For a high-speed limiting amplifier with small phase deviation, a new design technique using a uniplanar passive balun and a narrow-band matching network is proposed. It is revealed that a device with small bias-dependence of parasitic capacitances, such as a high electron mobility transistor (HEMT), and a differential operation using passive baluns enables phase deviations at very high frequency to be reduced. Furthermore, suppressing harmonics can reduce phase deviation in a limiting amplifier, which operates in the highly nonlinear region. To verify these design techniques, two types of limiting amplifier ICs using an InP HEMT and AlGaAs/GaAs BCT technology were designed and fabricated. These IC's feature narrow-band matching circuits for input and output using transmission lines and uniplanar CPW-slot transition baluns for differential signals. Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is synthesized as a long form (L-form; 23 kDa) and a short form (S-form; 20 kDa). The L-form contains a leader sequence that is required for transport to mitochondria, whereas the S-form lacks the leader sequence. A construct encoding the leader sequence of PHGPx tagged with green fluorescent protein was used to transfect RBL-2H3 cells, and the fusion protein was transported to mitochondria. The L-form of PHGPx was identified as the mitochondrial form of PHGPx and the S-form as the non-mitochondrial form of PHGPx since preferential enrichment of mitochondria for PHGPx was detected in M15 cells that overexpressed the L-form of PHGPx, whereas no similar enrichment was detected in L9 cells that overexpressed the S-form. Cell death caused by mitochondrial injury due to potassium cyanide (KCN) or rotenone (chemical hypoxia) was considerably suppressed in the M15 cells, whereas the L9 cells and control RBL-2H3 cells (S1 cells, transfected with the vector alone) succumbed to the cytotoxic effects of KCN. Flow cytometric analysis showed that mitochondrial PHGPx suppressed the generation of hydroperoxide, the loss of mitochondrial membrane potential, and the loss of plasma membrane integrity that are induced by KCN. Mitochondrial PHGPx might prevent changes in mitochondrial functions and cell death by reducing intracellular hydroperoxides. Mitochondrial PHGPx failed to protect M15 cells from mitochondrial injury by carbonyl cyanide m-chlorophenylhydrazone, which directly reduces membrane potential without the generation of hydroperoxides. M15 cells were more resistant than L9 cells to cell death caused by direct damage to mitochondria and to extracellular oxidative stress. L9 cells were more resistant to tert-butylhydroperoxide than S1 cells, whereas resistance to t-butylhydroperoxide was even more pronounced in M15 cells than in L9 cells. These results suggest that mitochondria might be a target for intracellular and extracellular oxidative stress and that mitochondrial PHGPx, as distinct form non-mitochondrial PHGPx, might play a primary role in protecting cells from oxidative stress. 相似文献

Ion-induced photon emission under heavy-ion bombardment has been studied over an energy range from near-infrared to vacuum-ultraviolet. Time-resolved optical devices with fast-response CCD cameras detected photons from a-SiO2 under implantation with negative Cu ions of 60 keV, up to a dose rate of 100 μA/cm2. The ion-induced photon spectra consisted of sharp line spectra due to isolated atoms of Si, Cu and a broad band of visible light. The line spectra resulted from an ion-induced glow above the surface and indicated pronounced outward transport of Cu atoms to the vacuum. Intensity of the line spectra and the broad band varied non-linearly with dose and dose rate. The in-situ spectroscopy provided diagnostic information of surface- and intra-solid processes associated with nanoparticle formation at high dose rates. The Cu sublimation reflected from presence of a Cu-depleted zone beneath the surface, which is one of the important factors to form a two-dimensional arrangement of nanoparticles. 相似文献

Messenger RNA has been isolated from rat lens tissue. The mRNA species which codes for the A2 chain of alpha-crystallin, revealed the same extremely high sedimentation value (14S) as the corresponding messenger from calf lens. However, it has been shown that in rat lens the 14-S messenger preparation directs the synthesis of an additional alpha-crystallin chain, designated as alpha-X with an approximate molecular weight of 24000. Wash load sedimentation during flood events is studied through comprehensive field measurements at the Shichikashuku Reservoir, Japan and numerical simulation. Field data during a flooding event caused by a typhoon in 1996 are used to set the boundary conditions for the model and to verify the simulation. In the field experiments, continuous monitoring of inflow turbidity is shown to be an effective and reliable means of estimating sediment influx. In the simulations, the proposed model is shown to provide an accurate reproduction of floodwater currents and sedimentation in the reservoir. The model achieves good accuracy and resolution by adopting an orthogonal curvilinear grid to discretize the reservoir and by solving the flow field using a three-dimensional standard k–ε turbulence model. The simulation reproduces the observed velocity, turbidity, and distribution of sedimentation well, demonstrating the potential utility of such models in the management of reservoir water quality and sedimentation. 相似文献

To investigate basic characteristics of 10 virulent phages active on silage-making lactobacilli, morphological properties, host ranges, protein composition and genome characterization were separated into five groups based on host ranges and basic properties. The seven phages of groups I, II and V were active on Lactobacillus plantarum and Lactobacillus pentosus. Phage phiPY4 (group III) infected both L. casei and Lactobacillus rhamnosus. Phage phiPY5 (group IV) specifically infected Lactobacillus casei. Morphologically, three phages of groups I belonged to the Myoviridae family, while seven other phages of groups II, III and V belonged to the Siphoviridae family. SDS-PAGE profiles, restriction analysis, G + C contents of DNA and Dot blot hybridization revealed a high degree of homology in each group. Clustering derived from host range analysis was closely related to results of DNA and protein analyses. These phages may be applicable to phage typing for silage-making lactobacilli. 相似文献
