Deficiencies in glycosylphosphatidyl-inositol (GPI)-linked proteins on erythrocytes and leucocytes in patients with paroxysmal nocturnal haemoglobinuria (PNH) are well known; however, expression on platelets in these patients is less well documented. We have studied CD55 and CD59 on the platelets of PNH and aplastic anaemia (AA) patients using flow cytometry. In all cases of PNH, CD55 and CD59 negative populations of platelets were detected with single or bimodal distribution and these results showed close correlation with the CD55 and CD59 patterns of neutrophils. Previous published studies have not demonstrated this distribution. We suggest that our findings may be due to the methodology used.  相似文献

An insulating YBa2Cu3O7-δ (YBCO) thin film was grown by inverted cylindrical magnetron (ICM) sputtering and under nitrogen cooling instead of oxygen in situ annealing. This insulating film went through different annealing processes in flowing oxygen, each time with increasing annealing temperature. The resistivity as a function of temperature was measured after each annealing. The fluctuation conductivity σ(T) is determined from the resistivity data and an analysis of the possible mechanisms for scattering of the fluctuating pairs is presented in this work. The crystal structure of the film through these different steps is also reported.  相似文献

We empirically examined S. H. Budman and A. S. Gurman's (see record 1984-04498-001) theoretical proposals concerning major differences in the value systems of long- vs. short-term therapists. Ss were 222 randomly selected licensed psychologists who indicated their preferred approach (short-term or long-term). Values were assessed with a scale designed for the study. Overall, results indicate that therapists who prefer a short-term approach are more likely to endorse the proposed values of the short-term therapist than are therapists who prefer a long-term approach. This finding held even after the authors controlled for the significant contributions of theoretical orientation and therapeutic practice variables. Specifically, short-term (vs. long-term) therapists believed more that psychological change could occur outside of therapy and that setting time limits would intensify the therapeutic work. Lipopolysaccharide (LPS) administration to mice elicited the activation of nuclear factor kappaB (NF-kappaB) in several tissues including liver and macrophages. Maximal activation was observed 1 h after treatment but declined at 3 and 6 h. The levels of IkappaBalpha and IkappaBbeta were analyzed during this period in an attempt to correlate NF-kappaB activity with IkappaB resynthesis. Degradation of IkappaBalpha was very rapid and was followed by recovery 1 h after LPS administration. IkappaBbeta degradation, which has been associated with persistent NF-kappaB activation, was complete at 1 h. However, a rapid recovery of IkappaBbeta in these tissues was observed at 3 h in parallel with the abrogation of NF-kappaB activity. Immunolocalization of newly synthesized IkappaBbeta by confocal microscopy revealed its preferential accumulation in the cytosol. Analysis of IkappaBbeta by Western blot using high resolution polyacrylamide gel electrophoresis showed the presence of two bands in cytosolic extracts of LPS-treated macrophages at 3 h, but only one band with the same mobility as the control was detected at 6 h. Moreover, treatment of extracts of resynthesized IkappaBbeta with alkaline phosphatase resulted in the accumulation of the protein of slightly higher electrophoretic mobility, indicating the prevalence of a rapid phosphorylation of the newly synthesized IkappaBbeta. At the mRNA level, up-regulation of IkappaBbeta was observed in macrophages stimulated for 1 h with LPS. When the effect of pro-inflammatory cytokines was investigated, tumor necrosis factor alpha, but not interleukin-1 or interferon-gamma, promoted an important degradation of IkappaBbeta followed by an increase in the mRNA at 1 h. These results suggest the existence of LPS- and tumor necrosis factor alpha- specific pathways involved in a rapid IkappaBbeta degradation and resynthesis and might explain the transient period of activation of NF-kappaB in these tissues upon stimulation with these factors. This rapid control of NF-kappaB function may contribute to the attenuation of the inflammatory response of these cells.  相似文献

Since batch chemical reactors exhibit an integrating response, temperature control for these systems can be a real problem for conventional PID controllers. Tuning can be extremely difficult due to the reduced stability margins proved for this type of processes. In this work, a simple robust control strategy for temperature regulation in batch and semi-batch chemical reactors is proposed. The feedback controller is composed by an approximate I/O linearizing feedback equipped with a calorimetric balance estimator. Based on standard results from singular perturbations, it is proven that the proposed feedback controller (i) can track a bounded temperature trajectory as close as desired (i.e., practical stability) by adjusting a single estimation parameter, and (ii) after a short transient, the performance of the exact I/O linearizing feedback can be recovered as the calorimetric balance estimation rate is increased.  相似文献

