Chirped gratings with very broad reflection bandwidths up to 44 nm and high reflectivity have been produced holographically in boron-codoped and standard telecommunications fibres which have been hydrogen-soaked. The role of protein kinase C (PKC) in ischemic preconditioning remains controversial because of difficulties with both its measurement and pharmacological manipulation. We investigated preconditioning in isolated neonatal rat cardiocytes by expressing constitutively active isotypes of PKC. Observations at differing durations of simulated ischemia suggested beta-galactosidase (beta-gal) activity reflected viability within transfected myocytes. Preconditioning with 90 min of ischemia significantly increased beta-gal activity and myocyte survival after 6 h of ischemia; an effect abolished by PKC inhibitors. After co-transfection with plasmids encoding beta-gal and either constitutively active mutants of PKC-delta, PKC-alpha, wild type PKC-delta, or empty vector, cardiocytes were subjected to 6 h of ischemia. Only PKC-delta, rendered constitutively active by a limited deletion within the pseudosubstrate domain, consistently increased resistance to simulated ischemia (beta-gal activity was 85.6 +/- 11.9% versus 53.7 +/- 6.5% (p 相似文献

Efficient, wide-stopband fibre transmission filters with single narrow transmission bands produced using multiply exposed chirped fibre grating resonators are reported. Tailored combinations of these structures with additional chirped gratings have achieved single passband transmission with an effective finesse of 48. Intracellular metabolism of the carcinogen chromate [Cr(VI)] produces the oxidative stress and oxidative DNA damage associated with its genotoxicity. Such oxidative stress has previously been measured by fluorescence using oxidant-sensitive dyes and attributed to the formation of reactive oxygen species (ROS). However, metabolism of Cr(VI) also produces Cr(IV) and Cr(V) which can directly damage biological macromolecules without forming ROS. We used the high-valence chromium species, bis(2-ethyl-2-hydroxybutyrato)oxochromate(V) [Cr(V)-EHBA], to test whether high-valence chromium would also react with the oxidant-sensitive dyes 2',7'-dichlorofluorescin (DCFH) and dihydrorhodamine (DHR). Cr(V)-EHBA caused both dyes to fluoresce over a wide dynamic range and under conditions which indicated that Cr(V) had reacted directly with both dyes without first forming a diffusible radical species. Dimethylthiourea (DMTU) and ethanol did not affect Cr(V)-induced fluorescence in vitro or Cr(VI)-induced fluorescence in A549 cells. Under the same conditions, ethanol and DMTU increased the extent of hydrogen peroxide-induced fluorescence. As chromium-induced fluorescence was unaffected by radical scavengers and was qualitatively different from hydrogen peroxide-induced fluorescence, we conclude that DCF and R123 fluorescence in chromate-treated A549 cells is a qualitative and cumulative measure of intracellular Cr(V) formation and not ROS.  相似文献

Nonmetallic inclusions in low-alloy steel welds have an important effect on the microstructure and properties of weld deposits. This work is an attempt at understanding the factors controlling the spatial distribution of such inclusions, with particular emphasis on the uniformity of the distribution and the effect of solidification mode during manual-metal-arc welding. The solidification mode has been controlled by using unusual combinations of base plates and experimental electrodes. It is found that the first phase to solidify (in the form of columnar grains) is delta-ferrite (δ) when a medium carbon electrode is deposited onto a low carbon substrate, but that it is austenite (γ) when a low carbon electrode is deposited onto a high carbon substrate. Relatively large inclusions have been found to position themselves preferentially, during solidification, to the columnar grain boundaries of the first phase to solidify, whether this is 8-ferrite or austenite. The results can be understood qualitatively in terms of a surface tension driven Marangoni effect, or in terms of the pushing of solid inclusions by the solidification front. Both mechanisms drive the larger inclusions into cusps in the interface while smaller ones are passively trapped. The implications of the observed nonuniform distribution of inclusions are more severe for solidification with austenite as the primary phase, since the larger inclusions are in that case located in the weakest region of the weld where they also do not contribute to the intragranular nucleation of acicular ferrite.  相似文献

A novel concept of hybrid-type fibre Bragg gratings is proposed. The complex growth behaviour of such a grating has been observed experimentally. A new dynamic model has been proposed and developed to simulate the complex and dynamic growth of hybrid-type gratings. The modelling results showed excellent agreement with the experimentally observed spectral evolution of the hybrid-type grating.  相似文献

