The atomic force microscope (AFM) is a powerful tool for the measurement of forces at the micro/nano scale when calibrated cantilevers are used. Besides many existing calibration techniques, the thermal calibration is one of the simplest and fastest methods for the dynamic characterization of an AFM cantilever. This method is efficient provided that the Brownian motion (thermal noise) is the most important source of excitation during the calibration process. Otherwise, the value of spring constant is underestimated. This paper investigates noise interference ranges in low stiffness AFM cantilevers taking into account thermal fluctuations and acoustic pressures as two main sources of noise. As a result, a preliminary knowledge about the conditions in which thermal fluctuations and acoustic pressures have closely the same effect on the AFM cantilever (noise interference) is provided with both theoretical and experimental arguments. Consequently, beyond the noise interference range, commercial low stiffness AFM cantilevers are calibrated in two ways: using the thermal noise (in a wide temperature range) and acoustic pressures generated by a loudspeaker. We then demonstrate that acoustic noises can also be used for an efficient characterization and calibration of low stiffness AFM cantilevers. The accuracy of the acoustic characterization is evaluated by comparison with results from the thermal calibration.  相似文献

Drilling of composite material structures is widely used for aeronautical assemblies. When drilling, damage to the composite laminate is directly related to the cutter geometry and the cutting conditions. Delamination of the composite materials at the hole exit as directly related to the axial force (F Z) of the cutter is considered to be the major such defect. To address this issue, an orthotropic analytical model is developed in order to calculate the critical force of delamination during drilling and a number of hypotheses for loading are proposed. This critical axial load is related to the delamination conditions (propagation of cracks in the last layers) and the mechanical characteristics of the composite material machined. A numerical model is also drawn up to allow for numerical validation of the analytical approach. A comparison between these analytical and numerical modellings and experimental results from quasi-static punch tests led to the choice of the loading hypothesis closest to the experimental conditions. The selection of corresponding load permits to model the drilling critical thrust force on delamination and then to optimise the cutting conditions. The dimensions and geometrical shape of the cutter are of considerable importance when it comes to choosing this load. The present article focuses on the case of the twist drill, which is commonly used to drill thick plates. However, this work can be adapted to different cutter geometries.  相似文献

The oxidation behavior of the alloy Fe–25%Cr–6%Al-RE (rich in lanthanum) was investigated by means of TEM analysis. The results show that after 2 hr oxidation of the alloy, in pure oxygen at 1200° C, La precipitated in the oxide scale at the top of -Al2O3 grains and at the grain-boundary regions in the form of tiny particles of hexagonal La2O3. These tiny particles prevented aluminum from diffusing toward the surface and suppressed lateral growth of the oxide scale. The rare-earth constituents accelerated the internal oxidation of the alloy during thermal cycling between 1200° C and room temperature. They appeared as particles of aluminum oxide and lanthanum oxide. Particles of cubic La2O3 precipitated in the alloy matrix near the oxide scale-metal interface in a direction parallel to grain boundaries.  相似文献

Free-living amoebae (FLA) are protozoa that can be found in water networks where they prey on bacteria within biofilms. Most bacteria are digested rapidly by phagocytosis, however some are able to survive within amoebae and some are even able to multiply, as it is the case for Legionella pneumophila. These resisting bacteria are a potential health problem as they could also resist to macrophage phagocytosis. Several publications already reported intra-amoebal bacteria but the methods of identification did not allow metagenomic analysis and are partly based on co-culture with one selected amoebal strain. The aim of our study was to conduct a rRNA-targeted metagenomic analysis on amoebae and intra-amoebal bacteria found in drinking water network, to provide the first FLA microbiome in environmental strains. Three sites of a water network were sampled during four months. Culturable FLA were isolated and total DNA was prepared, allowing purification of both amoebal and bacterial DNA. Metagenomic studies were then conducted through 18S or 16S amplicons sequencing. Hartmannella was by far the most represented genus of FLA. Regarding intra-amoebal bacteria, 54 genera were identified, among which 21 were newly described intra-amoebal bacteria, underlying the power of our approach. There were high differences in bacterial diversity between the three sites. Several genera were highly represented and/or found at least in two sites, underlying that these bacteria could be able to multiply within FLA. Our method is therefore useful to identify FLA microbiome and could be applied to other networks to have a more comprehensive view of intra-amoebal diversity.  相似文献

Bioactive glasses are able to bond to bone through the formation of hydroxy-carbonate apatite in body fluids while strontium (Sr)-releasing bioactive glasses are of interest for patients suffering from osteoporosis, as Sr was shown to increase bone formation both in vitro and in vivo. A melt-derived glass series (SiO2–P2O5–CaO–Na2O) with 0–100% of calcium (Ca) replaced by Sr on a molar base was prepared. pH change, ion release and apatite formation during immersion of glass powder in simulated body fluid and Tris buffer at 37°C over up to 8 h were investigated and showed that substituting Sr for Ca increased glass dissolution and ion release, an effect owing to an expansion of the glass network caused by the larger ionic radius of Sr ions compared with Ca. Sr release increased linearly with Sr substitution, and apatite formation was enhanced significantly in the fully Sr-substituted glass, which allowed for enhanced osteoblast attachment as well as proliferation and control of osteoblast and osteoclast activity as shown previously. Studying the composition–structure–property relationship in bioactive glasses enables us to successfully design next-generation biomaterials that combine the bone regenerative properties of bioactive glasses with the release of therapeutically active Sr ions.  相似文献

In this study, criteria on the artificial compliance due to intrinsic cohesive zone models are presented. The approach is based on a micromechanical model for a collection of cohesive zone models embedded between each mesh of a finite element-type discretization. The overall elastic behaviour of this cohesive volumetric medium is obtained using homogenization techniques and is given in a closed-form as function of bulk properties of the relevant material and mesh parameters (the mesh type and size). Practical criteria are obtained for the calibration of the cohesive stiffnesses bounding the additional compliance inherent to intrinsic cohesive zone models by lower value. For isotropic planar discretizations (e.g. Delaunay mesh), a rigorous bound is derived whereas convenient estimates are given for non-isotropic discretizations (e.g. regular mesh).  相似文献
