The hydrodynamic transport equations for charges in a semiconductor have been solved for a periodic excitation by means of a harmonic approach, in order to model microwave and millimetre-wave active devices. The solution is based on the expansion of physical variables in a Fourier series in the time domain, and on discretisation in the space domain. A waveform-balance technique in the TD is used to solve the nonlinear equations system. This approach allows for a longer time step with respect to standard TD solutions for most cases of interest, greatly reducing simulation time by at least two orders of magnitude in typical cases. © 2003 Wiley Periodicals, Inc. Peritoneal dialysis (PD) represents the dialysis modality of choice for pediatric patients with end-stage kidney disease. Indeed, compared with hemodialysis (HD), it offers many advantages, including more flexibility, reduction of the risk of hospital-acquired infections, preservation of residual kidney function, and a better quality of life. However, despite these positive aspects, PD may be associated with several long-term complications that may impair both patient’s general health and PD adequacy. In this view, chronic inflammation, caused by different factors, has a detrimental impact on the structure and function of the peritoneal membrane, leading to sclerosis and consequent PD failure both in adults and children. Although several studies investigated the complex pathogenic pathways underlying peritoneal membrane alterations, these processes remain still to explore. Understanding these mechanisms may provide novel approaches to improve the clinical outcome of pediatric PD patients through the identification of subjects at high risk of complications and the implementation of personalized interventions. In this review, we discuss the main experimental and clinical experiences exploring the potentiality of the proteomic analysis of peritoneal fluids and extracellular vesicles as a source of novel biomarkers in pediatric peritoneal dialysis.  相似文献

mitochondrial neuro-gastrointestinal encephalomyopathy (MNGIE) is a rare genetic disorder characterized by thymidine phosphorylase (TP) enzyme defect. The absence of TP activity induces the imbalance of mitochondrial nucleotide pool, leading to impaired mitochondrial DNA (mtDNA) replication and depletion. Since mtDNA is required to ensure oxidative phosphorylation, metabolically active tissues may not achieve sufficient energy production. The only effective life-saving approach in MNGIE has been the permanent replacement of TP via allogeneic hematopoietic stem cell or liver transplantation. However, the follow-up of transplanted patients showed that gut tissue changes do not revert and fatal complications, such as massive gastrointestinal bleeding, can occur. The purpose of this study was to clarify whether the reintroduction of TP after transplant can recover mtDNA copy number in a normal range. Using laser capture microdissection and droplet-digital-PCR, we assessed the mtDNA copy number in each layer of full-thickness ileal samples of a naive MNGIE cohort vs. controls and in a patient pre- and post-TP replacement. The treatment led to a significant recovery of gut tissue mtDNA amount, thus showing its efficacy. Our results indicate that a timely TP replacement is needed to maximize therapeutic success before irreversible degenerative tissue changes occur in MNGIE.  相似文献

The calibration of a three-dimensional digitizer is a very important issue to take into consideration that good quality, reliability, accuracy and high repeatability are the features which a good digitizer is expected to have. The aim of this paper is to propose a new method for the calibration of a 3-D laser scanner, mainly for robotic applications. The acquisition system consists of a laser emitter and a webcam with fixed relative positions. In addition, a cylindrical lens is provided with the laser housing so that it is capable to project a plane light. An optical filter was also used in order to segment the laser stripe from the rest of the scene. For the calibration procedure it was used a digital micrometer that move a target with known dimensions. The calibration method is based on the modeling of the geometrical relationship between the 3-D coordinates of the laser stripe on the target and its digital coordinates in the image plane. By this method it is possible to calibrate the intrinsic parameters of the video system, the position of the image plane and the laser plane in a given frame, all in the same time.  相似文献

Neuropathy development is a major dose-limiting side effect of anticancer treatments that significantly reduces patient’s quality of life. The inadequate pharmacological approaches for neuropathic pain management warrant the identification of novel therapeutic targets. Mitochondrial dysfunctions that lead to reactive oxygen species (ROS) increase, cytosolic Ca2+ imbalance, and lactate acidosis are implicated in neuropathic pain pathogenesis. It has been observed that in these deregulations, a pivotal role is played by the mitochondrial carbonic anhydrases (CA) VA and VB isoforms. Hence, preclinical studies should be conducted to assess the efficacy of two novel selenides bearing benzenesulfonamide moieties, named 5b and 5d, and able to inhibit CA VA and VB against paclitaxel-induced neurotoxicity in mice. Acute treatment with 5b and 5d (30–100 mg/kg, per os – p.o.) determined a dose-dependent and long-lasting anti-hyperalgesic effect in the Cold plate test. Further, repeated daily treatment for 15 days with 100 mg/kg of both compounds (starting the first day of paclitaxel injection) significantly prevented neuropathic pain development without the onset of tolerance to the anti-hyperalgesic effect. In both experiments, acetazolamide (AAZ, 100 mg/kg, p.o.) used as the reference drug was partially active. Moreover, ex vivo analysis demonstrated the efficacy of 5b and 5d repeated treatments in reducing the maladaptive plasticity that occurs to glia cells in the lumbar portion of the spinal cord and in improving mitochondrial functions in the brain and spinal cord that were strongly impaired by paclitaxel-repeated treatment. In this regard, 5b and 5d ameliorated the metabolic activity, as observed by the increase in citrate synthase activity, and preserved an optimal mitochondrial membrane potential (ΔΨ) value, which appeared depolarized in brains from paclitaxel-treated animals. In conclusion, 5b and 5d have therapeutic and protective effects against paclitaxel-induced neuropathy without tolerance development. Moreover, 5b and 5d reduced glial cell activation and mitochondrial dysfunction in the central nervous system, being a promising candidate for the management of neuropathic pain and neurotoxicity evoked by chemotherapeutic drugs.  相似文献

