The selective production of lactic acid by anaerobic acidogenesis with low pH control was examined using a chemostat culture. By decreasing culture pH to 3.5 in a chemostat culture containing mixed microbial populations for anaerobic acidogenesis, heterolactic fermentation became dominant, resulting in the selective production of lactic acid and ethanol. This phenomenon was reversible between the acidic and neutral conditions, and was not affected by the dilution rate. The extremely low pH operation was effective for selective lactic acid production in anaerobic acidogenesis.  相似文献

This study conducted in vivo and in situ experiments with rats to investigate the glucagon‐like peptide‐1 (GLP‐1) secretion in response to oral or ileal administration of α‐glucosyl‐isoquercitrin (20–40 mmol in 2 mL; Q3G), fructooligosaccharides (200 mmol in 2 mL; FOS) and Q3G + FOS. Direct effects on GLP‐1‐producing l ‐cells were also examined by an in vitro study using a murine enteroendocrine cell line, GLUTag. To evaluate the plasma GLP‐1 level, blood samples from jugular cannula for in vivo and portal cannula for in situ experiments were obtained before and after administration of Q3G, FOS, or Q3G + FOS. We found tendencies for increases but transient stimulation of GLP‐1 secretion by Q3G in in vivo and in situ experiments. Although FOS alone did not have any effects, Q3G + FOS enhanced and prolonged high plasma GLP‐1 level in both experiments. In addition, application of Q3G on GLUTag cells stimulated GLP‐1 secretion while FOS enhanced the effect of Q3G. Our results suggest that Q3G + FOS possess the potential for the management or prevention of Type 2 diabetes mellitus (T2DM) by enhancing and prolonging the GLP‐1 secretion via direct stimulation of GLP‐1 producing l ‐cell.  相似文献

A chitinase gene (Chi3K) was cloned from the genomic DNA of Vitis vinifera cv. Koshu. The structural gene comprised 891 by without introns and encoded 297 amino acids. The Chi3K product showed high similarity to the class III chitinase of V. vinifera cv. Pinot noir. Chi3K was expressed using a bacterial expression vector for purification and enzymatic characterization of its gene product. The recombinant chitinase exhibited hydrolytic activity toward glycol chitin and its optimum pH was 4.0. It also inhibited the growth of Botrytis cinerea, which causes grey mold disease in grapes.  相似文献

During a hypothetical core-disruptive accident in a sodium-cooled fast breeder reactor, degraded core materials can form debris beds on the core-support structure and/or in the lower inlet plenum of the reactor vessel from rapid quenching and fragmentation of core material pool. Coolant boiling may lead ultimately to leveling of the debris bed that is crucial to the relocation of molten core and heat-removal capability of the debris bed. In the present study, we elected to use depressurization boiling to simulate an axially increasing void distribution in the debris bed. Bottom-heating boiling was also chosen to confirm that characteristics of the self-leveling process do not depend on the boiling mode. Particle size (between 0.5 and 6 mm), shape (spherical and non-spherical), bed volume (between 5 and 8 l) and density (namely of alumina, zirconia, lead and stainless steel) along with boiling intensity and total volume were taken as experimental parameters to obtain the general characteristics of the self-leveling process. A series of experiments with simulant materials were conducted and analyzed in detail. The good concordance of the transient processes obtained from the different boiling methods sufficiently demonstrates that the present results obtained using the depressurization boiling method exhibit these general self-leveling characteristics. Detailed comparisons of deduced time variations of the inclination angle provides qualitative tendencies based on the experimental parameters considered influential to self-leveling behavior. The rationale behind the definition introduced for equivalent power density is also presented.  相似文献

SHR/NDmcr-cp (cp/cp) rats (SHR/NDcp) are an animal model of metabolic syndrome. A previous study of ours revealed drastic increases in the mass of palmitic (16:0), oleic (18:1n-9), palmitoleic (16:1n-7), cis-vaccenic (18:1n-7) and 5,8,11-eicosatrienoic acids in the liver of SHR/NDcp. However, detailed information on the class of lipid accumulated and the mechanism responsible for the overproduction of the accumulated lipid in the liver was not obtained. This study aimed to characterize the class of lipid accumulated and to explore the mechanism underlying the lipid accumulation in the liver of SHR/NDcp, in comparison with SHR/NDmcr-cp (+/+) (lean hypertensive littermates of SHR/NDcp) and Wistar Kyoto rats. In the liver of SHR/NDcp, de novo synthesis of fatty acids (16:0, 18:1n-9 and 16:1n-7) and triacylglycerol (TAG) synthesis were up-regulated and fatty acid β-oxidation was down-regulated. These perturbations of lipid metabolism caused fat accumulation in hepatocytes and accumulation of TAG, which were enriched with 16:0, 18:1n-9 and 16:1n-7, in the liver of SHR/NDcp. On the other hand, no changes were found in hepatic contents of diacylglycerol and unesterified fatty acid (FFA); among FFA, there were no differences in the hepatic concentrations of unesterified 16:0 and stearic acid between SHR/NDcp and two other groups of rats. Moreover, little change was brought about in the expression of genes responsive to endoplasmic reticulum stress in the liver of SHR/NDcp. These results may reinforce the pathophysiological role of stearoyl-CoA desaturase 1 and fatty acid elongase 6 in the liver of SHR/NDcp.  相似文献