A solar heated experimental drying kiln was installed in Madrid (INIA-Forest Industries Department) to study the efficiency of solar drying of sawn lumber in Spain, as compared with normal air drying. The kiln has been designed as a low cost semi-greenhouse type, but was equipped with a semi-automated control of the drying schedules. The performance of the solar kiln has been studied during ten test runs over a period of two years particularly with respect to drying rate, initial and final wood MC, reliability and efficiency. The results have proven that the solar kiln is capable of drying (economically) from an initial moisture content of 70 to 80%, to a final MC of 8 to 10% up to 3.5 times faster than air drying. With respect to checks, splits and internal stresses, the final wood quality is mostly better, not only compared to normal air drying but also to conventional kiln drying.  相似文献

The nonlinearities generated by wide-band, high-current photodetectors are discussed in the context of the performance of an externally modulated analog radio-frequency fiber-optic link. The quantitative impart of the detector's third-order intermodulation product intercept (IP3) on the link's spur-free dynamic range is analyzed. Waveguide p-i-n photodiodes were developed to specifically address these stringent IP3 goals. The IP3 was measured using a four laser heterodyne system resulting in an IP3 of 27 dBm measured at a center frequency of 20 GHz with 20 mA of de photocurrent. Recent legislation in several states has called for the removal of methyl tert-butyl ether (MTBE) from gasoline. In order to comply with Federal Clean Air Act requirements for carbon monoxide and ozone attainment, ethanol is being considered as a replacement for MTBE. The objective of this study is to evaluate the potential impact of ethanol on benzene plume lengths in subsurface environments following accidental spills of ethanol-blended gasoline. Two types of studies were conducted here. First, laboratory studies were performed using a pure culture indigenous to a gasoline-contaminated aquifer to evaluate the effect of ethanol on the rate of benzene biodegradation under aerobic conditions. Results from microbial studies showed that the biodegradation of 25 mg/L benzene was severely inhibited in the presence of 25 mg/L ethanol. While the enzymes responsible for benzene biodegradation by the culture were inducible, ethanol degradation appeared to be constitutive. Second, a two-dimensional model was developed to quantify the impact of ethanol on benzene plume lengths using weighted-average aerobic and anaerobic biodegradation rates for benzene in the presence and absence of ethanol. Model simulations indicated that benzene plume lengths are likely to increase by 16–34% in the presence of ethanol.  相似文献

Cryoprotectants such as hydrocolloids (amidated low-methoxyl (ALM) and high methoxyl (HM) pectins), kappa- and iota-carrageenans (k-C and i-C), xanthan gum (XG)) and dairy proteins [whey protein (WP), sodium caseinate (SC)] were added to mashed potatoes to investigate ways of improving the effects of freezing and thawing. It was found that each hydrocolloid and protein, depending on concentration, affected the mechanical properties [instrumental textural profile analysis (ITPA), cone penetration (CP) test], the total colour difference (ΔE*) with respect to fresh control (FC) and the sensory attributes of fresh (F) and frozen/thawed (F/T) mashed potatoes in a different way. In the F/T samples, adding 5 and 8 g kg−1 ALM, 3, 5 and 8 g kg−1 k-C, 1.5, 3, 5 and 8 g kg−1 i-C and 1.5, 5 and 15 g kg−1 WP significantly increased ITPA consistency. Also, adding 2.5 and 5 g kg−1 XG significantly increased ITPA consistency of the F/T product. In both F and F/T samples, k-C provided the highest ITPA consistency and also high CP average force evidencing a stronger synergistic effect in κ-C/denatured milk protein systems, although the excessive thickening and stickiness provided was judged undesirable by the panellists. Adding 8 g kg−1 HM pectin had a disruptive effect on the mashed potatoes and decreased both ITPA consistency and CP average force. In all cases, freezing and thawing reinforced the gel structure of the products as compared to F and FC counterparts. The ΔE* values were higher in F samples containing ALM and HM pectins. Dairy proteins affected the taste and odour of the mashed potatoes and were judged unacceptable in the sensory analysis. Samples containing 0.5 and 1.5 g kg−1 added XG were preferred organoleptically due to the creamy mouthfeel it produced. ITPA consistency correlated well with sensory texture attributes.  相似文献
