This study examined the use of a low-cost incentive program to decrease the rate of unanticipated no-shows in a nontreatment study of the genetics of substance dependence. Low-cost retail items (such as calling cards or gift certificates) were offered contingent on attendance at the first scheduled research appointment. Although the intervention did not result in an increase in the rate of attendance at appointments, it reduced the likelihood of unanticipated no-shows by almost 50%. This reduction was accompanied by a significant increase in advance cancellations. Despite limitations due to the study's A-B design, this research demonstrates that it may be possible to use modest incentives to increase the efficiency of research enrollment and increase service provision for substance abusers. A study was conducted to verify the efficacy of a fracture mechanics methodology to model the crack growth behavior of fretting fatigue-nucleated cracks obtained under test conditions similar to those found in turbine engine blade attachments. Experiments were performed to produce cracked samples, and fretting fatigue crack propagation lives were calculated for each sample. Cracks were generated at 106 cycles (10%-of-life) under applied stress conditions previously identified as the fretting fatigue limit conditions for a 107 cycle fatigue life. Resulting cracks, ranging in size from 30 to 1200 μm, were identified and measured using scanning electron microscopy. Uniaxial fatigue limit stresses were determined experimentally for the fretting fatigue-cracked samples, using a step loading technique, for R=0.5 at 300 Hz. Fracture surfaces were inspected to characterize the fretting fatigue crack front indicated by heat tinting. The shape and size of the crack front were then used in calculating ΔKth values for each crack. The resulting uniaxial fatigue limit and ΔKth values compared favorably with the baseline fatigue strength (660 MPa) for this material and the ΔKth value (2.9 MPa√m) for naturally initiated cracks tested at R=0.5 on a Kitagawa diagram.Crack propagation lives were calculated using stress results of FEM analysis of the contact conditions and a weight function method for determination of ΔK. Resulting lives were compared with the nine million-cycle propagation life that would have been expected in the experiments, if the contact conditions had not been removed. Scatter in the experimental results for fatigue limit stresses and fatigue lives had to be considered as part of an explanation why the fatigue life calculations were unable to match the experiments that were modeled. Analytical life prediction results for the case where propagation life is observed to be very short experimentally were most accurate when using a coefficient of friction, μ=1.0, rather than for the calculations using μ=0.3  相似文献

Supporting Mobile Commerce Applications Using Dependable Wireless Networks   总被引:1,自引:0,他引:1  
Mobile commerce (m-commerce) is an emerging discipline involving applications, mobile devices, middleware, and wireless networks. While most of existing e-commerce applications can be modified to run in a wireless environment, m-commerce also involves many more new applications that become possible only due to the wireless infrastructure. These applications include mobile financial services, user and location specific mobile advertising, mobile inventory management, wireless business re-engineering, and mobile interactive games. In addition to device and wireless constraints, mobile commerce would also be impacted by the dependability of wireless infrastructure. Unlike e-commerce applications that usually run on fixed networking infrastructure of fairly high dependability (approaching to about 100%), m-commerce applications may not receive such high dependability from the existing and emerging wireless infrastructure. So far, most of the m-commerce research focuses on applications, devices, and security issues. We believe that some work is necessary in addressing the dependability challenges of the wireless infrastructure. In this paper, we present (a) the dependability issues of wireless infrastructure, (b) several architectures to improve the dependability of wireless networks, and (c) a simulation model and results on wireless dependability for m-commerce. The results show that a significant improvement is possible in wireless dependability for supporting m-commerce applications.  相似文献

