Developing photoactivatable theranostic platforms with integrated functionalities of biocompatibility, targeting, imaging contrast, and therapy is a promising approach for cancer diagnosis and therapy. Here, we report a theranostic agent based on a hybrid nanoparticle comprising fullerene nanocrystals and gold nanoparticles (FGNPs) for photoacoustic imaging and photothermal therapy. Compared to gold nanoparticles and fullerene crystals, FGNPs exhibited stronger photoacoustic signals and photothermal heating characteristics by irradiating light with an optimal wavelength. Our studies demonstrated that FGNPs could kill cancer cells due to their photothermal heating characteristics in vitro. Moreover, FGNPs that are accumulated in tumor tissue via the enhanced permeation and retention effect can visualize tumor tissue due to their photoacoustic signal in tumor xenograft model mice. The theranostic agent with FGNPs shows promise for cancer therapy.  相似文献

Sequestration of m2 receptors (muscarinic acetylcholine receptor m2 subtypes), which was assessed as loss of N-[3H]methylscopolamine ([3H]NMS) binding activity from the cell surface, was examined in COS 7 and BHK-21 cells that had been transfected with expression vectors encoding the m2 receptor and, independently, vectors encoding a G protein-coupled receptor kinase (GRK2) (beta-adrenergic receptor kinase 1) or a GRK2 dominant-negative mutant (DN-GRK2). The sequestration of m2 receptors became apparent when the cells were treated with 10(-5) M or higher concentrations of carbamylcholine. In this case, approximately 40% or 20-25% of the [3H]NMS binding sites on COS 7 or BHK-21 cells, respectively, were sequestered with a half-life of 15-25 min. In cells in which GRK2 was also expressed, the sequestration became apparent in the presence of 10(-7) M carbamylcholine. Approximately 40% of the [3H]NMS binding sites on both COS 7 and BHK-21 cells were sequestered in the presence of 10(-6) M or higher concentrations of carbamylcholine. When DN-GRK2 was expressed in COS 7 cells, the proportion of [3H]NMS binding sites sequestered in the presence of 10(-5) M or higher concentrations of carbamylcholine was reduced to 20-30%. These results indicate that the phosphorylation of m2 receptors by GRK2 facilitates their sequestration. These results are in contrast with the absence of a correlation between sequestration and the phosphorylation of beta-adrenergic receptors by the GRK2 and suggests that the consequences of phosphorylation by GRK2 are different for different receptors.  相似文献

A detailed analysis has been carried out on the deasphaltened Cold Lake bitumen using two different series of chromatographic separations followed by i.r., u.v., n.m.r. and computerized GC/MS studies of the separated fractions: saturates (21.3), monoaromatics (8.3), diaromatics (3.6), polyaromatics and non-specific polar compounds (24.35), acids (15.2), bases (6.38), and neutral nitrogen compounds (1.15% of bitumen). The acyclic paraffin content of the maltene is low but decidedly higher than in the related Athabasca bitumen, indicating that the Cold Lake bitumen suffered a less complete biodegradation. Straight-chain paraffins and the isoprenoids phytane and pristane are present. The mono- and diaromatic fractions were analysed by high-voltage, low-resolution mass spectrometry. The monoaromatic fraction contains alkylbenzenes, naphthenebenzenes and dinaphthenebenzenes in a ratio of approximately 1:1:1. The ratio for naphthalenes, acenaphthenes + dibenzofurans and fluorenes in the diaromatic fraction is approximately 1:0.9:0.6. The composition of Cold Lake bitumen closely resembles that of the Athabasca bitumen except that its asphaltene content is somewhat lower and its acidic and saturate content is slightly higher.  相似文献

Mechanochemical modification of previously synthesized LaFeO3 perovskite‐type oxide by a high‐energy ball milling was investigated to introduce Fe4+ ions or transform some Fe3+ into Fe4+ in LaFeO3. X‐ray absorption fine structure studies revealed that the formation of Fe4+ ions into LaFeO3 perovskite has been achieved at first time by ball milling at room temperature without any additives or replacement of La3+ ions by some divalent cations. The structural model of Fe4+ containing LaFeO3 could be described as with a modified perovskite having equal amounts of La and Fe vacancies, which is supported by a good correlation between the results of Fe K‐edge XANES spectra and O2‐TPD. The synthesis of Fe4+‐containing LaFeO3 perovskite by ball milling was able to produce the O2 adsorption capacity of nonsubstituted perovskite‐type oxide.  相似文献

