Purpose : We aimed to identify novel chemotherapy responsiveness biomarkers for osteosarcoma (OS) by investigating the global protein expression profile of 12 biopsy samples from OS patients. Experimental design : Six patients were classified as good responders and six as poor responders, according to the Huvos grading system. The protein expression profiles obtained by 2‐D DIGE consisted of 2250 protein spots. Results : Among them, we identified 55 protein spots whose intensity was significantly different (Bonferroni adjusted p‐value<0.01) between the two patient groups. Mass spectrometric protein identification demonstrated that the 55 spots corresponded to 38 distinct gene products including peroxiredoxin 2 (PRDX 2). Use of a specific antibody against PRDX 2 confirmed the differential expression of PRDX 2 between good and poor responders, while PRDX 2 levels as measured by Western blotting correlated highly with their corresponding 2‐D DIGE values. The predictive value of PRDX 2 expression was further confirmed by examining an additional four OS cases using Western blotting. Conclusions and clinical relevance : These results establish PRDX 2 as a candidate for chemotherapy responsiveness marker in OS. Measuring PRDX 2 in biopsy samples before treatment may contribute to more effective management of OS. 相似文献

A procedure is presented for designing a control system for distributed parameter systems of parabolic type based on the reduced-order decoupled state-space model obtained by a finite integral-transform technique. A Kalman filter is used as an observer to estimate the state variables, and state feedback control is performed. The method was applied to a one-dimensional heat conduction process and a moving bed adsorber. Both state estimation and control performances were satisfactory in spite of the model and parameter uncertainties. Following this controller design approach, the searching algorithms for the optimal sensors' and the optimal actuators' allocation problems were solved. These algorithms were applied to a one-dimensional heat conduction process in order to confirm the state estimator and controller performance. The fastest state estimation could be achieved by assigning the sensors at the optimal locations and the desired state distribution was realized with a few actuators located at the optimal positions. 相似文献

This paper discusses the complexity of packingk-chains (simple paths of lengthk) into an undirected graph; the chains packed must be either vertex-disjoint or edge-disjoint. Linear-time algorithms are given for both problems when the graph is a tree, and for the edge-disjoint packing problem when the graph is general andk = 2. The vertex-disjoint packing problem for general graphs is shown to be NP-complete even when the graph has maximum degree three andk = 2. The response of the structure subjected to nonstationary random vibration such as earthquake excitation is nonstationary random vibration. Calculating method for statistical characteristics of such a response is complicated. Mean square value of the response is usually used to evaluate random response. Integral of mean square value of the response corresponds to total energy of the response. In this paper, a simplified calculation method to obtain integral of mean square value of the response is proposed. As input excitation, nonstationary white noise and nonstationary filtered white noise are used. Integrals of mean square value of the response are calculated for various values of parameters. In this paper, we aim to solve the problem of resource‐constrained project scheduling with multiple modes (rc‐PSP/mM), in which multiple execution modes are available for each of the project's activity and with minimization of makespan as objective. We present a new genetic algorithm approach in order to solve this problem. In this procedure, we propose a new mutation operator that exploits a critical path and two new local search procedures, i.e. critical path improvement local search (cpiLS) and iterative forward/backward local search (ifbLS), using activity list characteristics. The cpiLS reduces the critical path and the ifbLS improves resource allocation of the schedule of rc‐PSP/mM. Also, to evaluate the proposed approach, we compare the results of the computational experiment on certain standard project instances with the several competing heuristic procedures presented in the literature, and it has been revealed that our procedure is one of the most competitive among the algorithms. © 2014 Institute of Electrical Engineers of Japan. Dynamic pH junction is an on-line preconcentration method in capillary electrophoresis (CE) based on electrokinetic focusing of weakly ionic analytes with in large sample volumes in a multisection electrolyte system. In this report, experiments and computer simulations were performed to gain a better insight of the analyte focusing mechanism when a dynamic pH junction was used. A computer program, SIMUL, was used to simulate the band-narrowing process of a group for phenol derivatives under optimized buffer conditions, which were compared with experimental results. Computer simulations revealed the formation of a sharp moving pH boundary within the sample zone causing efficient focusing of long plugs of weakly acidic analytes based on their pKa. These studies offered useful information for understanding the band-narrowing process by control of the depth and lifetime of the moving pH boundary as a function of analyte pKa, sample pH, and injection length. The change in pH of the sample within the capillary was also estimated by measuring the absorbances of an analyte at two different wave-lengths. Optimization of analyte focusing resulted in enhanced detection responses of about 60-450-fold in terms of peak heights for some phenol derivatives' relation to conventional injections. Dynamic pH junction represents a novel approach to control band dispersion (peak width) and selectivity (mobility) of specific analytes for high-resolution CE separations. 相似文献

This paper presents about conceptual designs of Advanced Recycling Reactor (ARR) focusing on enhancement in transuranics (TRU) burning and americium (Am) transmutation. The design has been conducted in the context of the Global Nuclear Energy Partnership (GNEP) seeking to close nuclear fuel cycle in ways that reduce proliferation risks, reduce the nuclear waste in the US and further improve global energy security. This study strives to enhance the TRU burning and the Am transmutation, assuming the development of related technologies in this study, while the ARR based on mature technologies was designed in the previous study. It has followed that the provided TRU burning core is designed to burn TRU at 28 kg/TWthh, by adding moderator pins of B4C (Enriched B-11) and the Am transmutation core will be able to transmute Am at 34 kg/TWthh, by locating Am blanket of AmN around the TRU burning core. It indicates that these concepts improve TRU burning by 40-50% than the previous core and can transmute Am effectively, keeping the void reactivity acceptable. 相似文献
