Lignin fibers were developed from a commercial available soda hardwood lignin (SHL) with a melt‐spinning approach. SHL showed spinnability to form the fine fibers when poly(ethylene oxide) was used as a plasticizer with lignin. The thermal properties of lignin provided valuable information to assist the processing steps of the lignin fiber formation. The guaiacyl/syringyl ratio in SHL was determined by 31P‐NMR because it had great influence on the thermal mobility of lignin. Molybdenum (Mo) nitrogenases consist of two components: dinitrogenase reductase (encoded by nifH) and the dinitrogenase or MoFe protein (encoded by nifDK). Nitrogenase enzyme of photosynthetic bacteria is responsible for hydrogen production. Therefore, primers were designed for the nitrogenase gene only. In this study, two primers (ND and NH) were designed after comparative genomic analysis of nifH and nifD gene sequences from public databases. The designed primers were used for the amplification of nifH and nifD genes to detect nitrogenase genes in photosynthetic bacteria. Initial detection was done using a monoplex Polymerase Chain Reactions (PCRs) followed by optimization of the PCR protocols. Subsequently, a duplex PCR was designed for amplification and detection of nifH and nifD genes in indigenous photosynthetic bacteria. Evaluation of the duplex PCR on six samples isolated from Palm Oil Mill Effluent (POME) showed that only four isolates contained both the nifH and nifD genes, indicating that these isolates were potential hydrogen-producing bacteria. PCR detection provides a rapid and efficient pre-identification of potential photosynthetic bacterial hydrogen producers. 相似文献

Thermostable enzymes are currently being investigated to improve industrial processes of starch saccharification. A novel glucoamylase was purified to electrophoretic homogeneity from the culture supernatant of Fusarium solani on a fast protein liquid chromatographic system (FPLC). The recovery of glucoamylase after gel filtration on FPLC was 31.8% with 26.2-fold increase in specific activity. The enzyme had a molecular mass of 40 kDa by SDS-PAGE and 41 kDa by gel filtration. The glucoamylase exhibited optimum activity at pH 4.5. The Kcat and Km were 441/min and 1.9 mg/ml, respectively, for soluble starch, specificity constant (Kcat/Km) was 232. The enzyme was thermally stable at 50 °C and retained 79% activity after 60 min at this temperature. The half-life of the enzyme was 26 min at 60°C. The enzyme was slightly stimulated by Cu2+ and Mg2+ and strongly inhibited by Hg2+, Pb2+, Zn2+, Ni2+ and Fe3+. 相似文献

The volatile components of dalieb fruit have been investigated by capillary column gas chromatography/mass spectrometry. The main components were C1 to C4 aliphatic alcohols and the corresponding acids together with esters derived from them. Acetic acid and ethanol were particularly abundant, the fruit pulp containing approximately 0.1% of each compound. Energy and exergy analyses previously performed by the authors for a single effect absorption refrigeration system have been extended to double effect vapor absorption refrigeration system with the expectation of reducing energy supply as well as an interest in the diversification of the motive power employed by HVAC technologies. The total exergy destruction in the system as a percentage of the exergy input from a generator heating water over a range of operating temperatures is examined for a system operating on LiBr–H2O solution. The exergy destruction in each component, the coefficient of performance (COP) and the exergetic COP of the system are determined. It is shown that exergy destructions occur significantly in generators, absorbers, evaporator2 and heat exchangers while the exergy destructions in condenser1, evaporator1, throttling valves, and expansion valves are relatively smaller within the range of 1–5%. The results further indicate that with an increase in the generator1 temperature the COP and ECOP increase, but there is a significant reduction in total exergy destruction of the system for the same. On the other hand, the COP and ECOP decrease with an increase in the absorber1 temperature while the total exergy destruction of the system increases significantly with a small increase in the absorber1 temperature. The results show that the exergy method can be used as an effective criterion in designing an irreversible double effect absorption refrigeration system and may be a good tool for the determination of the optimum working conditions of such systems. An optimisation study of the large‐scale enzymatic synthesis of a liquid wax ester from oleic acid and oleyl alcohol using Novozym 435 was carried out. Investigations were performed in batch mode with a stirred tank reactor (STR) with one multi‐bladed impeller. Response surface methodology (RSM) based on a five‐level, three‐variable central composite rotatable design (CCRD) was used to evaluate the interactive effects of various parameters. The parameters are amount of enzyme (A) (90–120 g), impeller speed (B) (100–400 rpm) and temperature (C) (40–60 °C). The optimum conditions derived via RSM at a fixed reaction time of 1 h were successfully optimised as A = 104 g, B = 388.0 rpm and C = 49.7 °C. The actual experimental yield was 96.7% under the optimum conditions, which compared well with the maximum predicted value of 97.6%. Copyright © 2005 Society of Chemical Industry 相似文献

