Monogenic hypertension is rare and caused by genetic mutations, but whether factors associated with mutations are disease-specific remains uncertain. Given two factors associated with high mutation rates, we tested how many previously known genes match with (i) proximity to telomeres or (ii) high adenine and thymine content in cardiovascular diseases (CVDs) related to vascular stiffening. We extracted genomic information using a genome data viewer. In human chromosomes, 64 of 79 genetic loci involving >25 rare mutations and single nucleotide polymorphisms satisfied (i) or (ii), resulting in an 81% matching rate. However, this high matching rate was no longer observed as we checked the two factors in genes associated with essential hypertension (EH), thoracic aortic aneurysm (TAA), and congenital heart disease (CHD), resulting in matching rates of 53%, 70%, and 75%, respectively. A matching of telomere proximity or high adenine and thymine content projects the list of loci involving rare mutations of monogenic hypertension better than those of other CVDs, likely due to adoption of rigorous criteria for true-positive signals. Our data suggest that the factor–disease matching rate is an accurate tool that can explain deleterious mutations of monogenic hypertension at a >80% match—unlike the relatively lower matching rates found in human genes of EH, TAA, CHD, and familial Parkinson’s disease. 相似文献

The influence of PVT1 and MALAT1 variants on colorectal cancer (CRC) susceptibility and their impact on PVT1/miRNA-186/epithelial-mesenchymal transition (EMT) and MALAT1/miRNA-101/EMT axes in CRC are unknown. We investigated the influence of PVT1 rs13255292 and MALAT1 rs3200401 on the risk of CRC and adenomatous polyps (AP), their impact on the long noncoding RNAs PVT1 and MALAT1 expression and their target miRNA-186, miRNA-101/E-cadherin pathways, along with their potential as early CRC biomarkers. Overall, 280 individuals were recruited: 140 patients with CRC, 40 patients with AP, and 100 healthy volunteers. Genotyping and serum expression profiles were assessed using qPCR. The EMT biomarker, E-cadherin, was measured by ELISA. rs3200401 was associated with increased CRC risk, whereas rs13255292 was protective. Serum PVT1 and MALAT1 were upregulated in CRC and AP patients versus healthy controls, whereas, miRNA-186, miRNA-101 and E-cadherin were downregulated in CRC versus non-CRC groups. MALAT1 showed superior diagnostic potential for CRC and predicted CRC risk among non-CRC groups in the multivariate logistic analysis. PVT1, MALAT1, miRNA-186 and miRNA-101 levels were correlated with E-cadherin, tumor stage, lymph node and distant metastasis. E-cadherin was lost in metastatic vs. non-metastatic CRC. rs3200401CC genotype carriers showed higher E-cadherin levels than CC + CT carriers. rs3200401 was correlated with lymph node status. For the first time, rs13255292 and rs3200401 are potential genetic CRC predisposition markers, with rs3200401 possibly impacting the EMT process. Serum PVT1, MALAT1, miRNA-186 and miRNA-101 are novel non-invasive diagnostic biomarkers that could improve the clinical outcome of CRC. 相似文献

High temperatures appear on friction clutch surfaces due to the heat generated between the contacting surfaces during the slipping period. In some cases the maximum temperatures will exceed the allowable working temperature; friction clutches will fail rapidly when working under these conditions for a long time. Therefore, it is necessary to study the effect of design parameters (e.g., grooves) on the thermoelastic behavior of friction clutches to avoid these kinds of failures or at least increase the lifetime of friction clutch. A finite element method will be used to investigate the effect of the circumferential groove on the thermoelastic behavior of a single-disc clutch during a sliding phase. Axisymmetric models have been developed in this analysis to simulate the dry clutch systems during the sliding period. The effect of the groove area ratio G.R (defined as the groove area divided by the nominal contact area) on thermal and elastic behaviors is investigated. The results showed that the groove size has a significant effect on the magnitude and distribution of contact pressure, temperature field and heat generated along the frictional surfaces. 相似文献

Vanda Mimi Palmer (VMP), an orchid hybrid of Vanda tesselata and Vanda Tan Chay Yan is a highly scented tropical orchid which blooms all year round. Previous studies revealed that VMP produces a variety of isoprenoid volatiles during daylight. Isoprenoids are well known to contribute significantly to the scent of most fragrant plants. They are a large group of secondary metabolites which may possess valuable characteristics such as flavor, fragrance and toxicity and are produced via two pathways, the mevalonate (MVA) pathway or/and the 2-C-methyl-D-erythritol-4-phosphate (MEP) pathway. In this study, a sesquiterpene synthase gene denoted VMPSTS, previously isolated from a floral cDNA library of VMP was cloned and expressed in Lactococcus lactis to characterize the functionality of the protein. L. lactis, a food grade bacterium which utilizes the mevalonate pathway for isoprenoid production was found to be a suitable host for the characterization of plant terpene synthases. Through recombinant expression of VMPSTS, it was revealed that VMPSTS produced multiple sesquiterpenes and germacrene D dominates its profile. 相似文献