Meat quality can be affected by stress, exhaustion, feed composition, and other physical and environmental conditions. These stressors can alter the pH in postmortem muscle, leading to high pH and low-quality dark cutting (DC) beef, resulting in considerable economic loss. Moreover, the dark cutting prediction may equally provide a measure for animal welfare since it is directly related to animal stress. There are two needs to advance on-site detection of dark cutters: (1) a clear indication that biomarker (signature compounds) levels in cattle correlate with stress and DC outcome; and (2) measuring these biomarkers rapidly and accurately on-farm or the abattoir, depending on the objectives. This critical review assesses which small molecules and proteins have been identified as potential biomarkers of stress and dark cutting in cattle. We discuss the potential of promising small molecule biomarkers, including catecholamine/cortisol metabolites, lactate, succinate, inosine, glucose, and β-hydroxybutyrate, and we identify a clear research gap for proteomic biomarker discovery in live cattle. We also explore the potential of chemical-sensing and biosensing technologies, including direct electrochemical detection improved through nanotechnology (e.g., carbon and gold nanostructures), surface-enhanced Raman spectroscopy in combination with chemometrics, and commercial hand-held devices for small molecule detection. No current strategy exists to rapidly detect predictive meat quality biomarkers due to the need to further validate biomarkers and the fact that different biosensor types are needed to optimally detect different molecules. Nonetheless, several biomarker/biosensor combinations reported herein show excellent potential to enable the measurement of DC potential in live cattle.  相似文献

Casein kinase II (CK2) and cyclin-dependent kinases (CDKs) frequently interact within multiple pathways in pancreatic ductal adenocarcinoma (PDAC). Application of CK2- and CDK-inhibitors have been considered as a therapeutic option, but are currently not part of routine chemotherapy regimens. We investigated ten PDAC cell lines exposed to increasing concentrations of silmitasertib and dinaciclib. Cell proliferation, metabolic activity, biomass, and apoptosis/necrosis were evaluated, and bioinformatic clustering was used to classify cell lines into sensitive groups based on their response to inhibitors. Furthermore, whole exome sequencing (WES) and RNA sequencing (RNA-Seq) was conducted to assess recurrent mutations and the expression profile of inhibitor targets and genes frequently mutated in PDAC, respectively. Dinaciclib and silmitasertib demonstrated pronounced and limited cell line specific effects in cell death induction, respectively. WES revealed no genomic variants causing changes in the primary structure of the corresponding inhibitor target proteins. RNA-Seq demonstrated that the expression of all inhibitor target genes was higher in the PDAC cell lines compared to non-neoplastic pancreatic tissue. The observed differences in PDAC cell line sensitivity to silmitasertib or dinaciclib did not depend on target gene expression or the identified gene variants. For the PDAC hotspot genes kirsten rat sarcoma virus (KRAS) and tumor protein p53 (TP53), three and eight variants were identified, respectively. In conclusion, both inhibitors demonstrated in vitro efficacy on the PDAC cell lines. However, aberrations and expression of inhibitor target genes did not appear to affect the efficacy of the corresponding inhibitors. In addition, specific aberrations in TP53 and KRAS affected the efficacy of both inhibitors.  相似文献

The novel application of two‐dimensional (2D) single‐layer ternary chalcogenide nanosheets as “capture‐release” fluorescence‐based biomolecular nanosensors is demonstrated. Fluorescently labeled biomolecular probe is first captured by the ultrathin Ta2NiS5 nanosheets and then released upon adding analyte containing a target biomolecule due to the higher probe‐target affinity. Here, the authors use a nucleic acid probe for the model target biomolecule Plasmodium lactate dehydrogenase, which is an important malarial biomarker. The ultrathin Ta2NiS5 nanosheet serves as a highly efficient fluorescence quencher and the nanosensor developed from the nanosheet is highly sensitive and specific toward the target biomolecule. Apart from the specificity toward the target biomolecule in homogeneous solutions, the developed nanosensor is capable of detecting and differentiating the target in heterogeneous solutions consisting of either a mixture of biomolecules or serum, with exceptional specificity. The simplicity of the “capture‐release” method, by eliminating the need for preincubation of the probe with the test sample, may facilitate further development of portable and rapid biosensors. The authors anticipate that this ternary chalcogenide nanosheet‐based biomolecular nanosensor will be useful for the rapid detection and differentiation of a wide range of chemical and biological species.  相似文献

