The characteristics of millimeter-wave radiation in a corrugated ferrite slab supported by a grounded dielectric slab are investigated both theoretically and experimentally. Theoretical analysis is performed by the perturbation theory combined with multiple space scales for the transverse electric mode. A pair of amplitude transport equations are derived, and the characteristics of the leaky wave are explained in terms of the radiation angle and the radiation efficiency. Experiments were carried out using a layered structure composed of a corrugated polycrystalline yttrium iron garnet slab and a Teflon slab in the frequency range from 40 to 50 GHz. We looked for p16/p19 deletion and p16 promoter methylation, as well as loss of 9p21 heterozygosity in pure squamous cell carcinomas (SCC), and in transitional cell carcinomas (TCC) of the bladder with SCC components. Homozygous deletion of p16/p19 was detected in 11 of 21 (52%) cases of pure SCCs and in three of ten (30%) cases of TCC with SCC. Three cases of TCC with SCC had p16/p19 deletion, hypermethylation of the p16 promoter, or LOH on 9p21 only in the SCC components, suggesting that these molecular alterations occurred preferentially in SCC. Interestingly, homozygous deletion of p16/p19 was observed in squamous metaplasia from bladder cancer patients (five of 11, 45%), showing that this change occurred in preneoplastic cells. On the other hand, p16/p19 deletions were not found in squamous metaplasias from non cancerous patients. Hypermethylation of the p16 promoter was observed in two of 14 tumors (14%) and none of seven metaplasias examined. These data suggest that: (a) p16/p19 deletion is associated with early carcinogenesis of SCC of the bladder, and squamous metaplasia of the bladder cancer patient has already sustained genetic changes found in cancer, and (b) genetic mosaicism occurs in cases of TCC with SCC, with the SCC component showing more frequent 9p21 alterations than the TCC component.  相似文献

We show that tetraphenylphosphonium inhibits oxidation of palmitoylcarnitine, pyruvate, malate, 2-oxoglutarate and glutamate in heart mitochondria in the range of concentration (1-5 microM) commonly used for the determination of mitochondrial membrane potential. The inhibition of 2-oxoglutarate (but not other substrate) oxidation by tetraphenylphosphonium is dependent on the concentration of 2-oxoglutarate and on extramitochondrial free calcium, and the kinetic plots are consistent with a mixed type of inhibition. Our results indicate that tetraphenylphosphonium interacts with enzymes, specifically involved in the oxidation of 2-oxoglutarate, most possibly, 2-oxoglutarate dehydrogenase.  相似文献

Recombinant human m-calpain was produced in a soluble form at a level of 20 mg/liter of Sf-9 cell culture by the coexpression of recombinant human m-calpain large (m80K) and small (30K) subunits using a baculovirus expression system. The expressed m-calpain was purified by sequential column chromatographies on DEAE-Toyopearl, gel-filtration, and Mono Q by the same method used to purify native m-calpain. The recombinant m-calpain had a specific activity of 691 U/mg and a Ka value (Ca2+ requirement for 50% caseinolysis activity) of 0.4 mM, which are essentially identical to those of native rabbit m-calpain. A mutant m-calpain large subunit, m-C105S-80K, where the active-site cysteine-105 is converted to serine by site-directed mutagenesis, was coexpressed with 30K in Sf-9 cells, purified, and characterized. m-C105S-calpain does not degrade casein nor an artificial tetra-peptide substrate, succinyl-Leu-Leu-Val-Tyr-MCA. Further, it shows no autolytic activity with Ca2+. This is the first report of the large-scale production of a fully active m-calpain species in the baculovirus system.  相似文献

Regular network polyesters were prepared from trimesic (Y), pyromellitic (X) or mellitic (YH) acids and 1,6-hexanediol (6G). Prepolymers prepared by melt-polycondensation were cast from dimethylformamide solution and post-polymerized at 260°C for various times to form a network. The resultant films were transparent, flexible and insoluble in organic solvents. Degrees of reaction estimated from the infra-red absorbance of ester and methylene groups were 95, 95 and 96% for 6GY, 6GX and 6GYH post-polymerized for 12 h, respectively. X-ray diffraction intensity curves showed the formation of some ordered structure owing to the regular networks, and the degrees of ordering were 6GY> 6GX> 6GYH. Densities were 1.227, 1.290 and 1.289 g cm−3 and heat distortion temperatures measured by thermomechanical analysis were 75, 72 and 75°C for 6GY, 6GX and 6GYH post-polymerized for 12 h, respectively. Tensile strengths were in the order 6GY> 6GX> 6GYH and Young's moduli were 6GYH = 6GX> 6GY. Dye absorptions were 6GX> 6GY> 6GYH, and water absorptions and weight losses in alkali solution were 6GYH > 6GX> 6GY. These effects of the number of functional groups on the benzene ring on the structure and properties were discussed, and compared with those of previously reported network polymers.  相似文献

