We have previously reported that vitamin K2 (VK2) but not VK1 has a potent apoptosis-inducing effect on freshly isolated leukemia cells from patients with various types of leukemia. By multi-color flow cytometric analysis using monoclonal antibody (mAb), APO2.7, which detects mitochondrial 7A6 antigen specifically expressed by cells undergoing apoptosis, we further investigated the apoptosis-inducing effect of VK2 on minor populations of leukemic blast cells in bone marrow from patients with myelodysplastic syndrome (MDS) and overt myeloid leukemia (post-MDS AML). Limiting dilution of CD95 (anti-Fas) mAb-treated apoptotic Jurkat cells with nonapoptotic CTB-1 cells revealed that APO2.7-positive Jurkat cells were consistently detectable by flow cytometry when present at levels of at least 5% in the CTB-1 suspension. In patient samples the gating area for leukemic clone was determined using cell surface antigen-specific mAbs conjugated with either fluorescein isothionate (FITC) or phycoerythrin (PE) and subsequently the cells stained with phycoerythrin cyanine (PE-Cy5)-conjugated APO2.7 mAb were assessed within the gating area of the leukemic clone for monitoring apoptosis. Treatment of the bone marrow mononuclear cells with 3-10 microM of VK2 (menaquinone-3, -4 and -5) in vitro potently induced apoptosis of the leukemic blast cells as compared with the untreated control cells in all 15 MDS patients tested. This effect was more prominent on blastic cells than that on mature myeloid cells such as CD34-/CD33+ gated cells. In addition, VK2 performed much less effectively on CD3-positive lymphoid cells. In contrast to VK2, VK1 did not show apoptosis-inducing activity. These data suggest that VK2 may be used for treatment of patients with MDS in blastic transformation. 相似文献

This study presents the effect of direct current (DC) electric field application on ice sterilization. The sample used was Escherichia coli (JM103) in a 0.001, 0.01, or 0.1 mol/L NaCl aqueous solution. The experimental sample was placed in a cuvette with parallel electrode plates (distance between electrodes: 1 mm) and then cooled at −30 °C or −80 °C for 40 min. After freezing, a 100 V DC voltage was applied to the sample in the freezer with a temperature of −30 °C or −80 °C. Consequently, the survival ratio of E. coli in all samples after freezing was lower than that before freezing due to the freezing injury. In conclusion, we found the bactericidal effect of the DC electric fields in the frozen 0.01 mol/L NaCl samples. © 2021 Institute of Electrical Engineers of Japan. A morphometric study has shown that 10% of the fibers of the ulnar nerve should suffice to reinnervate the biceps muscle in brachial plexus palsies. The aim of this study was to evaluate, by a morphometric study using computerized microanalysis, the cross-sectional surface areas of the different collateral and terminal branches of the ulnar nerve. This was expressed in terms of percentage of the cross-sectional surface area of the main trunk of the ulnar nerve. The study revealed that the branch to the flexor digitorum profundus bellies to the ring and little fingers formed 9.5% of the cross-sectional area of the ulnar nerve. Thus use of these fascicles destined for the flexor digitorum profundus, identified by intra-operative nerve stimulation, at the level of the arm would be sufficient for neurotisation of the nerve to the biceps. This paper describes a new class of saturable absorber device based on single-wall carbon nanotube (SWNT)-the saturable absorber incorporating nano tube (SAINT). The device possesses ultrafast optical properties comparable to that of the industrial standard semiconductor saturable absorber mirror (SESAM). Passively mode-locked picosecond fiber lasers in different configurations are demonstrated using SAINTs as mode lockers. This is the first demonstration of optical pulsed lasers based on the carbon nanotube technology, and the first practical application of carbon nanotubes in the field of applied optics. 相似文献

Spontaneous transition of the plasma confinement mode was observed in the helicalaxis heliotron device “Heliotron J“ for three different plasma heating schemes, i.e. ECHonly, NBI-only and the combination of ECH and NBI. The transition seems to occur above a certain critical density. In addition to the confinement transition, a spontaneous shift of the hitting position of the divertor plasma flux on the wall was observed. Rubber is an important industrial material. However, mechanical properties and deformation of the high polymer including rubber are not clear. A method of estimating the mechanical properties which can be used for the deteriorated rubber is demanded to improve the reliability of it. Improved VBO (Viscoplasticity theory based on overstress) model is a constitutive equation. We applied it for the compression stress-strain behavior of deteriorated NBR (acryloNitrile-Butadiene rubber). Furthermore, we proposed a method of estimating the mechanical properties of deteriorated NBR. In this paper, deterioration tests in water and in air were conducted. To confirm the validity of the proposed improved VBO model and estimated mechanical properties, simulations of the stress-strain curves obtained in the tests were conducted. 相似文献

A technique for preparation of a pillar-shaped specimen and its multidirectional observation using a combination of a scanning transmission electron microscope (STEM) and a focused ion beam (FIB) instrument has been developed. The system employs an FIB/STEM compatible holder with a specially designed tilt mechanism, which allows the specimen to be tilted through 360 degrees [T. Yaguchi, M. Konno, T. Kamino, T. Microanal. 10 (Suppl. 2) (2004) 1030]. This technique was applied to obtain the three-dimensional (3D) elemental distributions around a contact plug of a Si device used in a 90-nm technology. A specimen containing only one contact plug was prepared in the shape of a pillar with a diameter of 200nm and a length of 5mum. Elemental maps were obtained from the pillar specimen using a 200-kV cold-field emission gun (FEG) STEM model HD-2300C equipped with the EDAX genesis X-ray energy-dispersive spectrometry (XEDS) system through a spectrum imaging technique. In this study, elemental distributions of minor elements with weak signals were enhanced by applying principal component analysis (PCA), which is a superior technique to extract weak signals from a large dataset. The distributions of elements, especially the metallization component Ti and minor dopant As in this particular device, were successfully extracted by PCA. Finally, the 3D elemental distributions around the contact plug could be visualized by reconstruction from the tilt series of maps. 相似文献

The purpose of this study is to clarify damage process of “Type IV cracking” in weldment of a 2.25Cr-1Mo steel and to propose a micro damage prediction method. From continuous observation under a creep-fatigue test, it was found that spherical shape voids initiated and grew on grain boundaries in fine grain region and these voids grow continuously by changing their shape to crack-like. Both spherical and crack-like void growth rate equations were derived from the proposed void growth model. It was indicated that measured void growth rate under the creep-fatigue loading was well predicted by the growth rate equations. 相似文献
