Phosphatidylcholine acyltransferase (lecithin:cholesterol acyltransferase or LCAT; EC 2.3.1.43) activity was found to be present in pig ovarian follicular fluid (POFF), in addition to pig serum (PS). The cholesterol esterification rate in both POFF and PS is linear with incubation time up to 2 hr. The mean absolute rate of POFF-cholesterol esterification was 8.1±0.4 nmoles per ml per hr approximately one-fourth of that in PS. However, the fractional rate (percent of labeled cholesterol esterified per hr) of POFF-cholesterol esterification was similar to that observed in PS. There was little variation of absolute rate of cholesterol esterification in the fluid obtained from different sizes of follicles. Fatty acid or triacylglycerol did not participate in the reaction of cholesterol esterification in POFF. No appreciable change in enzymatic activity was found from storing POFF at 4 C for periods of time up to 24 hr or at −70 C up to 2 months, but activity was lost thereafter. On the other hand, PS showed a much longer period of stability (5 days at 4 C and 9 months at −70 C). A discrepancy between the fatty acid composition of cholesteryl esters formed by the LCAT reaction and the fatty acid composition at the C-2 position of phosphatidylcholine led us to propose a two-step mechanism for the LCAT reaction. It is concluded that the LCAT of POFF, as well as that of plasma, is specific for individual fatty acids rather than for the fatty acid composition of phosphatidylcholine. The fatty acid concentration of lysophosphatidylcholine decreased during prolonged incubation times (6 to 21 hr) suggesting that the increased lysophosphatidylcholine formed as a product of the LCAT reaction may be reused as substrate for the LCAT reaction or for hydrolysis by lysophosphatidylcholine hydrolase. General-base catalysis in the active site of serine proteasesis carried out by the imidazole side chain of a histidine. Duringformation of the transition state, an adjacent carboxylic acidgroup stabilizes the positive charge that forms on the general-basecatalyst and as a result contributes several orders of magnitudeto the catalytic efficiency of these enzymes. In the recentlydiscovered family of self-cleaving proteins exemplified by theLexA repressor of Escherichia coli, instead of the imidazoleof a histidine, the active-site general-base catalyst was foundto be the -amino of a lysine. The considerably higher capacityof the lysine side chain for proton acceptance raises interestingquestions concerning the role of electrostatic interactionsin the mechanism of proton transfer by this highly basic group.The negative charge elimination studies described here and theireffects on the kmax and pK of LexA self-cleavage are consistentwith a model in which electrostatic interactions between anacidic side chain and the general-base catalyst form a barrierto proton transfer. The implications are that the -amino group,unlike the imidazole group, is capable of effecting proton transferwithout the intervention of a countercharge.  相似文献

The paper examines and evaluates the transposition of live theatre performance into a digital multimedia environment utilising QuickTime (or equivalent) movie software for use on standard PCs. Writers such as Brenda Laurel have discussed the close correspondences between theatre and computers, and CD-ROM authoring software such as Director clearly adopts theatrical paradigms and terminology. The differences between attending a performance in a theatre and viewing a recording on a computer screen are outlined, and how these two experiences differ in what is expected from the audience/user. The distortions inherent in any video recording of a theatre event are explored, with particular reference to differentials in performance scale. The flickering, low-r esolution picture typifying video playback on CD-ROMs is then argued to be a positive virtue in the remediation of theatrical footage, and far preferable to full-screen video recordings. A number of reasons are proposed which explore ideas around documentary realism, visual and aesthetic convention, totemic fetishisation, and Brecht's alienation theory. It is suggested that the small-window digital movie placed within the larger multimedia screen where other graphical codes and images can co-exist and interact with the video footage, creates a dual proscenium that is poetic, evocative, and inherently theatrical.  相似文献