Findings obtained from dental charts and from radiograph taken one year later for the same individuals were examined and classified according to different criteria. Similarities between the two sources investigated by the canonical correlation method. The results obtained were as follows: 1. The correlation coefficient and the squared multiple correlations (R2) were calculated. Consequently, high similarities were seen between the two sources for the mandibular anterior teeth [sequence: see text], maxillary incisor teeth [sequence: see text], mandibular and maxillary first premolar teeth [sequence: see text], and the corresponding teeth of the same jaws. 2. Three roots showed high elucidative values by the canonical correlation method. The first root was understood to be the mandibular incisor factor, the second the maxillary incisor factor, and the third the first premolar factor. This seemed to indicate, that the similarities between the two sources in these regions were very high. 3. A study of the similarities between the two sources was done based on the individual scores of canonical variable loadings calculated in this analysis. High similarities were seen at [sequence: see text] in category 1 (intact & C1) for chart findings and category 1'(intact, C1-C3 & resin filling) for radiographic findings: at [sequence: see text] in category 2 (C2-C4, inlay & filling of repair) for chart findings and category 3'(stump of tooth) for radiographic findings; and at [sequence: see text] in category 3 (complete crown, jacket crown & post crown) for chart findings and category 3'(stump of tooth) for radiographic findings. 4. The present results suggest that if proper case in taken, one set of information can serve as a substitute for the other. In personal identification, higher efficiency may be achieved if this is borne in mind.  相似文献

We have studied the train-of-four (TOF) response mechanomyographically during onset of neuromuscular block produced by subclinical doses of suxamethonium in order to follow the augmentation of the first twitch of the TOF (T1) and TOF fade compared with control TOF responses before the drug was given. In the groups given suxamethonium 0.05, 0.1, 0.2 and 0.3 mg kg-1, the increments in T1 after administration of the drug were observed before twitch depression occurred; these were mean 22.3 (SEM 8.1)%, 19.2 (3.3)%, 10.8 (2.0)% and 4.2 (2.2)%, respectively. This effect was more marked with the lower doses (P < 0.05). The degree of TOF fade was moderate during onset of neuromuscular block and depended on the dose of drug. The results of this study suggest that low doses of suxamethonium produced transient increase in muscle tension and twitch depression with significant TOF fade. We conclude that suxamethonium was associated with presynaptic effects as a consequence of brief stimulation of acetylcholine release followed by progressive diminution at the neuromuscular junction.  相似文献

The effects of methionine on hepatocarcinogenesis induced by coadministration of a choline-deficient L-amino acid-defined (CDAA) diet and ethionine were examined. F344 male rats were divided into 4 experimental groups. Groups 1 and 2 received the CDAA diet and a choline-supplemented L-amino acid-defined (CSAA)++ diet, respectively. Group 3 received the CDAA diet containing 0.05% ethionine, and group 4 the CDAA diet containing 0.05% ethionine and 0.47% methionine. Animals were killed after 12 weeks of treatment. Histologically, the CDAA diet induced intracellular fat accumulation and foci. In contrast, ethionine caused not only foci, but also hyperplastic nodules, cholangiofibrosis and the proliferation of oval cells without such fat accumulation. Methionine abolished the development of all of the liver lesions induced by coadministration of the CDAA diet and ethionine. To investigate the effects of methionine on induction of c-myc and c-Ha-ras expression, as well as generation of 8-hydroxyguanine (8-OHGua) and 2-thiobarbituric acid-reacting substances (TBARS), by coadministration of the CDAA diet and ethionine, subgroups of 3 to 5 animals were killed at 2, 4, 8, or 11 days after the beginning of the experiment. Coadministration of the CDAA diet and ethionine markedly enhanced the level of expression of c-myc and c-Ha-ras, 8-OHGua formation and TBARS generation as compared with the CDAA or CSAA diet within 11 days, and methionine blocks these actions. These results indicate that addition of methionine prevents the induction of c-myc and c-Ha-ras expression, 8-OHGua formation and TBARS generation, as well as hepatocellular lesions, by coadministration of the CDAA diet and ethionine in rats, and suggest a possible involvement of oxidative stress and gene expression in hepatocarcinogenesis by these agents.  相似文献

Sarcophytol A (SaA), a cembrane-type diterpene, inhibits pancreatic carcinogenesis induced by N-nitrosobis(2-oxypropyl)amine (BOP) in hamsters. The experimental groups received two injections of BOP at 70 mg/kg dose, followed 2 weeks later by a 20 mg/kg dose injection, and were fed a basal diet or 0.01 and 0.05% SaA diets starting 1 week after the second injection of BOP. Control groups were injected with normal saline and fed the basal diet or the 0.05% SaA diet. All animals were killed 30 weeks after the start of the experiments. Seventeen BOP-treated hamsters fed the basal diet developed pancreatic tumors (77.3%) while only 12 of 21 hamsters fed the 0.01% SaA diet (57.1%) and 12 of 23 hamsters fed the 0.05% SaA diet (52.2%) developed pancreatic tumors. Pancreatic lesions included ductal hyperplasia, atypical ductal hyperplasia, and carcinoma in situ. Microscopic invasive carcinoma induced by BOP and the incidence of larger pancreatic tumors in hamsters were significantly higher in hamsters fed the basal diet than in hamsters fed the SaA diet (p < 0.05). The proliferating cell nuclear antigen (PCNA) labeling index of pancreatic carcinoma in BOP-treated hamsters fed the basal diet was 41.2 +/- 13.4%, whereas BOP-treated hamsters fed 0.01 and 0.05% SaA diets yielded PCNA indexes of 26.8 +/- 8.3 and 28.4 +/- 7.0%, respectively. k-ras mutation was detected in 40% of cancers in both groups. No pancreatic tumors developed in saline-treated groups, and no differences in body weights or histological findings in their organs, including the pancreas, were observed in either group. These findings suggest that SaA not only inhibits BOP-induced pancreatic carcinogenesis in hamsters, but also provides antipromotion and antiprogression effects on these tumors, even when SaA commences 1 week after the initiation of pancreatic carcinogenesis.  相似文献