Formulation of a cost‐effective, high‐performance and eco‐friendly lubricant, largely depends on the base oil quality, then the selection of suitable additives and their proportions. Vegetable oils, identified to be eco‐friendly, renewable, future‐available and cost‐effective treasures for lubricant formulation, apart from processing, will rely much on suitable additives to meet the performance requirements for automotive shock absorber (ASA) fluids. Additives that will guarantee performance, longevity and eco‐friendliness of formulated vegetable‐based functional fluids have to be uncommonly effective, resistant to depletion, non‐toxic and highly biodegradable. Their selection in these regards will require skills and experience, which will harness the various arms of synergism as effective tools to succour the known weaknesses of the base oil. This is a review on additives that could be used in formulation of vegetable oil‐based (ASA) fluids. Inhibitors of the human enzyme dimethylarginine dimethylaminohydrolase‐1 (DDAH‐1) can raise endogenous levels of asymmetric dimethylarginine (ADMA) and lead to a subsequent inhibition of nitric oxide synthesis. In this study, N5‐(1‐imino‐2‐chloroethyl)‐L ‐ornithine (Cl‐NIO) is shown to be a potent time‐ and concentration‐dependent inhibitor of purified human DDAH‐1 (KI=1.3±0.6 μM ; kinact=0.34±0.07 min?1), with >500‐fold selectivity against two arginine‐handling enzymes in the same pathway. An activity probe is used to measure the “in cell” IC50 value (6.6±0.2 μM ) for Cl‐NIO inhibition of DDAH‐1 artificially expressed within cultured HEK293T cells. A screen of diverse melanoma cell lines reveals that a striking 50/64 (78 %) of melanoma lines tested showed increased levels of DDAH‐1 relative to normal melanocyte control lines. Treatment of the melanoma A375 cell line with Cl‐NIO shows a subsequent decrease in cellular nitric oxide production. Cl‐NIO is a promising tool for the study of methylarginine‐mediated nitric oxide control and a potential therapeutic lead compound for other indications with elevated nitric oxide production, such as septic shock and idiopathic pulmonary fibrosis. 相似文献

In this study, we devised a new concept for the precise nanofabrication of Au-Si fibrous nanostructures using megahertz femtosecond laser irradiation in air and atmospheric pressure conditions. The weblike fibrous nanostructures of Au thin layer on silicon substrate, which are proposed for the application of solar cells, exhibit a specific improvement of the optical properties in visible wavelength. Varying numbers of laser interaction pulses were used to control the synthesis of the nanofibrous structures. Electron microscopy analysis revealed that the nanostructures are formed due to the aggregation of polycrystalline nanoparticles of the respective constituent materials with diameters varying between 30 and 90 nm. Measurement of the reflectance through a spectroradiometer showed that the coupling of incident electromagnetic irradiation was greatly improved over the broadband wavelength range. Lower reflectance intensity was obtained with a higher number of laser pulses due to the bulk of gold nanoparticles being agglomerated by the mechanism of fusion. This forms interweaving fibrous nanostructures which reveal a certain degree of assembly.
