A method for the large-scale isolation of β-casein from renneted skim milk was developed. The curd from renneted skim milk was dispersed in hot (?70 °C) water to inactivate residual chymosin. The heated curd was subsequently recovered by centrifugation, resuspended in water and incubated at 5 °C, during which β-casein dissociated from the curd; the suspension was centrifuged and the aqueous phase lyophilised. The isolated protein consisted mainly of β-casein, containing a minor amount of γ-caseins and traces of other caseins. Unless chymosin was fully inactivated by heating, some β-casein was hydrolysed at the Leu192–Tyr193 bond. The yield of β-casein increased with incubation time, up to ∼20% of the β-casein present in the milk after 24 h at 5 °C. Reducing milk pH to 5.5 or 6.0, prior to renneting, caused a high level of contamination with αs-caseins. This isolation procedure can be easily scaled-up to an industrial process and the β-casein-depleted curd may be used for the manufacture of rennet casein or processed cheese.  相似文献

The influence of the microscale local mechanical properties of the interfacial transition zone (ITZ) on macro-level mechanical response and impact behavior is studied for concretes made with copper slag and gravel aggregates. 3D nanotech vertical scanning interferometry, scanning electron microscopy coupled with energy dispersive X-ray micro-analysis, digital image analysis, and 3D X-ray computed tomography were used to characterize the microstructures and the ITZs. It was deduced that a stronger and denser ITZ in the copper slag specimen would reduce its vulnerability to stiffness loss and contribute to its elastic and more ductile response under impact loading. The analysis also indicated that a significant degeneration in the pore structure of the gravel specimen associated with a relatively weaker and non-homogeneous ITZ occurred under impact. Finally, it was also concluded that increased roughness of ITZ may contribute to the load-carrying capacity of concrete under impact by improving contact point interactions and energy dissipation.  相似文献

β–SiC nanoprecipitates can be patterned in crystalline silicon with an almost monomodal size distribution by simultaneous-dual-beam of C+ and Si+ ion implantations at 550 °C. Their shape appears as spherical (average diameter ~4–5 nm) ,and they are in epitaxial relationship with the crystalline silicon matrix. The narrow size distribution follows the left wing of the carbon distribution where the nuclear ion stopping, and thus the point defect generation rate is largest. This observation allows us to conclude that the induced damage act as sinks for C atoms leading to the SiC nanoprecipitates formation centered at the maximum of the simulated damage distribution. The nuclear reaction analysis, X-ray diffraction, Raman spectroscopy, and transmission electron microscopy techniques were used to characterize the samples.  相似文献

Computational modelling is one of the most significant developments in the practice of scientific inquiry in the 20th century. During the past decade, advances in computing technologies have increased the speed of computers by a factor of 100; an increase of a factor of 1000 can be expected in the next decade. These advances have, however, come at a price, namely, radical change(s) in computer architecture. Will computational scientists and engineers be able to harness the power offered by these high-performance computers to solve the most critical problems in science and engineering? In this paper, we discuss the challenges that must be addressed if we are to realize the benefits offered by high-performance computing. The task will not be easy; it will require revision or replacement of much of the software developed for vector supercomputers as well as advances in a number of key theoretical areas. Because of the pace of computing advances, these challenges must be met by close collaboration between computational scientists, computer scientists and applied mathematicians. The effectiveness of such a multidisciplinary approach is illustrated in a brief review of NWCHEM, a general-purpose computational chemistry code designed for parallel supercomputers.  相似文献

The statistical properties of atmospheric water vapor mixing ratio (WVMR) determined as the ratio of Raman lidar signals backscattered from water vapor and nitrogen molecules are studied. It is shown that WVMR estimates can be biased by a small percentage at low signal photon-counting rates due to fluctuations in the nitrogen signal in the denominator of the ratio, the magnitude of the bias being linked to the signal-to-noise ratio of the nitrogen signal. This is particularly important when unbiased estimates are required as in the case of climate studies and global positioning system (GPS) signal calibration. Different bias corrections and a modified ratio formulation are proposed in order to correct or eliminate this bias. The method is successfully applied in processing signals obtained with an experimental Raman lidar system devoted to calibrate GPS signals for slant path delays. It is shown to reduce biases into negligible values in both WVMR and wet path delay estimates in the range interval of 0-7 km.  相似文献

Cold bitumen emulsion mixture (CBEM) is not yet widely used as a surface course around the world. In this study, 0/14-mm-size dense-graded surface course CBEMs have been investigated. The mechanical performance was evaluated in terms of stiffness modulus over 3 months and resistance to permanent deformation under three different stress levels (100, 200, 300 kPa), whilst durability evaluation was carried out in terms of resistance to moisture and frost damage. The study has also investigated the incorporation of low cement content (1%) with relatively sustainable by-product fillers, namely ground-granulated blast furnace slag (GGBS) and fly ash (FA) type 450-S on both mechanical and durability performance. A comparison has been carried out between the low and high cement content CBEM, as well as with respect to corresponding hot mix asphalt (HMA). The results revealed that the incorporation of GGBS and FA in CBEMs leads to superior performance, similar to CBEMs treated with high cement content and comparable to an equivalent HMA. Furthermore, GGBS replacement exhibited better performance than that of FA replacement. The findings suggest that the new sustainable types of CBEM can be developed for using as a surface layer for medium- to heavy-trafficked roads.  相似文献

In this study, effects of high pressure (HP) on some constituents and properties of buffalo milk were examined. HP treatment at 100-600MPa for 30 min affected casein micelle size only slightly, whereas treatment at 800 MPa increased it by approximately 35%. Levels of non-micellar alpha(S1)and beta-caseins were increased by treatment > or = 250MPa, and were highest after treatment at 400-800MPa. The level of non-micellar calcium increased with increasing pressure up to 600 MPa. The L*-value of the milk decreased gradually with increasing pressure, from approximately 82 for untreated milk to approximately 65 for milk treated at 800 MPa. Milk pH was increased by approximately 0.07 units after treatment at 100-800 MPa, with no significant difference between treatment pressures. Denaturation of alpha-lactalbumin occurred at pressures > or = 400 MPa, and reached >90% after treatment at 800 MPa, whereas beta-lactoglobulin (beta-Ig) was denatured > 100 MPa, reaching approximately 100% after treatment at 400MPa; after treatment > or = 400MPa, all beta-Ig was associated with the casein micelles. The rennet coagulation time of buffalo milk increased with increasing pressure, whereas the strength of the coagulum formed decreased after treatment at 250-800 MPa. Overall, HP treatment affected many constituents and properties of buffalo milk; some of these effects have also been observed in the milk from other species, but the extent of the effects, and the pressure at which they occurred, differed considerably.  相似文献
