Cationic liposomes have been extensively explored as gene delivery vector for several reasons. It is because disadvantages of viral vectors include risk of replication, possible immunogenicity, and the difficulty of obtaining a large quantity of viral vectors. Currently, a variety of cationic components for liposome formulations have been developed. The components are broadly divided into two classes based on the chemical structure of hydrophobic moieties: long aliphatic (saturated or unsaturated) hydrocarbons and cholesterol ring. A variety of hydrophilic moieties also include tertiary amines, ammonium salt and spermine. The role of liposomes is to condense DNA to form complexes with high affinity to cell surfaces where possible fusion or destabilization of the membrane and/or endocytosis are involved. However, at present, little structure-activity relationships are known. Some vectors are on clinical trials approved by NIH.  相似文献

The absorption of unbranched aliphatic alcohols in the rabbit vagina was studied using a perfusion method, and the absorption rates were found to be first order with respect to the drug concentration in the vagina from methanol to octanol. A physical model involving an aqueous diffusion layer in series with a membrane consisting of aqueous pores and lipoidal pathways was used for analyzing the data. The physically based parameters in the model were determined. An effective diffusion layer thickness ("unstirred layer") of around 0.035 cm was found. The increase in the permeability coefficient for the lipoidal pathway per methylene group was around 2.5 for this homologous series.  相似文献

STUDY OBJECTIVES: The peak inspiratory flow rates (PIFRs) generated by cystic fibrosis (CF) and COPD patients through a range of clinically relevant resistances have not yet been reported (to our knowledge). The objectives of this study were to (1) explore a relevant range of resistive loads and address whether patients with stable CF and COPD can generate the PIFR sufficient to disperse dry-powder inhalants (DPI) and (2) determine whether the optimal inspiratory flow rate effective for delivery of aerosolized pharmacologic therapeutic agents can be attained with a comfort rating acceptable to subjects. DESIGN: Prospective, controlled, subject-blinded study. SETTING: Pulmonary function laboratory at the VA Palo Alto Health Care System. PATIENTS OR PARTICIPANTS: Thirty-six subjects, including 12 healthy volunteers, 12 subjects with CF, and 12 subjects with COPD were studied. MEASUREMENTS: Studies of dynamic lung function and PIFR without and with varying resistances were obtained at a single laboratory visit. RESULTS: Dynamic lung function and PIFR varied inversely with the resistive load for all patient groups and did not correlate with the disease severity, as indicated by FEV1 of percent predicted. The average subjective comfort rating for any given resistive load was similar for subjects with CF and COPD. CONCLUSIONS: These results support the conclusion that subjects with stable CF and COPD of varying severity can comfortably generate the necessary flow rates to operate new and currently available DPIs over a wide range of inspiratory resistances.  相似文献

A 2.125-Gb/s transmitter meeting the specifications of the emerging ANSI Fiber Channel standard has been developed using BiCMOS technology. This transmitter features (1) a fully bipolar 10:1 multiplexer (MUX) and a 2.125-GHz retimer for high-accuracy transmission of data, (2) an emitter-coupled logic (ECL) CMOS analog phase-locked loop, (3) pure ECL-level output for direct connection to the currently available optical modules, and (4) BiCMOS process technology that includes 0.25-μm CMOS devices and 20-GHz bipolar devices. The LSI serializes 32-bit-wide, 53.125-Mb/s data into 2.125-Gb/s data through a CMOS 8B10B encoder. The chip area is 3×2 mm2, and the power dissipation is 860 mW  相似文献

We have developed a simple algorithm for scalp localization for craniotomy. Using a series of CT/MRI data, the point on the scalp closest to the center of the lesion (T) is determined by our program. A plane (plane R) is defined by three points: bilateral auditory meati and T. Point S is the intersection of three planes: the plane R, the scalp surface and the midsagittal plane. The distance from either S or the ipsilateral external auditory meatus to T is measured along the scalp surface on the plane R. The distance from the nasion to S along the scalp surface on the midsagittal plane is also measured. To determine the craniotomy site, these distances are measured directly on a patient's scalp in the operating room. This simple and accurate method for scalp localization could be used by installing our program in conventional CT/MR scanners.  相似文献

The non-Newtonian behavior of commercial linear polyethylene samples and their fractions were studied at 190°C. The viscosity η versus shear rate \documentclass{article}\pagestyle{empty}\begin{document}$ \dot \gamma $\end{document} curves of whole polymers could be superimposed onto a single master curve despite the variations of their molecular weights and molecular weight distributions. For fractions, however, the same master curve was inapplicable, and the sensitivity of the viscosity to shear rate was found to be greater than those of the whole polymers. The zero-shear viscosities η0 of fractions were related to the 3.42 power of the weight-average molecular weight Mu as follows: For whole polymers, the zero-shear viscosities were found to be considerably higher at the same Mw and markedly lower at the same z-average molecular weight Mz than those of the fractions. Thus, it was concluded that η0 corresponds to an average of molecular weight between Mw and Mz. It was found that the molecular relaxation time τ is proportional to Mz5.3 for whole polymers and to η0Mw for fractions. Using these relations it was possible to relate the flow ratio, the ratio of flow rates at two different shear stresses, with the molecular weight distribution.  相似文献

Changes in temperature, relative humidity, and moisture content of green coffee beans were monitored during transportation of coffee from Brazil to Italy. Six containers (three conventional and three prototype) were stowed in three different places (hold, first floor, and deck) on the ship. Each prototype was located next to a conventional container. The moisture content of the coffee in the container located on the first floor was less affected by environmental variations (0.7%) than that in the hold and on the deck. Coffee located in the hold showed the highest variation in moisture content (3%); in addition, the container showed visible condensation. Coffee transported on the deck showed an intermediary variation in moisture (2%), and there was no visible condensation. The variation in coffee moisture content of the prototype containers was similar to that of the conventional ones, especially in the top layers of coffee bags (2 to 3%), while the increase in water activity was 0.70. This suggests that diffusion of moisture occurs very slowly inside the cargo and that there are thus sufficient time and conditions for fungal growth. The regions of the container near the wall and ceiling are susceptible to condensation since they are close to the headspace with its high relative humidity. Ochratoxin A production occurred in coffee located at the top of the container on the deck and in the wet bags from the hold (those found to be wet on opening the containers at the final destination).  相似文献

A 1.5 V 8 mW BiCMOS video A/D converter has been developed by using a BiCMOS pumping comparator. Combining Bipolar high-speed and good-matching characteristics with CMOS switched capacitor techniques, this A/D converter is suitable for use in battery-operated multimedia terminals.  相似文献
