Galectins have been recognized as potential novel therapeutic targets for the numerous fundamental biological processes in which they are involved. Galectins are key players in homeostasis, and as such their expression and function are finely tuned in vivo. Thus, their modes of action are complex and remain largely unexplored, partly because of the lack of dedicated tools. We thus designed galectin inhibitors from a lactosamine core, functionalized at key C2 and C3′ positions by aromatic substituents to ensure both high affinity and selectivity, and equipped with a spacer that can be modified on demand to further modulate their physico‐chemical properties. As a proof‐of‐concept, galectin‐3 was selectively targeted. The efficacy of the synthesized di‐aromatic lactosamine tools was shown in cellular assays to modulate collective epithelial cell migration and to interfere with actin/cortactin localization. 相似文献

Detecting the active forms of proteases by using activity‐based probes in complex proteomes has become an intensively investigated field of research over the past years because many pathogenic conditions involve alterations in protease activities. The detection of lysosomal cysteine proteases, the cathepsins, has mostly relied on the use of probes that incorporate reactive electrophilic moieties to modify a cysteine in the active site covalently. Here we report the first example of an activity‐based probe that targets the cathepsins and incorporates a photoactivatable benzophenone group for covalent labelling. When tested on a set of five cathepsins (B, K, L, S and V), this probe selectively labelled the active site of cathepsin L. Furthermore, when tested on crude cell extracts, the probe specifically detected cathepsin L quantities as low as a few picomoles. This study suggests that photoaffinity labelling is a promising approach for developing highly selective and useful cathepsin L probes. In particular, this probe might allow the detection of small amounts of the secreted active cathepsin L form in the cellular microenvironment in vitro and ex vivo. 相似文献

In this paper we study a symbiotic aerial vehicle-ground vehicle robotic team where unmanned aerial vehicles (UAVs) are used for aerial manipulation tasks, while unmanned ground vehicles (UGVs) aid and assist them. UGV can provide a UAV with a safe landing area and transport it across large distances, while UAV can provide an additional degree of freedom for the UGV, enabling it to negotiate obstacles. We propose an overall system control framework that includes high-accuracy motion planning for each individual robot and ad-hoc decentralized mission planning for complex missions. Experimental results obtained in a mockup arena for parcel transportation scenario show that the system is able to plan and execute missions in various environments and that the obtained plans result in lower energy consumption. 相似文献

This study examines cross-cultural differences beliefs related to e-commerce use for Italy and the United States. We argue that for both cultures, the user's decision to make an online purchase is simultaneously influenced by a set of contrary factors. These include decision facilitators such as propensity to trust and institutional trust, and decision inhibitors such as perceived risk and privacy concerns. We argue that substantial cultural differences exist that affect the above factors and the relationships among them. We use Hofstede's cultural theory and Fukuyama's theory of trust and social capital, along with emic factors important for the Italian society, to develop the study's propositions. The hypotheses were empirically tested using LISREL structural equation modeling and multigroup analysis. The results revealed that the Italian society exhibited lower propensity to trust, institutional trust, privacy concerns, and higher perceived risk. The relationships between institutional trust and e-commerce use, privacy concerns and e-commerce use, and perceived risk and institutional trust are all weaker for Italy. The relationship between perceived risk and privacy concerns is stronger for Italy. The paper's major contribution is in validating an important model of e-commerce use across two cultures and showing the moderating effects of culture. 相似文献

This study explored online personal ads of 294 heterosexual and homosexual men and women in the United States through a qualitative analysis and comparison of participant-generated “personal” and “preferred partner” narratives. Nine characteristics were identified and combined into three overarching categories: physical, lifestyle, and personality characteristics. These three personal and preferred partner characteristics were examined for difference by gender, sexual orientation, age and desired relationship type of the advertisers. Main effects emerged for all four predictors, most notably for age and desired relationship type. Additionally, this study explored the possibility that personal and preferred partner narratives contained similar constellations of characteristics, finding significant correlations on all three variables, lending support for the matching hypothesis in dating partner characteristics. 相似文献