We have developed a preliminary method for the qualitative assessment of polycyclic aromatic hydrocarbon (PAH) tetrol metabolites in humans using serum albumin adducts. The method was used to investigate the presence of tetrol metabolites of PAHs released following acid hydrolysis of serum albumin-diol epoxide adducts. Liquid-liquid extraction and solid phase extraction were used for isolation and purification of the tetrol metabolites. Purified tetrols were derivatized to convert them to their trimethylsilyl derivatives and analyzed using gas chromatography/high resolution mass spectrometry. A total of 26 tetrol isomers were investigated, resulting in observation of the following 8 tetrols in 3 smoking and 7 non-smoking donors: 5-methylchrysene-1,2,3,4-anti-tetrol, benz(a)anthracene-1,2,3,4-anti-tetrol, benz(a)anthracene-1,2,3,4-syn-tetrol, benz(a)anthracene-8,9,10,11-anti-tetrol, (±)-benzo(a)pyrene-r-7,t-8,9,c-10-tetrahydrotetrol (BPTI-1), (±)-benzo(a)pyrene-r-7,t-8,c-9,t-10-tetrahydrotetrol (BPTII-1), (±)-benzo(a)pyrene-r-7,t-8,c-9,10-tetrahydrotetrol (BPTII-2), and (±)-benzo(a)pyrene-r-7,t-8,9,10-tetrahydrotetrol (BPTI-2). This preliminary work presents the first reported method for the assessment of PAH multi-analyte tetrol isomers extracted from serum albumin adducts in human donors.  相似文献

Stem cell-based therapies carry significant promise for treating human diseases. However, clinical translation of stem cell transplants for effective treatment requires precise non-destructive evaluation of the purity of stem cells with high sensitivity (<0.001% of the number of cells). Here, a novel methodology using hyperspectral imaging (HSI) combined with spectral angle mapping-based machine learning analysis is reported to distinguish differentiating human adipose-derived stem cells (hASCs) from control stem cells. The spectral signature of adipogenesis generated by the HSI method enables identifying differentiated cells at single-cell resolution. The label-free HSI method is compared with the standard techniques such as Oil Red O staining, fluorescence microscopy, and qPCR that are routinely used to evaluate adipogenic differentiation of hASCs. HSI is successfully used to assess the abundance of adipocytes derived from transplanted cells in a transgenic mice model. Further, Raman microscopy and multiphoton-based metabolic imaging is performed to provide complementary information for the functional imaging of the hASCs. Finally, the HSI method is validated using matrix-assisted laser desorption/ionization-mass spectrometry imaging of the stem cells. The study presented here demonstrates that multimodal imaging methods enable label-free identification of stem cell differentiation with high spatial and chemical resolution.  相似文献

This study was conducted using response surface methodology to predict fluid loss from injected beef strip steaks as influenced by levels of salt and sodium phosphates (SP) in the injection brine. Also, a beef-based dehydrated beef protein (DBP) water binding ingredient was evaluated. Paired U.S. select beef strip loins were quartered before being injected with 110% of initial weight with brines containing various concentrations of salt and SP (CON) or salt, SP and 5% DBP. Steaks were sliced, overwrapped and stored in the dark for 4 d. Purge values ranged from 0.6% to 4.6% for CON and 0.3% to 2.1% for DBP. Fluid losses when accounting for the fluid lost from injection to slicing were as high as 6.8% for CON brines, but only 2.8% for DBP brines. The equations generated here and the DBP product could help producers achieve acceptable purge while reducing sodium use.  相似文献

We compared food choice and the initial response to deterrent treated diet between fifth instars of Helicoverpa armigera, a polyphagous generalist pest, and Bombyx mori, an oligophagous specialist beneficial. Bombyx mori was more behaviorally sensitive to salicin than to caffeine. The relative sensitivities were reversed for H. armigera, which was tolerant to the highest levels of salicin found in natural sources but sensitive to caffeine. A single gustatory receptor neuron (GRN) in the medial styloconic sensillum of B. mori was highly sensitive to salicin and caffeine. The styloconic sensilla of H. armigera did not respond consistently to either of the bitter compounds. Phagostimulants also were tested. Myo-inositol and sucrose were detected specifically by two GRNs located in B. mori lateral styloconic sensillum, whereas, in H. armigera, sucrose was sensed by a GRN in the lateral sensillum, and myo-inositol by a GRN in the medial sensillum. Myo-inositol responsiveness in both species occurred at or below 10?3 mM, which is far below the naturally occurring concentration of 1 mM in plants. Larval responses to specific plant secondary compounds appear to have complex determinants that may include host range, metabolic capacity, and gustatory repertoire.  相似文献