Anaerobic digestion is a well established technology for the reduction of organic matter and stabilization of wastewater. Biogas, a mixture of methane and carbon dioxide, is produced as a useful by-product of the process. Current solid waste management at the city of Prince George is focused on disposal of waste and not on energy recovery. Co-digestion of fresh fruit and vegetable waste with sewer sludge can improve biogas yield by increasing the load of biodegradable material. A six week full-scale project co-digesting almost 15,000 kg of supermarket waste was completed. Average daily biogas production was found to be significantly higher than in previous years. Digester operation remained stable over the course of the study as indicated by the consistently low volatile acids-to-alkalinity ratio. Undigested organic material was visible in centrifuged sludge suggesting that the waste should have been added to the primary digester to prevent short circuiting and to increase the hydraulic retention time of the freshly added waste.  相似文献

Since 2002, the Royal Air Force (RAF) has been working towards developing role-related physical tests for use as an operational fitness test (OFT). The purpose of this study was to establish reliability of the OFT (comprising four tests), investigate gym-based tests as predictors of performance and establish performance standards. Fifty-eight RAF personnel performed the OFT on three occasions. A separate cohort carried out fitness and anthropometric tests before performing the OFT, by way of establishing performance predictors. Documented evidence and views of an expert panel were used to determine OFT standards. Reliability ranged from moderate to good for three tests, with one test (Dig) showing poor reliability. The 95% limits of agreement for the prediction models ranged from good to poor (6.7-34.2%). The prediction models were not sufficiently accurate to estimate confidently OFT performance, but could be used as a guide to quantify likely outcome and training needs.  相似文献

A microfluidic valve based on electrochemical (ECM) actuation was designed, fabricated using UV-LIGA microfabrication technologies. The valve consists of an ECM actuator, polydimethylsiloxane (PDMS) membrane and a micro chamber. The flow channels and chamber are made of cured SU-8 polymer. The hydrogen gas bubbles were generated in the valve microchamber with Pt black electrodes (coated with platinum nanoparticles) and filled with 1 M of NaCl solution. The nano particles coated on the working electrode helps to boost the surface-to-volume ratio of the electrode for faster reversible electrolysis and faster valve operation. To test the functionality of the microvalve, a simple micropump based on ECM principle was also integrated in the system to deliver a microscopic volume of fluid through the valve. The experimental results have showed that an approximately 300 μm deflection of valve membrane was achieved by applying a bias voltage of ?1.5 V across the electrodes. The pressure in the valve chamber was estimated to be about 200 KPa. Experimental results proved that the valve can be easily operated by controlling the electrical signals supplied to the ECM actuators.  相似文献

The performance of five hole-transporting layers (HTLs) is investigated in both single-junction perovskite and Cu(In, Ga)Se2 (CIGSe)-perovskite tandem solar cells: nickel oxide (NiOx,), copper-doped nickel oxide (NiOx:Cu), NiOx+SAM, NiOx:Cu+SAM, and SAM, where SAM is the [2-(3,-6Dimethoxy-9H-carbazol-9yl)ethyl]phosphonic acid (MeO-2PACz) self-assembled monolayer. The performance of the devices is correlated to the charge-carrier dynamics at the HTL/perovskite interface and the limiting factors of these HTLs are analyzed by performing time-resolved and absolute photoluminescence ((Tr)PL), transient surface photovoltage (tr-SPV), and X-ray/UV photoemission spectroscopy (XPS/UPS) measurements on indium tin oxide (ITO)/HTL/perovskite and CIGSe/HTL/perovskite stacks. A high quasi-Fermi level splitting to open-circuit (QFLS-Voc) deficit is detected for the NiOx-based devices, attributed to electron trapping and poor hole extraction at the NiOx-perovskite interface and a low carrier effective lifetime in the bulk of the perovskite. Simultaneously, doping the NiOx with 2% Cu and passivating its surface with MeO-2PACz suppresses the electron trapping, enhances the holes extraction, reduces the non-radiative interfacial recombination, and improves the band alignment. Due to this superior interfacial charge-carrier dynamics, NiOx:Cu+SAM is found to be the most suitable HTL for the monolithic CIGSe-perovskite tandem devices, enabling a power-conversion efficiency (PCE) of 23.4%, Voc of 1.72V, and a fill factor (FF) of 71%, while the remaining four HTLs suffer from prominent Voc and FF losses.  相似文献

