Flaujeac trait is the functional deficiency of a plasma protein of the intrinsic coagulation, kinin-forming, and plasma fibrinolytic pathways. The Flaujeac factor in man has been isolated and tentatively identified as a kininogen of high molecular weight (HMW). Highly purified bovine HMW-kininogen, but not bovine low molecular weight kininogen, repaired Flaujeac factor deficiency. The two subspecies of this molecule, HMW-kininogen a and HMW-kininogen b, also corrected Flaujeac factor deficiency. When bovine HMW-kininogen was incubated with bovine plasma kallikrein, kinin-free HMW-kininogen, bradykinin, and a glycopeptide fragment (peptide 1-2; 12,584 daltons) were rapidly released. None of these fragmentation products corrected Flaujeac factor deficiency alone or in mixtures. The function of HMW-kininogen appeared to depend upon the structural integrity of the native molecule. When injected in concentrations of 2 pmol-8 nmol/0.1 ml, peptide 1-2 caused increased vascular permeability in rabbits, rats, or guinea pigs. The enhanced permeability was maximal within 1-2 min and terminated in 5-10 min, differing from that of bradykinin or histamine. Injected together in equimolar amounts, peptide 1-2 and bradykinin produced a synergistic permeability response which was immediate in onset as well as prolonged in duration. Peptide 1-2 is a rapidly acting, highly basic glyco-peptide which mediates increased vascular permeability in a complementary and synergistic manner with bradykinin. 相似文献

Leak-before-break (LBB) assessment of primary heat transport piping of nuclear reactors involves detailed fracture assessment
of pipes and elbows with postulated throughwall cracks. Fracture assessment requires the calculation of elastic–plastic J-integral and crack opening displacement (COD)1 for these piping components. Analytical estimation schemes to evaluate elastic–plastic J-integral and COD simplify the calculations. These types of estimation schemes are available for pipes with various crack
configurations subjected to different types of loading. However, such schemes for elbow (or pipe bend), which is one of the
important components for LBB analyses, is very meager. Recently, elastic–plastic J and COD estimation scheme has been developed for throughwall circumferentially cracked elbow subjected to closing bending
moment. However, it is well known that the elbow deformation characteristics are distinctly different for closing and opening
bending modes because the ovalisation patterns of elbow cross section are different under these two modes. Development of
elastic–plastic J and COD estimation scheme for an elbow with throughwall circumferential crack at intrados subjected to opening bending moment
forms the objective of the present paper. Experimental validation of proposed J-estimation scheme has been provided by comparing the crack initiation, unstable ductile tearing loads and crack extension

The three-dimensional structure of a new crystal form of methanol dehydrogenase from Methylophilus W3A1 has been obtained in the presence of substrate using data recorded at a synchrotron. The structure of this approximately 140 kDa heterotetramer, refined at 1. 9 A resolution, reveals the detailed configuration of its redox cofactor, pyrroloquinoline quinone (PQQ). C4, one of the oxygen-bearing atoms of this orthoquinone is in a planar configuration while C5, which bears the other quinone oxygen, is tetrahedral, suggesting that the PQQ is in the semiquinone redox state. The substrate binding site has been identified close to PQQ and to the side chain of Asp297, the putative active site base. The proximity of the hydroxyl of methanol to C5 of PQQ compared to the greater separation of the substrate methyl group from C5 supports the addition-elimination reaction mechanism involving a hemiketal intermediate. 相似文献

Summary The paper is concerned with the propagation of Love waves in a porous layer over an inhomogeneous half-space due to a point source. The dispersion equation for Love waves in a porous layer has been derived. The effects of porosity and inhomogeneity in the half-space have been shown graphically on the dispersion curves.
