High level languages used for programming microprocessors still need some assembly code to cover all the programming requirements and aspects. Using assembly code in a high level language program introduces certain difficulties and disadvantages. We describe six new yeast episomal vectors which encode glutathione S-transferase (GST) affinity tags. These allow for the production of GST-fusion proteins in Saccharomyces cerevisiae under the control of the CUP1 promoter. Affinity chromatography with glutathione-Sepharose permits convenient purification of the fusion protein from a yeast lysate. The presence of a protease cleavage site facilitates subsequent removal of the GST tag. The expression and single-step purification of both GST and a functional GST-metallothionein fusion from yeast are shown as an example of the application of these vectors.  相似文献

Introduced a laboratory measure of motor function, synchronization of tapping with an auditory stimulus, as an index of motor disorganization in schizophrenia. It was hypothesized that (a) schizophrenics' performance on this task would be distinguishable from that of controls and would reflect a relative inability to take advantage of stimulus predictability, independent of task difficulty; (b) clinical measures of disturbed motor behavior would be associated with poorer synchronization; and (c) synchronization performance would be associated with clinical ratings of formal thought disorder and type-token ratio assessment of disorganization in spoken language. 16 schizophrenic, 8 affective, and 8 normal controls (18–48 yrs) were studied. The schizophrenics showed a specific pattern of disrupted synchrony not consistent with explanations based on S motivation, task difficulty, motor dexterity, a general psychosis factor, drug effects, or tapping speed ability. Deficient motor performance was associated with clinical evidence of abnormal movement and disturbed thinking and with the type-token ratio. In order to utilise sardinelle (Sardinellaaurita) protein by-products, which is normally discarded as industrial waste in the process of fish manufacturing, heads and viscera proteins were hydrolysed by different proteases to obtain antioxidative peptides. All hydrolysates showed different degrees of hydrolysis and varying degrees of antioxidant activities. Hydrolysate generated with crude enzyme extract from sardine (Sardinapilchardus) displayed high antioxidant activity, and the higher DPPH radical-scavenging activity (87 ± 2.1% at 2 mg/ml) was obtained with a degree of hydrolysis of 6%. This hydrolysate was fractionated by size exclusion chromatography on a Sephadex G-25 into eight major fractions (P1–P8). Fraction P4, which exhibited the highest DPPH scavenging activity, was then fractionated by reversed-phase high performance liquid chromatography (RP-HPLC). Seven antioxidant peptides were isolated. The molecular masses and amino acids sequences of the purified peptides were determined using ESI-MS and ESI-MS/MS, respectively. Their structures were identified as Leu-His-Tyr, Leu-Ala-Arg-Leu, Gly-Gly-Glu, Gly-Ala-His, Gly-Ala-Trp-Ala, Pro-His-Tyr-Leu and Gly-Ala-Leu-Ala-Ala-His. The first peptide displayed the highest DPPH radical-scavenging activity (63 ± 1.57%; at 150 μg/ml) among these peptides.  相似文献

A new method was developed to determine the presence of some refined vegetable oils in olive oil based on the sum of campesterol and stigmasterol percentages. Model systems of corn, soybean, sunflower and cotton seed oils in olive oil at levels of 5%, 10% and 20% were prepared. The unsaponifiables of these model systems were analysed by GLC using polar column with high thermal stability. An olive oil authenticity factor based on the summation of campesterol and stigmasterol percentages was established as an indicator of olive oil adulteration with vegetable oils. The results indicate the possibility to detect the presence as little as 5% of these plant oils in olive oil.  相似文献

BACKGROUND: Plant essential oils are widely used as fragrances and flavours. Therefore, the essential oils from the leaves of Cinnamomum pubescens Kochummen (CP) and the whole plant of Etlingera elatior (EE) were investigated for their antioxidant, antibacterial and phytochemical properties. RESULTS: CP and EE were found to contain appreciable levels of total phenolic contents (50.6 and 33.41 g kg?1 as gallic acid equivalent) and total flavonoid contents (205.6 and 244.8 g kg?1 as rutin equivalent), respectively. DPPH free radical scavenging activity of CP is superior to EE (P < 0.05) showing IC50 of 77.2 and 995.1 µg mL?1, respectively. Methicillin‐resistant Staphylococcus aureus (MRSA), Bacillus subtilis, Pseudomonas aeruginosa and Salmonella choleraesuis were tested against CP and EE. Only MRSA was the most susceptible bacteria to CP. GC/MS studies resulted in the identification of 79 and 73 compounds in CP and EE, respectively. The most abundant components of EE included β‐pinene (24.92%) and 1‐dodecene (24.31%). While the major compound in CP were 1,6‐octadien‐3‐ol,3,7‐dimethyl (11.55%), cinnamaldehyde (56.15%) and 1‐phenyl‐propane‐2,2‐diol diethanoate (11.38%). CONCLUSION: This study suggests that the essential oils from Cinnamomum pubescens Kochummen and Etlingera elatior could be potentially used as a new source of natural antioxidant and antibacterial in the food and pharmaceutical industries. In this paper, a new framework for designing static and low order anti-windup compensator (AWC) for industrial cascade control systems with actuator saturation constraint is presented. Based on less conservative block diagonal quadratic Lyapunov function, sector boundedness, decoupled architecture, norm reduction and cascade loop compensation, linear matrix inequalities are developed which guarantee stability and suitable performance for overall closed-loop system. Static AWC parameters are obtained by comparing the full order AWC architecture with generalized architecture for cascade control system. Low order AWC is designed by sub-optimal approach in which AWC weights are tuned by designer. Anti-windup compensator is divided into inner and outer loop compensators which compensate the effect of saturation at each level. It is observed that the proposed methodology is less conservative than the traditional AWC schemes when applied to cascade control systems. The proposed scheme is successfully tested experimentally on a temperature-based process control system and results are outlined.  相似文献
