The challenge of developing gene therapies for genetic forms of blindness is heightened by the heterogeneity of these conditions. However, mechanistic commonalities indicate key pathways that may be targeted in a gene-independent approach. Mitochondrial dysfunction and axon degeneration are common features of many neurodegenerative conditions including retinal degenerations. Here we explore the neuroprotective effect afforded by the absence of sterile alpha and Toll/interleukin-1 receptor motif-containing 1 (SARM1), a prodegenerative NADase, in a rotenone-induced mouse model of retinal ganglion cell loss and visual dysfunction. Sarm1 knockout mice retain visual function after rotenone insult, displaying preservation of photopic negative response following rotenone treatment in addition to significantly higher optokinetic response measurements than wild type mice following rotenone. Protection of spatial vision is sustained over time in both sexes and is accompanied by increased RGC survival and additionally preservation of axonal density in optic nerves of Sarm1−/− mice insulted with rotenone. Primary fibroblasts extracted from Sarm1−/− mice demonstrate an increased oxygen consumption rate relative to those from wild type mice, with significantly higher basal, maximal and spare respiratory capacity. Collectively, our data indicate that Sarm1 ablation increases mitochondrial bioenergetics and confers histological and functional protection in vivo in the mouse retina against mitochondrial dysfunction, a hallmark of many neurodegenerative conditions including a variety of ocular disorders.  相似文献

The experiments were performed to extract and purify substrates for polyphenol oxidase (PPO) from pericarp tissue of postharvest litchi fruit. Two purified PPO substrates were identified as (−)-epicatechin and procyanidin A2. The antioxidant properties of two PPO substrates were further evaluated in the present study. Variation in the content of the major substrate (−)-epicatechin of litchi fruit during storage at 25 °C was analysed using the HPLC-UV method. The results showed that (−)-epicatechin exhibited stronger antioxidant capability than procyanidin A2, in terms of reducing power and scavenging activities of DPPH radical, hydroxyl radical and superoxide radical. Furthermore, (−)-epicatechin content in pericarp tissue tended to decrease with increasing skin browning index of litchi fruit during storage at 25 °C. Thus, these two compounds can be used as potential antioxidants in litchi waste and the fresh pericarp tissue of litchi fruit exhibited a better utilisation value.  相似文献

Smart meters (SM) may provide large benefits to all stakeholders by providing the information required to implement certain sets of demand response actions. Benefits produced by SM related actions depend on the features of these actions, the system and the targeted consumer group.We first lay out an analytical framework to analyze the application of Demand Response (DR) actions. Based on this framework, we describe a way to determine the response of domestic load in a system to the implementation of DR actions. We propose determining the overall change in the consumption behavior of domestic load based on previously obtained estimates of the peak load and overall consumption reduction for different types of consumers in different types of systems resulting from the application of each set of actions. We have carried out a comprehensive literature review to provide here such estimates.We apply the analytical framework and methodology developed to characterize the reaction of consumers in the Austrian system to different SM related actions. Finally, we provide guidelines on which DR actions to implement in this system and how to implement them. The use of advanced indirect feedback on consumption behavior, critical peak prices and simple time-of-use tariffs is advocated.  相似文献

The favorable and adjustable transport properties of porous media make them suitable components in reactors used for solar energy conversion and storage processes. The directed engineering of the porous media’s morphology can significantly improve the performance of these reactors. We used a multiscale approach to characterize the changes in performance of exemplary solar fuel processing and solar power production reactors incorporating porous media as multifunctional components. The method applied uses imaging-based direct numerical simulations and digital image processing in combination with volume averaging theory to characterize the transport in porous media. Two samples with varying morphology (fibrous vs. foam) and varying size range (mm vs. μm scale), each with porosity between 0.46 and 0.84, were characterized. The obtained effective transport properties were used in continuum-scale models to quantify the performance of reactors incorporating multifunctional porous media for solar fuel processing by photoelectrochemical water splitting or power production by solar thermal processes.  相似文献

The coach–athlete relationship has been purported to be shaped by coaches’ and athletes’ self-perceptions (e.g., I trust my coach/athlete) and metaperceptions (e.g., My coach/athlete trusts me) of closeness, commitment, and complementarity (3Cs). The development of the Coach–Athlete Relationship Questionnaire (CART-Q; S. Jowett & N. Ntoumanis, 2004) has enabled the assessment of coaches’ and athletes’ self-perceptions of the relationship as defined by the 3Cs. The author conducted 2 studies to examine the factor structure and criterion-related validity of the CART-Q when its items are worded to express metaperceptions. In the 1st study, confirmatory factor analyses employing a sample of athletes (n = 201) supported the validity of a 3-dimensional model in which the factors were separate but correlated for metacloseness, metacommitment, and metacomplementarity. The 2nd study provided further evidence of validity when the 3 factors were examined relative to criterion variables with 2 independent samples: athletes (n = 189) and coaches (n = 138). These results highlight that the conceptual model from which the CART-Q was developed captures the relationship quality through athletes’ and coaches’ feelings (closeness), thoughts (commitment), and behaviors (complementarity). Eight types of PVC + CPE blends with different weight ratios (PVC/CPE = 100/0, 90/10, 80/20, 60/40, 40/60, 20/80, 10/90, and 0/100) are tested. Data on the heat conductivity, thermal diffusivity, and heat capacity of the blends investigated are reported. Primary attention is paid to the structural approach for effective elastic constants: bulk, shear, and Young's modulae. The blends are considered as random mixtures of two isotropic constituents. It is found that the elastic moduli may be well represented by the Kerner and Budiansky equations.  相似文献

The present study examined quantitative electroencephalographic (QEEG) profile for children with autistic spectrum disorder (ASD). Five-minute QEEG data were obtained from 90 normal controls (NCs) and 66 children with ASD. Spectrum analyses revealed that ASD children showed significantly less relative alpha and more relative delta than NC. Specifically, 26% of ASD children and 2% of NCs showed 1.5 SDs of relative alpha below the normative mean. Children with this QEEG profile had 17 times the risk of having ASD than those without such a profile. Sensitivity and specificity of relative alpha were 91% and 73%, respectively. This paper deals with the problem of the colorimetric fidelity of digitised colour slides of painted works of art. A six-matrix conversion model was derived that permitted the transformation of any RGB device-dependent measurement on digitised colour slides to XYZ CIE 1931 device-independent values. The model was calibrated against a reference colour chart. Eighty-one uniformly-painted colour samples were photographed together with a painting on a 4 × 5 inch colour slide that was digitised using a high resolution scanner. A 3 × 3 transformation matrix, of the corrected R, G and B values and the respective X, Y and Z tristimulus values provided by spectrophotometry, was calculated. The calculated matrix was then applied to a 13th century Byzantine fresco, captured on the same digitised slide, to transform the RGB measurements to XYZ CIE 1931 device-independent values which were then converted into the L* a* b* CIE 1976 colorimetric system.  相似文献