Adipose-tissue lipolysis (assessed from glycerol release) and glucose uptake were examined in parametrial and mesenteric adipocytes prepared from control or hyperthyroid rats in relation to changes in insulin sensitivity. Basal rates of lipolysis did not differ significantly between adipose-tissue depots. Lipolysis was maximally stimulated by noradrenaline at 1 microM, half-maximal anti-lipolytic effects of insulin were observed at approximately 11 microU/ ml insulin, and half-maximal stimulation of glucose uptake was observed at approximately 16 microU/ml insulin in adipocytes from both depots. Wortmannin caused a dose-dependent inhibition of the anti-lipolytic effect of insulin (150 microU/ml) on noradrenaline-stimulated lipolysis. Half-maximal effects of wortmannin were observed at 20-40 nM. The p70S6K inhibitor rapamycin and the mitogen-activated protein kinase kinase inhibitor PD098059 had no effects on noradrenaline-stimulated lipolysis. Hyperthyroidism increased basal rates of lipolysis and the maximal response of lipolysis to noradrenaline stimulation (3.1-fold, P < 0.001 and 2.1-fold, P < 0.05 respectively) in parametrial adipocytes. Hyperthyroidism markedly blunted the sensitivity of noradrenaline-stimulated lipolysis to half-maximal suppression by insulin in both parametrial and mesenteric adipocyte depots, and noradrenaline-stimulated lipolysis at a maximal insulin concentration remained significantly higher in adipocytes prepared from hyperthyroid rats compared with controls. Hyperthyroidism had no effect on basal and little effect on insulin-stimulated glucose uptake. Tri-iodothyronine administered at a low dose selectively influenced the anti-lipolytic action of insulin in parametrial adipocytes, and led to significantly less marked elevation in plasma non-esterified fatty acid concentrations in vivo. The results demonstrate a selective effect of hyperthyroidism to impair insulin's anti-lipolytic action, and are consistent with the operation of different downstream signalling mechanisms for the effects of insulin on adipocyte glucose transport and lipolysis.  相似文献

Technical advances in the study of ice-free parts of Antarctica can provide quantitative records that are useful for constraining and refining models of ice sheet evolution and behaviour. Such records improve our understanding of system trajectory, influence the questions we ask about system stability and help to define the ice-sheet processes that are relevant on different time-scales. Here, we illustrate the contribution of cosmogenic isotope analysis of exposed bedrock surfaces and marine geophysical surveying to the understanding of Antarctic ice sheet evolution on a range of time-scales. In the Dry Valleys of East Antarctica, 3He dating of subglacial flood deposits that are now exposed on mountain summits provide evidence of an expanded and thicker Mid-Miocene ice sheet. The survival of surface boulders for approximately 14Myr, the oldest yet measured, demonstrates exceptionally low rates of subsequent erosion and points to the persistence and stability of the dry polar desert climate since that time. Increasingly, there are constraints on West Antarctic ice sheet fluctuations during Quaternary glacial cycles. In the Sarnoff Mountains of Marie Byrd Land in West Antarctica, 10Be and 26Al cosmogenic isotope analysis of glacial erratics and bedrock reveal steady thinning of the ice sheet from 10400 years ago to the present, probably as a result of grounding line retreat. In the Antarctic Peninsula, offshore analysis reveals an extensive ice sheet at the last glacial maximum. Based on radiocarbon dating, deglaciation began by 17000cal yr BP and was complete by 9500cal yr BP. Deglaciation of the west and east sides of the Antarctic Peninsula ice sheet occurred at different times and rates, but was largely complete by the Early Holocene. At that time ice shelves were less extensive on the west side of the Antarctic Peninsula than they are today. The message from the past is that individual glacier drainage basins in Antarctica respond in different and distinctive ways to global climate change, depending on the link between regional topography and climate setting.  相似文献

"Stress-regulated" mitogen-activated protein kinases (SR-MAPKs) comprise the stress-activated protein kinases (SAPKs)/c-Jun N-terminal kinases (JNKs) and the p38-MAPKs. In the perfused heart, ischemia/reperfusion activates SR-MAPKs. Although the agent(s) directly responsible is unclear, reactive oxygen species are generated during ischemia/reperfusion. We have assessed the ability of oxidative stress (as exemplified by H2O2) to activate SR-MAPKs in the perfused heart and compared it with the effect of ischemia/reperfusion. H2O2 activated both SAPKs/JNKs and p38-MAPK. Maximal activation by H2O2 in both cases was observed at 0.5 mM. Whereas activation of p38-MAPK by H2O2 was comparable to that of ischemia and ischemia/reperfusion, activation of the SAPKs/JNKs was less than that of ischemia/reperfusion. As with ischemia/reperfusion, there was minimal activation of the ERK MAPK subfamily by H2O2. MAPK-activated protein kinase 2 (MAPKAPK2), a downstream substrate of p38-MAPKs, was activated by H2O2 to a similar extent as with ischemia or ischemia/reperfusion. In all instances, activation of MAPKAPK2 in perfused hearts was inhibited by SB203580, an inhibitor of p38-MAPKs. Perfusion of hearts at high aortic pressure (20 kilopascals) also activated the SR-MAPKs and MAPKAPK2. Free radical trapping agents (dimethyl sulfoxide and N-t-butyl-alpha-phenyl nitrone) inhibited the activation of SR-MAPKs and MAPKAPK2 by ischemia/reperfusion. These data are consistent with a role for reactive oxygen species in the activation of SR-MAPKs during ischemia/reperfusion.  相似文献

We deposit dense, ordered, thin films of Au–dodecanethiol core/shell nanoparticles by the Langmuir–Schäfer (LS) printing method, and find that their resistance at ambient temperature responds selectively and sensitively to alkane odours. Response is a rapid resistance increase due to swelling, and is strongest for alkane odours where the alkane chain is similar in length to the dodecane shell. For decane odours, we find a response to concentrations as low as 15 ppm, about 600 times below the lower explosive limit. Response is weaker, but still significant, to aromatic odours (e.g. Toluene, Xylene), while potential interferants such as polar and/or hydrogen-bonding odours (e.g. alcohols, ketones, water vapour) are somewhat rejected. Resistance is weakly dependent on temperature, and recovers rapidly and completely to its original value within the error margin of measurement.  相似文献