Birds are unrivaled windows into biotic processes at all levels and are proven indicators of ecological well-being. Understanding the determinants of species distributions and their dynamics is an important aspect of ecology and is critical for conservation and management. Through crowdsourcing, since 2002, the eBird project has been collecting bird observation records. These observations, together with local-scale environmental covariates such as climate, habitat, and vegetation phenology have been a valuable resource for a global community of educators, land managers, ornithologists, and conservation biologists. By associating environmental inputs with observed patterns of bird occurrence, predictive models have been developed that provide a statistical framework to harness available data for predicting species distributions and making inferences about species-habitat associations. Understanding these models, however, is challenging because they require scientists to quantify and compare multiscale spatialtemporal patterns. A large series of coordinated or sequential plots must be generated, individually programmed, and manually composed for analysis. This hampers the exploration and is a barrier to making the cross-species comparisons that are essential for coordinating conservation and extracting important ecological information. To address these limitations, as part of a collaboration among computer scientists, statisticians, biologists and ornithologists, we have developed BirdVis, an interactive visualization system that supports the analysis of spatio-temporal bird distribution models. BirdVis leverages visualization techniques and uses them in a novel way to better assist users in the exploration of interdependencies among model parameters. Furthermore, the system allows for comparative visualization through coordinated views, providing an intuitive interface to identify relevant correlations and patterns. We justify our design decisions and present case studies that show how BirdVis has helped scientists obtain new evidence for existing hypotheses, as well as formulate new hypotheses in their domain.  相似文献

Zusammenfassung  Entwicklungen im Bereich der Informations- und Kommunikationstechnologie führen derzeit zu fundamentalen Umw?lzungen der Gesellschaft. Eine Beschleunigung des Lebens und das Verschwimmen von Beruf und Privatleben sind nur zwei Beispiele hierfür. Ein Abweichen von dem gesellschaftlichen Ideal des mobilen, jungen und dynamischen Mitarbeiters erweist sich h?ufig als Weg in das Einsiedlerdasein eines Exoten. Ein Widerstreben gegen die wachsenden Begehrlichkeiten von Wirtschaft und Staat l?sst das Individuum verd?chtig erscheinen und kann zu einer weiteren Entkopplung aus der Gesellschaft führen. Eine entscheidende Aufgabe bei gesellschaftlichen Umw?lzungen ist die der Reflexion und Kritik. Diese Rolle scheint mehr und mehr den Juristen zuzukommen. Insbesondere beim Thema Datenschutz erfolgt h?ufig eine Reduktion auf die Verfassungskonformit?t einzelner staatlicher Ma?nahmen. Kritik sollte jedoch bereits weit früher geübt werden. Medien sollten diese zentrale Funktion unterstützen und der politische Diskurs sollte zu einer optimalen L?sung führen. Eine Gruppe, die in der Debatte viel beitragen k?nnte und müsste, wird h?ufig, wenn überhaupt, aufgrund ihres technischen Sachverstandes herangezogen – die Informatiker. Dies ist insbesondere verwunderlich, da Informatiker ma?geblich an der Ver?nderung der Gesellschaft beteiligt sind. Sie schaffen die technischen Grundlagen für neue Formen der Interaktion. Doch das daraus resultierende Bild der Informatik als Computerwissenschaft beraubt Informatiker ihrer gesellschaftskritischen Funktion, da sie auf die Rolle von Technikern reduziert werden. Eine Positionierung und Darstellung der Informatik als Struktur- und Koordinationswissenschaft sowie der Informatiker als interdisziplin?re Systementwickler und -analytiker würde es Informatikern erm?glichen die bedeutende Rolle der Gesellschaftskritiker einzunehmen und geh?rt zu werden.  相似文献
