The sarco-endoplasmic reticulum calcium ATPase (SERCA) is responsible for maintaining calcium homeostasis in all eukaryotic cells by actively transporting calcium from the cytosol into the sarco-endoplasmic reticulum (SR/ER) lumen. Calcium is an important signaling ion, and the activity of SERCA is critical for a variety of cellular processes such as muscle contraction, neuronal activity, and energy metabolism. SERCA is regulated by several small transmembrane peptide subunits that are collectively known as the “regulins”. Phospholamban (PLN) and sarcolipin (SLN) are the original and most extensively studied members of the regulin family. PLN and SLN inhibit the calcium transport properties of SERCA and they are required for the proper functioning of cardiac and skeletal muscles, respectively. Myoregulin (MLN), dwarf open reading frame (DWORF), endoregulin (ELN), and another-regulin (ALN) are newly discovered tissue-specific regulators of SERCA. Herein, we compare the functional properties of the regulin family of SERCA transmembrane peptide subunits and consider their regulatory mechanisms in the context of the physiological and pathophysiological roles of these peptides. We present new functional data for human MLN, ELN, and ALN, demonstrating that they are inhibitors of SERCA with distinct functional consequences. Molecular modeling and molecular dynamics simulations of SERCA in complex with the transmembrane domains of MLN and ALN provide insights into how differential binding to the so-called inhibitory groove of SERCA—formed by transmembrane helices M2, M6, and M9—can result in distinct functional outcomes.  相似文献

A novel control approach is proposed to optimize the fatigue distribution of wind turbines in a large‐scale offshore wind farm on the basis of an intelligent agent theory. In this approach, each wind turbine is considered to be an intelligent agent. The turbine at the farm boundary communicates with its neighbouring downwind turbines and organizes them adaptively into a wind delivery group along the wind direction. The agent attributes and the event structure are designed on the basis of the intelligent agent theory by using the unified modelling language. The control strategy of the intelligent agent is studied using topology models. The reference power of an individual wind turbine from the wind farm controller is re‐dispatched to balance the turbine fatigue in the power dispatch intervals. In the fatigue optimization, the goal function is to minimize the standard deviation of the fatigue coefficient for every wind turbine. The optimization is constrained such that the average fatigue for every turbine is smaller than what would be achieved by conventional dispatch and such that the total power loss of the wind farm is restricted to a few percent of the total power. This intelligent agent control approach is verified through the simulation of wind data from the Horns Rev offshore wind farm. The results illustrate that intelligent agent control is a feasible way to optimize fatigue distribution in wind farms, which may reduce the maintenance frequency and extend the service life of large‐scale wind farms. The aim of this study was to evaluate the antioxidant mechanisms of red ginseng essential oil (REO) in cells as well as in an animal model. REO was prepared by a supercritical CO(2) extraction of waste-products generated after hot water extraction of red ginseng. In HepG2 cells, REO diminished the H(2)O(2)-mediated oxidative stress and also restored both the activity and expression of antioxidant enzymes such as superoxide dismutase, catalase and glutathione peroxidase. Administration of REO inhibited the phosphorylation of upstream mitogen-activated protein kinases (MAPKs) such as c-Jun N-terminal kinase, extracellular signal-regulated kinase, and p38. In mice, the CCl(4)-mediated elevation of serum aspartate transaminase and alanine transaminase as well as the induction of hepatic lipid peroxidation were decreased by REO administration. REO treatments also resulted in up-regulation of the antioxidant enzyme expression in the liver. Moreover, increased phosphorylations of MAPKs were inhibited after REO administration. Overall, REO seems to protect the liver from oxidative stress through the activation and induction of antioxidant enzymes via inhibition of MAPKs pathways.  相似文献

An extraction and analytical method for the determination of pesticide residues in fruits and vegetables has been developed. The method includes extraction with a pressurised liquid solvent containing a mixture of 1,1,1,2-tetrafluoroethane and toluene, and identification/quantification of pesticides using ultrahigh-performance liquid chromatography coupled to tandem mass spectrometry (UPLC/MS/MS). Validation studies were carried out to evaluate the performance of the method for the determination of 71 different pesticides and metabolites in tomato, cucumber, pepper, spinach, zucchini, grape, cherry, peach and apricot. Matrix-matched calibration curves were applied and correlation coefficients (r(2)) came out to be greater than 0.99. Limit of quantification (LOQ) values of the active substances were found to be lower than the maximum residue limits (MRL) according to regulations in Turkey. The recovery values were found to be between 70% and 120% with relative standard deviations less than 20%. Based on these results, the proposed method is fast, cheaper, rugged and gives quantitative results with no additional clean-up steps.  相似文献

Polystyrene (PS) was prepared using two different polymerization methods (dispersion polymerization and seed polymerization) to investigate the steric stabilizer effect during the adsorption process of carbon nanotubes (CNTs) on the surface of PS microspheres. Experiments with different microsphere diameters and difference types of CNTs were conducted to analyze the curvature effect of the spheres on the adsorption mechanism. The results showed that PS microspheres prepared through dispersion polymerization exhibited preferable adsorption behavior compared to PS spheres prepared through seed polymerization, suggesting that poly(N-vinylpyrrolidone) led to improved adsorption interactions between the CNTs and the PS microspheres in the CNTs dispersion. Additionally, the PS diameter and CNT curvature were examined with respect to the adsorption behavior between the PS microspheres and the CNTs. Multiwalled carbon nanotubes (MWCNTs) were found to be well adsorbed on the surface of PS microspheres measuring 2 microm. However, the MWCNTs were adsorbed much less on the surface of submicron-sized PS microspheres, compared with thinwalled carbon nanotubes (TWCNTs). On the other hand, TWCNTs were found to be suitable for adsorption on submicron-sized PS microspheres. These results also indicate that the curvature of the CNTs and the polymer microspheres are important to the CNT adsorption process.  相似文献

To investigate the effect of starter isolated from kimchi, we selected the most effective strain among the 121 strains examined and identified the strain as Leuconostoc mesenteroides LK93 through 16S rDNA sequencing. This strain exhibited resistance to acid and bile salts with good survival capacity and showed antimicrobial and antifungal activity. We investigated the effects of starter on kimchi fermentation. The kimchi was fermented at 25°C for 25 d. Furthermore, the LK93 strain was examined for its ability to inhibit the growth of film-forming yeast. It was determined that the LK93 strain could be a potential starter in kimchi.  相似文献

The design of a three‐bladed wind turbine rotor is described, where the main focus has been highest possible mechanical power coefficient, CP, at a single operational condition. Structural, as well as off‐design, issues are not considered, leading to a purely theoretical design for investigating maximum aerodynamic efficiency. The rotor is designed assuming constant induction for most of the blade span, but near the tip region, a constant load is assumed instead. The rotor design is obtained using an actuator disc model, and is subsequently verified using both a free‐wake lifting line method and a full three‐dimensional Navier–Stokes solver. Excellent agreement is obtained using the three models. Global CP reaches a value of slightly above 0.51, while global thrust coefficient CT is 0.87. The local power coefficient Cp increases to slightly above the Betz limit on the inner part of the rotor; the local thrust coefficient Ct increases to a value above 1.1. This agrees well with the theory of de Vries, which states that including the effect of the low pressure behind the centre of the rotor stemming from the increased rotation, both Cp and Ct will increase towards the root.
