利用流式细胞仪及荧光定量PCR技术,分别检测细胞凋亡率和DNA损伤修复相关基因mRNA表达变化,以探讨曲古菌素A(Trichostatin A,TSA)能否在体外增强食管癌细胞对X射线的敏感性。结果显示,辐射前TSA的处理显著提高了X射线引起的细胞凋亡率(P〈0.05);X射线的单独处理显著上调了基因ATM、XRCC2和Lig4(8Gy除外)的mRNA水平(P〈0.05),TSA单独和TSA联合X射线都下调了ATM、XRCC2和Lig4的mRNA水平。提示TSA很可能通过下调ATM、Lig4和XRCC2mRNA水平使食管癌细胞对X射线敏感。 相似文献

The whole DNA of soybean was implanted into four varieties of wheat of Zhougyu 5,Huaiyin 9628,Wenyou 1,Jimai 5 respectively via ion-beam mediation.There were 5 plants obtained whose protein content was higher than 18.5%,the highest one was 21.44%,There were 3 plants obtained whose protein content was lower than 11.5%,the lowest one was 10.96%.We can see that the whole DNA of soybean transformed into wheat via ion beam implantation can induce the increase in wheat protein content dramatically.The result also shows that the transformation efficiency of different gene types of wheat receptor varies greatly that the implanting time has a certain effect on the efficiency of transformation. 相似文献

From a population of about 3500 single plants in Arabidopsis promoter trapping bank, one plant whose GUS-gene had been specifically expressed in vascular bundle, was screened by the method of gus tissue staining. The T-DNA flanking sequence was amplified using TAIL-PCR. This band will be purified and connected to TA cloning vector. 本实验利用正己烷萃取技术、超临界CO2萃取技术和亚临界低温萃取技术三种方法制备得到了三种栀子果油,并对其主要理化指标、脂肪酸组成、植物甾醇含量、谷维素含量、维生素E含量及氧化酸败时间进行比较分析,以研究不同萃取方法对栀子果油品质的影响。结果表明三种栀子果油的过氧化值为9.38~17.30 mmol/kg,酸值为4.06~14.28 mg KOH/g,皂化值为186.07~198.91 mg KOH/g,碘值为116.43?10-2~130.16?10-2 g I2/g。气相色谱法测定其脂肪酸组成以油酸和亚油酸等不饱和脂肪酸为主,相对含量占76.04~78.00%。栀子果油中植物甾醇和维生素E的含量分别是0.85~3.48%和17.06?10-2~29.80?10-2 mg/g,谷维素含量极少。其中超临界栀子果油的色泽最好,溶剂残留量最少,但酸价最高;正己烷栀子果油中甾醇含量最高,氧化稳定性相对较好;亚临界栀子果油的酸值最低,维生素E含量最高。本文为栀子果油制备方法的选择和精炼提供了理论依据,具有一定的应用意义。 相似文献

分析比较了亚临界丁烷/二甲醚萃取法、超临界CO2萃取法和乙醇浸提法所得到的小茴香籽提取物,利用气相色谱-质谱法对不同方法得到的净油进行了化学成分分析。经研究发现,小茴香籽所含挥发油占总提取物的3%~6%,其中含量最高的2种化合物是草蒿脑和反式茴香脑,都属于醚类。亚临界萃取对弱极性物质有较好的提取效果,所得净油中有机酸含量较高;超临界CO2萃取法对极性混合物提取效果较好,得到的净油种类最多;有机溶剂萃取对极性物质提取效果较好。通过比较不同萃取方法所得小茴香籽净油的成分,以期为以后有针对性的提取小茴香籽的特定成分、进一步对其开发利用提供数据支持。 相似文献

Pollen grains of Pinus thunbergii Parl. (Japanese black pine) were implanted with 30 keV nitrogen ion beams and the effects of nitrogen ion implantation on pollen tube growth in vitro and the organization of actin cytoskeleton in the pollen tube cell were investigated using a confocal laser scanning microscope after fluorescence labeling. Treatment with ion implantation significantly blocked pollen tube growth. Confocal microscopy showed that ion implantation disrupted actin filament cytoskeleton organization in the pollen tube. It was found that there was a distinct correlation between the inhibition of pollen tube growth and the disruption of actin cytoskeleton organization, indicating that an intact actin cytoskeleton is essential for continuous pollen tube elongation in Pinus thunbergii. Although the detailed mechanism for the ion-implantation-induced bioeffect still remains to be elucidated, the present study assumes that the cytoskeleton system in pollen grains may provide a key target in response to ion beam implantation and is involved in mediating certain subsequent cytological changes. 相似文献