Philadelphia-negative chronic myeloproliferative neoplasms (MPNs) represent a group of hematological disorders that are traditionally considered as indistinct slow progressing conditions; still, a subset of cases shows a rapid evolution towards myelofibrotic bone marrow failure. Specific abnormalities in the megakaryocyte lineage seem to play a central role in this evolution, especially in the bone marrow fibrosis but also in the induction of myeloproliferation. In this review, we analyze the current knowledge of prognostic factors of MPNs related to their evolution to myelofibrotic bone marrow failure. Moreover, we focused the role of the megakaryocytic lineage in the various stages of MPNs, with updated examples of MPNs in vitro and in vivo models and new therapeutic implications.  相似文献

This review aims to describe and discuss the different functions of the endolysosomal system, from homeostasis to its vital role during viral infections. We will initially describe endolysosomal system’s main functions, presenting recent data on how its compartments are essential for host defense to explore later how SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) and other coronaviruses subvert these organelles for their benefit. It is clear that to succeed, pathogens’ evolution favored the establishment of ways to avoid, escape, or manipulate lysosomal function. The unavoidable coexistence with such an unfriendly milieu imposed on viruses the establishment of a vast array of strategies to make the most out of the invaded cell’s machinery to produce new viruses and maneuvers to escape the host’s defense system.  相似文献

Obesity is one of the greatest health challenges affecting children of all ages and ethnicities. Almost 19% of children and adolescents worldwide are overweight or obese, with an upward trend in the last decades. These reports imply an increased risk of fat accumulation in hepatic cells leading to a series of histological hepatic damages gathered under the acronym NAFLD (Non-Alcoholic Fatty Liver Disease). Due to the complex dynamics underlying this condition, it has been recently renamed as ‘Metabolic Dysfunction Associated Fatty Liver Disease (MAFLD)’, supporting the hypothesis that hepatic steatosis is a key component of the large group of clinical and laboratory abnormalities of Metabolic Syndrome (MetS). This review aims to share the latest scientific knowledge on MAFLD in children in an attempt to offer novel insights into the complex dynamics underlying this condition, focusing on the novel molecular aspects. Although there is still no treatment with a proven efficacy for this condition, starting from the molecular basis of the disease, MAFLD’s therapeutic landscape is rapidly expanding, and different medications seem to act as modifiers of liver steatosis, inflammation, and fibrosis.  相似文献

The enhancement of the search capabilities of geo-spatial tools occupies one of the highest positions in the agenda of the INSPIRE initiative. This can be done by equipping applications with tools able to understand user terminology. However, this is in contrast with current approaches, which tend to fix in advance the terminology with a consequent rigidity in the way users interact with the system. In this paper we present the work we have done with the Semantic Geo-Catalogue (SGC) project in providing a semantic extension to the geo-catalogue of the Autonomous Province of Trento (PAT) in Italy. This was done through the adoption of a semantic matching tool and a faceted ontology that codifies knowledge about the geography of the PAT and that was created by reorganizing data extracted from the local geographical dataset. Thanks to the semantic extension, queries to the geo-catalogue are expanded with domain specific terms taken from the ontology thus obtaining a higher number of relevant documents in output. We also complied with the Open Government Data (OGD) initiative by publishing in RDF and by linking to relevant dictionaries some useful data taken from the local repository.  相似文献

A real time FFT-based impedance meter is here presented. It is realized simply by a standard PC in addition to a low cost two channels hardware device, and is based on a cost-effective algorithm to meaningfully reduce the known problem of the bias introduced by the relative phase and amplitude error between channels. The impedance meter exploits the implementation of an ad hoc cost-effective algorithm and a synchronous sampling allows eliminating the leakage error. The aliasing and the spectral interference are eliminated by means of an anti-aliasing filter and the use of very low distortion sinusoidal signals. Automatic measurements can be managed too, capturing different measures at different test frequencies and displaying them graphically on the PC screen. To validate the measured results comparative test were performed with respect to certified and calibrated commercially available multimeters known with good accuracies.Finally, this novel impedance meter integrates a home-made software, previously reported [1] capable to implement real time virtual instruments.  相似文献
