We describe a new cloning-free strategy to delete genes in the opportunistic pathogenic yeast Candida lusitaniae. We first constructed two ura3 Δ strains in C. lusitaniae for their use in transformation experiments. One was deleted for the entire URA3 coding sequence; the other possessed a partial deletion within the coding region, which was used to determine the minimum amount of homology required for efficient homologous recombination by double crossing-over of a linear DNA fragment restoring URA3 expression. This amount was estimated to 200 bp on each side of the DNA fragment. These data constituted the basis of the development of a strategy to construct DNA cassettes for gene deletion by a cloning-free overlapping PCR method. Two cassettes were necessary in two successive transformation steps for the complete removal of a gene of interest. As an example, we report here the deletion of the LEU2 gene. The first cassette was constituted by the URA3 gene flanked by two large fragments (500 bp) homologous to the 5' and 3' non-coding regions of LEU2. After transformation of an ura3 Δ recipient strain and integration of the cassette at the LEU2 locus, the URA3 gene was removed by a second transformation round with a DNA cassette made by the fusion between the 5' and 3' non-coding regions of the LEU2 gene. The overall procedure takes less than 2 weeks and allows the creation of a clean null mutant that retains no foreign DNA sequence integrated in its genome.  相似文献

The combination of experimental and numerical approaches is attempted to shed more light on 3D microstructural imperfections and mechanical performance of 3D printed acrylonitrile butadiene styrene parts. The starting point is the virtual building of airy structures using a reverse engineering approach. This approach combines microstructure generator, finite element model, and optimization strategy to propose virtual airy structures satisfying structural and mechanical criteria up to a desired porosity content of 60%. Optimal structures are printed using fused deposition modeling and X‐ray microtomography is used to assess all microstructural defects. Compression testing is performed for load levels above 50% of reduction in sample height. The main outcome of this work is the demonstration of small amount of process induced porosity inducing high pore connectivity. The interdependence of process induced and desired porosity reveals genuine microstructural effects that are only characteristics of 3D printed materials.

This study aims at gaining a better understanding of the microstructural features that control the mechanical and the tribological performances of WC–12 wt.% Co coatings under High Velocity Oxygen Fuel (HVOF) spraying conditions. This paper looks at the influences of the HVOF process parameters for WC–12Co material on the microstructural and the tribological behaviours of the coatings. The correlation between the coating microstructure and the wear behaviour is investigated by observing and analysing the microstructure and by studying the friction moment using enhanced statistical tool based on neural computations. According to the experimental and the numerical results, it has been shown that the spray parameters affect the phase composition, hardness and porosity of HVOF sprayed WC–12Co coatings and the correlations with HVOF process parameters are fully predictable in the steady-state regime.  相似文献

An adaptive fuzzy control approach is proposed for a class of multiple-input–multiple-output (MIMO) nonlinear systems with completely unknown non-affine functions. The global implicit function theorem is first used to prove the existence of an unknown ideal implicit controller that can achieve the control objectives. Within this scheme, fuzzy systems are employed the approximate the unknown ideal implicit controller, and robustifying control terms are used to compensate the approximation errors and external disturbances. The adjustable parameters of the used fuzzy systems are deduced from the stability analysis of the closed-loop system in the sense of Lyapunov. To show the efficiency of the proposed controllers, two simulation examples are presented.  相似文献

The major challenge of real-time Wireless Sensor Networks stills the optimization of both constraints: energy consumption, to get long network lifetime and the communication delay, to meet real-time requirements. In the context of IEEE 802.15.4/ZigBee networks, the association procedure has a direct effect on building paths optimizing those constraints. In this paper, we are interested on the definition of an ideal approach of load balancing to fairly distribute energy consumption among nodes in IEEE 802.15.4/ZigBee WSNs. This approach leads to conserve energy of each node in order to extend the network lifetime. To be closer to this ideal, we propose new dynamic association/re-association approaches allowing path alternation relative to association criteria and their threshold parameters. The implementation of those approaches in NS2 simulator highlights the efficiency of cooperative and dynamic association criteria particularly the one based on the sum of the inverses of remaining energy. Indeed, this approach gives better results with regard to energy distribution according to ideal approach which leads to a longer lifetime. The effects of the contents of lipids, pigments, α-tocopherol and phenols were studied in relation to the antioxidant capacity of five virgin olive oils obtained from five olive cultivars planted in Tunisia (Arbequina, Koroneiki, Leccino, Oueslati and Chemchali). The antioxidant capacities were evaluated by two different radical scavenging activities: radical scavenging activity by the DPPH assay (RSA-DPPH) and total antioxidant status by the ABTS test (TAA-ABTS). The highest contents of antioxidant compounds (75.96, 10.34, 6.32, 15.39 and 241.52 mg kg−1 for oleic acid, O/L ratio, carotenes, chlorophylls and total phenols, respectively) were found for the Koroneiki cultivar except for α-tocopherol and o-diphenols, which had the highest contents (369 and 160.7 mg kg−1, respectively) in the Leccino and Chemchali cultivars (cvs). Furthermore, the highest antioxidant capacity in virgin olive oil was observed in the Koroneiki cultivar (0.24 mmol TE kg−1) followed by the Chemchali and Leccino cvs (0.22 and 0.13 mmol TE kg−1) for the TAA-ABTS test. However, the RSA-DPPH activity was higher for the Chemchali cultivar (19.9%) than for the Koroneiki and Leccino cvs (18.4 and 13.5%, respectively). Correlation between these capacities and the oil composition revealed that they were mainly influenced by the carotene content, followed by chlorophyll and phenolic contents where the ABTS test was more pronounced. Then, the antioxidant capacity of the virgin olive oils was correlated with polar components and the lipid profile which are important for its shelf life.  相似文献

The effect of a rigid filler on the elastic properties of starch‐based composites is investigated. Thermomolding of the targeted composite is conducted using a starch matrix with varying silicone carbide content. Mechanical testing reveals that the composite's Young modulus cannot be rationalized using two‐phase analytical models. The effect of a weak interphase region is highlighted using a finite‐element model that assumes the generation of virtual microstructures. Numerical results are discussed by describing the influence of the structural and interphase parameters on the composite's elastic modulus. Identification of optimal interphase parameters quantitatively demonstrates the weak adhesion between intrinsic phases for all studied filler fractions.

This paper deals with a stochastic approach to evaluate the rut depth of hot and warm bituminous mixtures. First, the rutting performance of both mixtures using the French rutting tester device was evaluated. Given the random uncertainties derived from the numerous experimental measurements of the rut depth, statistical information was collected. Accordingly, the entropy maximum principle was used here to define adequate probability density function of the rut. Confidence regions with a high probability of 99% were determined for the estimation of the rut depth. In addition, comparison of mechanical and rheological results is performed with aged bitumen recovered from reclaimed asphalt and virgin bitumen to analyse the effect of ageing on bitumen viscoelastic properties. The experimental characterisation of the different binders based on rheological and conventional tests showed stiffening and hardening effects due to bitumen ageing.  相似文献
