Leishmania parasites cycle between sand fly vectors and mammalian hosts, transforming from extracellular promastigotes that reside in the vectors’ alimentary canal to obligatory intracellular non-motile amastigotes that are harbored by macrophages of the mammalian hosts. The transition between vector and host exposes them to a broad range of environmental conditions that induces a developmental program of gene expression, with translation regulation playing a key role. The Leishmania genome encodes six paralogs of the cap-binding protein eIF4E. All six isoforms show a relatively low degree of conservation with eIF4Es of other eukaryotes, as well as among themselves. This variability could suggest that they have been assigned discrete roles that could contribute to their survival under the changing environmental conditions. Here, we describe LeishIF4E-5, a LeishIF4E paralog. Despite the low sequence conservation observed between LeishIF4E-5 and other LeishIF4Es, the three aromatic residues in its cap-binding pocket are conserved, in accordance with its cap-binding activity. However, the cap-binding activity of LeishIF4E-5 is restricted to the promastigote life form and not observed in amastigotes. The overexpression of LeishIF4E-5 shows a decline in cell proliferation and an overall reduction in global translation. Immuno-cytochemical analysis shows that LeishIF4E-5 is localized in the cytoplasm, with a non-uniform distribution. Mass spectrometry analysis of proteins that co-purify with LeishIF4E-5 highlighted proteins involved in RNA metabolism, along with two LeishIF4G paralogs, LeishIF4G-1 and LeishIF4G-2. These vary in their conserved eIF4E binding motif, possibly suggesting that they can form different complexes.  相似文献

Despite incremental improvements in the field of tissue engineering, no technology is currently available for producing completely autologous implants where both the cells and the scaffolding material are generated from the patient, and thus do not provoke an immune response that may lead to implant rejection. Here, a new approach is introduced to efficiently engineer any tissue type, which its differentiation cues are known, from one small tissue biopsy. Pieces of omental tissues are extracted from patients and, while the cells are reprogrammed to become induced pluripotent stem cells, the extracellular matrix is processed into an immunologically matching, thermoresponsive hydrogel. Efficient cell differentiation within a large 3D hydrogel is reported, and, as a proof of concept, the generation of functional cardiac, cortical, spinal cord, and adipogenic tissue implants is demonstrated. This versatile bioengineering approach may assist to regenerate any tissue and organ with a minimal risk for immune rejection.  相似文献

Soft, mechanically deformable materials and systems that can, on demand, manipulate light propagation within both the visible and infrared (IR) regions of the electromagnetic spectrum are desirable for applications that include sensing, optoelectronics, robotics, energy conservation, and thermal management. However, the development of such technologies remains exceptionally difficult, with relatively few examples reported to date. Herein, this challenge is addressed by engineering cephalopod-inspired adaptive camouflage platforms with multispectral functionality. First, stretchable copolymer membranes that feature outstanding unstrained protonic conductivities of up to ≈90 mS cm−1, demonstrate increases of ≈80% in their conductivities at strains of 200%, and exhibit no loss in electrical performance even under extreme elongations of 500% are described. Next, the membranes are used for the fabrication of mechanically and electrically actuated camouflage devices that function over an unprecedented spectral window; can simultaneously modulate their visible and IR specular-to-diffuse transmittance ratios by >3000-fold and >4-fold, respectively; feature rapid response times of ≈0.6 s; and exhibit good performance after repeated actuation. These findings may afford new scientific and technological opportunities not only for adaptive optics and photonics but also for any platform that can benefit from simultaneously controlling visible light and heat.  相似文献

Designing functional materials as electrode coatings to transduce high-quality information about redox molecules in biofluids is crucial for developing the next-generation medical devices. Rapidly analyzing the neurotransmitter dopamine (DA) urinary levels can enable point-of-care testing for neuroendocrine tumors. A novel sensing electrodes array modified with biopolymer chitosan and electrocatalytic carbon nanotube films that can generate cross-reactive electrochemical signals from complex biofluids, such as undiluted urine, is presented. By generating cross-reactive signals, the feasibility of quantifying DA levels from unprocessed urine samples is demonstrated. The films’ electrochemical activity is characterized and modeled the additive effect of the main redox interferants in urine (norepinephrine and uric acid) on the generated electrochemical signals that overlap and mask the electrochemical signature of DA. Finally, the feasibility of successfully quantifying urinary DA levels is demonstrated by investigating two calibration approaches: 1) using a synthetic solution (1.15 µm root mean squared error (RMSE) and 4.2 µm  limit-of-detection (LoD) values), and 2) directly using the urine samples (2.5 µm RMSE and 9.3 µm LoD values). The outcome of this work will enhance the understanding of the overlapping and masking electrochemical signatures and their interactions with functional materials, providing better analytical tools to differentiate redox molecules in highly complex biofluids.  相似文献

Atomic force microscopy (AFM) and scanning electron microscopy with energy dispersive spectroscopy (SEM-EDS) have been used for both morphological and elemental mass analysis study of atmospheric particles. As part of the geometrical particle analysis, and in addition to the traditional height profile measurement of individual particles, AFM was used to measure the volume relative to the projection area for each particle separately, providing a particle shape model. The element identification was done by the EDS analysis, and the element mass content was calculated based on laboratory calibration with particles of known composition. The SEM-EDS mass measurements from two samples collected at 150 and 500 m above the surface of the Mediterranean Sea were found to be similar to mass calculations derived from the AFM volume measurements. The AFM results show that the volume of most of the aerosols that were identified as soluble marine sulfate and nitrate aerosol particles can be better estimated using cylindrical shapes than spherical or conical geometry.  相似文献

A novel sample holder that enables atomic force microscopy (AFM) tips to be mounted inside a scanning electron microscopy (SEM) for the purpose of characterizing the AFM tips is described. The holder provides quick and easy handling of tips by using a spring clip to hold them in place. The holder can accommodate two tips simultaneously in two perpendicular orientations, allowing both top and side view imaging of the tips by the SEM.  相似文献

One of the differences between industrial manufacturing or processing plants and construction sites is the temporary nature of the construction site, which has traditionally precluded installation of sophisticated production monitoring systems. Monitoring of production progress, cost, and quality is performed almost exclusively manually, with the result that it is expensive and approximate, and is commonly delivered with a time lag that does not allow for an effectively closed control loop. Automated monitoring of construction lifting equipment to provide useful feedback information for project management is a strong potential candidate; almost all components and materials must be transported by machines, and monitoring of machines is relatively straightforward. A system concept, employing a “black box” monitor and an electronic building information model, was developed. A field study was conducted to test the feasibility of the concept. The results indicate that the system is technically feasible, and offers the potential to deliver real-time, accurate project control information at very low cost.  相似文献

The electronic structure of polycrystalline C60 thin films has been investigated using surface photovoltage spectroscopy (SPS) and conductivity measurements. the films show n-type semiconductivity with an activation energy of ∼ 0.8 eV as found from the temperature dependence of the conductivity at high temperatures. the electronic structure emerging from our SPS results comprises a 1.6 eV photo-conduction gap, a mobility gap of about 2.25 eV and two gap states, a donor and an acceptor, at 0.35 eV and (1.0-1.1) eV, respectively, below the photo-conduction edge. the results indicate the possibility of the existence of band tails, extending into the optical gap of these films, as well as other deep gap states.  相似文献