Fine particles of photocatalytic anatase-type TiO2 prepared through hydrolysis of titanium-tetraisopropoxide were coated by carbon through their precipitation in poly(vinyl alcohol) (PVA) aqueous solution, followed by heat treatment at high temperatures of 400-1000 °C in a flow of high purity Ar. Without carbon coating, the phase transformation from anatase to rutile started above 600 °C, but it was suppressed up to 800 °C with carbon coating. Suppression of the phase transformation depended on the amount of carbon coated, apparent suppression being observed with carbon content above 5 mass%. The amount of carbon coated on anatase was controlled by changing the concentration of PVA in the solution. In order to have a carbon content of about 5 mass%, a PVA solution with more than 2 mass% had to be used.  相似文献

Reactive blending has been usually utilized to stabilize morphology and to improve the properties of multi-component polymer blends by generating copolymers in situ at the interface. However, the present study on blends composed of poly(butylene terephthalate) (PBT) and functionalized styrene-acrylonitrile random copolymer (SAN) demonstrates another possibility for this method, i.e. tailoring morphology and thereby controlling the properties of polymer blends. By varying reaction conditions it was demonstrated that blends could be formed having the same ratio of [PBT]/[SAN] but which possessed completely different microstructural forms: a sea-island morphology with and without micelles, a corded dispersed phase morphology, and a highly oriented, layer-like morphology.  相似文献

A new palm-sized optical PM2.5 sensor has been developed and its performance evaluated. The PM2.5 mass concentration was calculated from the distribution of light scattering intensity by considering the relationship between scattering intensity and particle size. The results of laboratory tests suggested that the sensor can detect particles with diameters as small as ~0.3 µm and can measure PM2.5mass concentrations as high as ~600 µg/m3. Year-round ambient observations were conducted at four urban and suburban sites in Fukuoka, Kadoma, Kasugai, and Tokyo, Japan. Daily averaged PM2.5 mass concentration data from our sensors were in good agreement with corresponding data from the collocated standard instrument at the Kadoma site, with slopes of 1.07–1.16 and correlation coefficients (R) of 0.90–0.91, and with those of the nearest observatories of the Ministry of the Environment of Japan, at 1.7–4.1 km away from our observation sites, with slopes of 0.97–1.23 and R of 0.89–0.95. Slightly greater slopes were observed in winter than in summer, except at Tokyo, which was possibly due to the photochemical formation of relatively small secondary particles. Under high relative humidity conditions (>70%), the sensor has a tendency to overestimate the PM2.5 mass concentrations compared to those measured by the standard instruments, except at Fukuoka, which is probably due to the hygroscopic growth of particles. This study demonstrates that the sensor can provide reasonable PM2.5 mass concentration data in urban and suburban environments and is applicable to studies on the environmental and health effects of PM2.5.

Ammonium nitrate (AN)‐based composite propellants have attracted a considerable amount of attention because of the clean burning nature of AN as an oxidizer. However, such propellants have several disadvantages such as poor ignition and a low burning rate. In this study, the burning characteristics of AN‐based propellants supplemented with Fe2O3 as a burning catalyst were investigated. The addition of Fe2O3 is known to improve the ignitability at low pressure. Fe2O3 addition also increases the burning rate, while the pressure exponent generally decreases. The increasing ratio (R) of the burning rate of the AN/Fe2O3 propellant to that of the corresponding AN propellant vs. the amount of Fe2O3 added (ξ) depends on the burning pressure and AN content. R decreases at threshold value of ξ. The most effective value of ξ for increasing the burning rate was found to be 4 % for the propellant at 80 % AN, and the value generally decreased with decreasing AN content. According to thermal decomposition kinetics, Fe2O3 accelerates the reactions of AN and binder decomposition gases in the condensed‐ and/or gas‐phase reaction zones. The burning characteristics of the AN‐based propellant were improved by combining catalysts with differing catalytic mechanisms instead of supplementing the propellant with a single catalyst owing to the multiplicative effect of the former.  相似文献