Calcium borate nanoparticles have been synthesized by a thermal treatment method via facile co-precipitation. Differences of annealing temperature and annealing time and their effects on crystal structure, particle size, size distribution and thermal stability of nanoparticles were investigated. The formation of calcium borate compound was characterized by X-ray diffraction (XRD) and Fourier Transform Infrared spectroscopy (FTIR), Transmission electron microscopy (TEM), and Thermogravimetry (TGA). The XRD patterns revealed that the co-precipitated samples annealed at 700 °C for 3 h annealing time formed an amorphous structure and the transformation into a crystalline structure only occurred after 5 h annealing time. It was found that the samples annealed at 900 °C are mostly metaborate (CaB2O4) nanoparticles and tetraborate (CaB4O7) nanoparticles only observed at 970 °C, which was confirmed by FTIR. The TEM images indicated that with increasing the annealing time and temperature, the average particle size increases. This paper presents a numerical study of the effect of oscillation amplitude in oscillatory baffled column (OBC) using computational fluid dynamics. The numerical work was carried out for single phase liquid flow for an unsteady 3-D model using commercial software, Fluent (2006). This work was concentrated on the effect of oscillation amplitude. Three amplitudes of 5, 10 and 15 mm with constant frequency of 1 Hz are applied. Vortex and cycle average velocities at different points are analyzed. The studies show the maximum velocity for 5 mm, 10 mm and 15 mm in an OBC are 0.11 m/s, 0.25 m/s and 0.40 m/s respectively in the first cycle of oscillation. At a constant frequency, greater oscillation amplitude displaces the liquid to a further distance and builds a larger vortex. Vortex length was 1.5 times bigger when oscillation amplitude changes from 5 mm to 10 mm and 2 times when the amplitude is triple from 5 mm. In this contribution, we reported the investigation of natural rubber (NR) reinforced by silicon dioxide‐graft‐polyisoprene (SiO2‐g‐PIP) core–shell nanoparticle. First, the hydroxyl on the surface of the SiO2 nanoparticles was reacted with 2‐methyl‐2‐[(dodecylsulfanylthiocarbonyl) sulfanyl] propanoic acid to produce trithioester‐capped SiO2 (denoted as SiO2‐CTA). SiO2‐CTA was used as a nanoparticle chain transfer agent, and SiO2‐g‐PIP core–shell nanoparticles were synthesized via reversible addition‐fragmentation chain transfer polymerization (RAFT). The results of FTIR spectroscopy and TGA showed that the grafting weight of the PIP block in SiO2‐g‐PIP was 2.1 wt%. SiO2‐g‐PIP and SiO2 were simultaneously incorporated into NR. The curing properties of the NR compounds showed that the vulcanization rates of the NR/SiO2‐g‐PIP compounds were much higher than those of NR/silica compounds. The results of scanning electron microscopy showed that SiO2 microdomains in the NR/SiO2‐g‐PIP vulcanizate were much better disperse and distribute than SiO2 microdomains in the NR/SiO2 composite. A conductometric study of the mixed micellization behavior between cetyltrimethylammonium bromide (CTAB, a cationic surfactant) and sodium dodecyl sulfate (SDS, an anionic surfactant) was carried out in the absence/presence of various percentages of chitosan in the temperature range of 298.15–318.15 K. The deviations of critical micelle concentration (cmc) from the ideal values indicate the interaction between CTAB and SDS. The micellar mole fraction values according to different proposed models X1Rub (Rubingh), X1M (Motomura), X1Rod (Rodenas), and X1id (ideal mole fraction) were estimated and the results obtained reveal the high contribution of CTAB in the mixed micellization, which enhances with the increase of the mole fraction of CTAB. The negative magnitudes of indicate the spontaneous formation of mixed micelles between CTAB and SDS. The values of activity coefficients (f1 and f2) were less than unity and the values of the interaction parameter (β) are negative in all cases, which indicate the attractive interaction between CTAB and SDS. The negative values of excess free energy of micellization (ΔGex) signify the stability of the mixed micelles. The negative values of in the chitosan systems indicate that micellization is exothermic. The values of were found to be positive in all cases. 相似文献

Middle molecule uremic toxins constitute to a quarter of uremic toxins present in human blood. In a condition where these uremic toxins accumulate in bloodstream due to renal failure, blood purification process using a high performance membrane is required. Here, we develop biocompatible mixed matrix membranes (MMMs) made up of polysulfone (PSf) and iron oxide nanoparticles (Fe2O3 NPs) with the focus to remove middle molecule uremic toxin effectively. The MMMs were evaluated in terms of their biocompatibility and separation performance. At higher Fe2O3 NPs loading, the MMM displayed a huge reduction of protein adsorption and platelet adhesion while maintaining normal blood coagulation time and acceptable complement activation. The optimized MMM exhibited high permeability (110.47 L m−2 h−1 bar−1), protein retention (99.9%) and demonstrated excellent clearance of urea (82%) and lysozyme (46.7%).
