involves the substitution of aryl groups on the phosphorus atom with perfluoroalkyl groups to successfully afford P‐perfluoroalkylated phosphines. In addition, the P‐perfluoroalkylated phosphines were found to promote the Cu‐free cross‐coupling reaction of acid chlorides with terminal alkynes.