The genetic control over the corticosterone response to ethanol (EtOH) and its possible relationship to other EtOH-related traits was examined using BXD recombinant inbred (RI) strains derived from an F2 cross of C57BL/6J (B6) and DBA/2J (D2) progenitor strains. Quantitative trait locus (QTL) analysis of corticosterone levels 1 hr following EtOH suggested the influence of a single major gene on this trait. Two loci were predicted to account for 47% of the genetic variance in plasma corticosterone levels 6 hr following EtOH, whereas 3 loci were predicted to account for 78% of the genetic variance in corticosterone levels 7 hrs following EtOH. Markers associated with corticosterone levels 7 hrs following EtOH and corrected corticosterone levels 6 hrs post-EtOH overlapped with ones found to influence acute and chronic EtOH withdrawal severity, suggesting some degree of common genetic determination between these traits. Overall these results indicate that gene action significantly influences stress responsiveness and suggest possible chromosomal locations of these genes. Two experiments examined the interrelationship of subject-matter knowledge, interest, and recall in the field of human immunology/human biology and assessed cross-domain performance in the field of physics. Framed by a stage model of domain learning, cluster-analytic methods were used to group individuals on the basis of their performance on cognitive and affective measures in immunology. Results using cluster profiles were compared with prediction expected from the stage model. Performance for students in these clusters was compared with their performance on similar measures in the field of physics. In Experiment 1, 30 premedical students and 17 graduate students in educational psychology served as participants. Results revealed 3 unique clusters of students that tended to conform to prediction of the domain learning model. Individuals' knowledge, interest, and recall of immunology generally paralleled their performance in the domain of physics. Experiment 2 sought to test and extend the findings of the initial experiment. Participants were 78 undergraduate students in education. New functional elastic polyurethane foams (PUF) degradable under environmental abiotic and biotic factors, retaining all the inherent properties of the conventional foams were synthesized using isocyanate precursors based on disaccharides (DS): lactose, maltose and saccharose. It was shown by the model reactions of monosaccharide glucose, and DS lactose and saccharose, with phenylisocyanate that both the primary and secondary hydroxyls of the carbohydrates reacted to form urethanes. The main properties of DS‐based foams (PUF/DS) were found to be similar to PUF foam (matrix) prepared with conventional polyols. However, the new PUF/DS were found to undergo enhanced acid/alkaline hydrolysis and degradation compared with PUF matrix when incubated in soil. Mass losses of incubated PUF/DSs significantly exceeded the actual carbohydrate content 28.6%, and in 12 months reached 39.58 (PUF‐4), 53.31(PUF‐8), and 47.25 (PUF‐12). In contrast, under the same conditions PUF matrix lost only 2–2.5%, confirming that incorporation of natural compounds into the polymer chain impacted the degradation processes. PUF/DS were characterized by FTIR, 1H NMR, ebullioscopy, and exclusion chromatography (molecular masses and molecular mass distribution of the oligomeric model), physical and mechanical tests (density, tensile strength, relative elongation, moisture absorption, vapor permeability), morphology, and degradation in the soil. © 2015 Wiley Periodicals, Inc. A hydrostatic head viscometer and its novel viscosity equation were developed to determine flow characteristics of Newtonian and non-Newtonian fluids. The objective of this research is to test capabilities of the hydrostatic head viscometer and its novel non-Newtonian viscosity equation by characterizing rheological behaviors of well-known polyethylene oxide aqueous solutions as non-Newtonian fluids with 60 wt.% sucrose aqueous solution as a reference/calibration fluid. Non-Newtonian characteristics of 0.3–0.7 wt.% polyethylene oxide aqueous solutions were extensively investigated with the hydrostatic head viscometer and its non-Newtonian viscosity equation over a 294–306 K temperature range, a 0.14–40 Reynolds number range, and a 55–784 s?1 shear rate range at atmospheric pressure. Dynamic viscosity values of 60 wt.% sucrose aqueous solution were determined with the calibrated hydrostatic head viscometer and its Newtonian viscosity equation over a 3–5 Reynolds number range at 299.15 K and atmospheric pressure and compared with the literature dynamic viscosity value. 相似文献