The hydrodechlorination of CCl2F–CClF2 (CFC-113) was studied using silica- and activated carbon-supported Ru, Rh, Pd, and Pt catalysts. The activity of the catalysts changed with time-on-stream. Ru was most stable among the four noble metals and gave a simple product distribution yielding CClF=CF2 (CFC-1113) and CHClF–CClF2 (HCFC-123a) as main products. Over silica-supported Ru, CFC-1113 yield decreased gradually with time-on-stream but HCFC-123a yield did not change throughout the reaction, suggesting that these products were formed on different sites of Ru. XRD studies and temperature-programmed reduction of the spent catalyst revealed that the deactivation was caused by halogen-containing carbonaceous species accumulated on the Ru surface during the reaction. The carbonaceous species seemed to be formed on the active site for CFC-1113 formation. Because CFC-1113 selectivity was higher over the catalyst having higher Ru dispersion, it was assumed that the hydrodechlorination of CFC-113 is structure-sensitive and CFC-1113 formation is promoted by Ru having highly unsaturated coordination sphere.  相似文献

Solid oxide fuel cells (SOFCs) are being researched around the world. In Japan, a compact SOFC system with rated alternative current (AC) power of 700 W has become available on the market, since the base load electricity demand for a standard home is said to be less than 700 W AC. To improve the generating efficiency of SOFC systems in the 700-W class, we focused on thermoelectric generation (TEG) technology, since there are a lot of temperature gradients in the system. Analysis based on simulations indicated the possibility of introducing thermoelectric generation at the air preheater, steam generator, and exhaust outlet. Among these options, incorporating a TEG heat exchanger comprising multiple CoSb3/SiGe-based TEG modules into the air preheater had potential to produce additional output of 37.5 W and an improvement in generating efficiency from 46% to 48.5%. Furthermore, by introducing thermoelectric generation at the other two locations, an increase in maximum output of more than 50 W and generating efficiency of 50% can be anticipated.  相似文献

Amorphous ZrW2O8 powder and amorphous SiO2 powder were prepared by a sol–gel process as raw materials, and high-density ZrW2O8–SiO2 were successfully prepared at a much lower temperature of 923 K for a much shorter holding time of 10 min by spark plasma sintering (SPS) method rather than by conventional melt-quenching method. The relative densities of 0.85ZrW2O8–0.15SiO2 and 0.70ZrW2O8–0.30SiO2 were 99.4% and 96.6%, respectively. The combined technique of a sol–gel process and SPS should enable us to prepare the varied types of high-density composites of ZrW2O8 without severe thermal cracking caused by melt-quenching. The thermal expansion properties and dielectric properties of ZrW2O8–SiO2 were also investigated.  相似文献

Silicon nanowires (SiNWs) with a single-crystalline Si core and a thermally oxidized shell (core-shell SiNWs) were synthesized by gold-catalyzed chemical vapor deposition followed by rapid thermal oxidation. To synthesize high-quality core-shell SiNWs, the relationship between the growth parameters and the crystallinity was studied. Furthermore, the formation process of the oxide shell was analyzed in detail by transmission electron microscopy. Using SiNWs as channels, back gate-type field effect transistors (FETs) were fabricated on p-type silicon wafers. FETs with core-shell SiNWs channels exhibited smaller hysteresis in the drain current (Id) vs. gate voltage (VGS) characteristics and higher on/off drain current ratio (ION/IOFF) than those with bare SiNWs channels.  相似文献

Recent software system contain many functions to provide various services. According to this tendency, it is difficult to ensure software quality and to eliminate crucial faults by conventional software testing methods. So taking the effect of test engineer's skill on test item generation into consideration, we propose a new test item generation method, which supports the generation of test items for illegal behavior of the system. The proposed method can generate test items based on use-case analysis, deviation analysis for legal behavior, and faults tree analysis for system fault situations. Prom the results of the experimental applications of our method, we confirmed that test items for illegal behavior of a system were effectively generated, and also the proposed method could effectively assist test item generation by an engineer with low-level skill.  相似文献
