Vitamin A is an essential fat-soluble vitamin that occurs in various chemical forms. It is essential for several physiological processes. Either hyper- or hypovitaminosis can be harmful. One of the most important vitamin A functions is its involvement in visual phototransduction, where it serves as the crucial part of photopigment, the first molecule in the process of transforming photons of light into electrical signals. In this process, large quantities of vitamin A in the form of 11-cis-retinal are being isomerized to all-trans-retinal and then quickly recycled back to 11-cis-retinal. Complex machinery of transporters and enzymes is involved in this process (i.e., the visual cycle). Any fault in the machinery may not only reduce the efficiency of visual detection but also cause the accumulation of toxic chemicals in the retina. This review provides a comprehensive overview of diseases that are directly or indirectly connected with vitamin A pathways in the retina. It includes the pathophysiological background and clinical presentation of each disease and summarizes the already existing therapeutic and prospective interventions.  相似文献

The factors determining corrosion are reviewed in this paper, with an emphasis on iron tolerance limit and the production of high-purity castings. To understand the iron impurity tolerance limit, magnesium phase diagrams were calculated using the Pandat software package. Calculated phase diagrams can explain the iron tolerance limit and the production of high-purity castings by means of control of melt conditions; this is significant for the production of quality castings from recycled magnesium. Based on the new insight, the influence of the microstructure on corrosion of magnesium alloys is reviewed.  相似文献

The influence of Cl and SO42− on the electrochemical behavior of AZ91D, AZCe2, and AZLa1 was studied. For all alloys, there was a current plateau in the anodic polarization curves in Na2SO4 solutions. In 0.5% NaCl solution, there was a small current plateau, whereas there was none in the 3.5% and 5% NaCl solutions. This indicated that SO42− is less aggressive than Cl. The range of the current plateau decreased with increasing SO42− concentration. For all alloys, the high frequency capacitive loop in the Nyquist plots decreased with increasing concentration consistent with the decrease in corrosion resistance with increasing Cl and SO42− concentration.  相似文献

Metabolically active gasotransmitters (nitric oxide, carbon monoxide and hydrogen sulfide) are important signalling molecules that show therapeutic utility in oxidative pathologies. The reduced form of selenium, hydrogen selenide (HSe/H2Se), shares some characteristics with these molecules. The simple selenide salt, sodium hydroselenide (NaHSe) showed significant metabolic activity, dose-dependently decreasing ex vivo O2 consumption (rat soleus muscle, liver) and transiently inhibiting mitochondrial cytochrome C oxidase (liver, heart). Pharmacological manipulation of selenoprotein expression in HepG2 human hepatocytes revealed that the oxidation status of selenium impacts on protein expression; reduced selenide (NaHSe) increased, whereas (oxidized) sodium selenite decreased the abundance of two ubiquitous selenoproteins. An inhibitor of endogenous sulfide production (DL-propargylglycine; PAG) also reduced selenoprotein expression; this was reversed by exogenous NaHSe, but not sodium hydrosulfide (NaHS). NaHSe also conferred cytoprotection against an oxidative challenge (H2O2), and this was associated with an increase in mitochondrial membrane potential. Anesthetized Wistar rats receiving intravenous NaHSe exhibited significant bradycardia, metabolic acidosis and hyperlactataemia. In summary, NaHSe modulates metabolism by inhibition of cytochrome C oxidase. Modification of selenoprotein expression revealed the importance of oxidation status of selenium therapies, with implications for current clinical practice. The utility of NaHSe as a research tool and putative therapeutic is discussed.  相似文献

An annular laser beam based powder cladding head, which enables an axial powder feeding and variation of the laser beam intensity distribution (LBID) on the workpiece surface is presented. The influence of typical LBIDs, including Ring, Tophat(?), Tophat(+), and Gaussian-like, on a cladding process has been characterized based on the process and melt pool visualization, powder catchment efficiency, clad layer geometry, and porosity. The results showed that the most stable process without plasma formation but with low dilution and porosity of the clad layer can be achieved within the range from a Ring to a Tophat(?) LBID. Additionally, axial powder feeding results in a high powder catchment efficiency above 80%.  相似文献

Fungal species of genus Sepedonium are rich sources of diverse secondary metabolites (e.g., alkaloids, peptaibols), which exhibit variable biological activities. Herein, two new peptaibols, named ampullosporin F (1) and ampullosporin G (2), together with five known compounds, ampullosporin A (3), peptaibolin (4), chrysosporide (5), c(Trp-Ser) (6) and c(Trp-Ala) (7), have been isolated from the culture of Sepedonium ampullosporum Damon strain KSH534. The structures of 1 and 2 were elucidated based on ESI-HRMSn experiments and intense 1D and 2D NMR analyses. The sequence of ampullosporin F (1) was determined to be Ac-Trp1-Ala2-Aib3-Aib4-Leu5-Aib6-Gln7-Aib8-Aib9-Aib10-GluOMe11-Leu12-Aib13-Gln14-Leuol15, while ampullosporin G (2) differs from 1 by exchanging the position of Gln7 with GluOMe11. Furthermore, the total synthesis of 1 and 2 was carried out on solid-phase to confirm the absolute configuration of all chiral amino acids as L. In addition, ampullosporin F (1) and G (2) showed significant antifungal activity against B. cinerea and P. infestans, but were inactive against S. tritici. Cell viability assays using human prostate (PC-3) and colorectal (HT-29) cancer cells confirmed potent anticancer activities of 1 and 2. Furthermore, a molecular docking study was performed in silico as an attempt to explain the structure-activity correlation of the characteristic ampullosporins (1–3).  相似文献

We aimed to assess the in vitro antimicrobial activity and the in vivo effect on the murine fecal microbiome and volatile organic compound (VOC) profile of (S)-reutericyclin. The antimicrobial activity of (S)-reutericyclin was tested against Clostridium difficile, Listeria monocytogenes, Escherichia coli, Enterococcus faecium, Staphylococcus aureus, Staphylococcus (S.) epidermidis, Streptococcus agalactiae, Pseudomonas aeruginosa and Propionibacterium acnes. Reutericyclin or water were gavage fed to male BALBc mice for 7 weeks. Thereafter stool samples underwent 16S based microbiome analysis and VOC analysis by gas chromatography mass spectrometry (GC-MS). (S)-reutericyclin inhibited growth of S. epidermidis only. Oral (S)-reutericyclin treatment caused a trend towards reduced alpha diversity. Beta diversity was significantly influenced by reutericyclin. Linear discriminant analysis Effect Size (LEfSe) analysis showed an increase of Streptococcus and Muribaculum as well as a decrease of butyrate producing Ruminoclostridium, Roseburia and Eubacterium in the reutericyclin group. VOC analysis revealed significant increases of pentane and heptane and decreases of 2,3-butanedione and 2-heptanone in reutericyclin animals. The antimicrobial activity of (S)-reutericyclin differs from reports of (R)-reutericyclin with inhibitory effects on a multitude of Gram-positive bacteria reported in the literature. In vivo (S)-reutericyclin treatment led to a microbiome shift towards dysbiosis and distinct alterations of the fecal VOC profile.  相似文献

Inclusion complexes of o-, m- and p-coumaric acid (CA) with β-cyclodextrin (βCD) were prepared in stoichiometric ratios (1:1) and stability constants and antioxidant activity of the complexes were studied. The apparent stability constants in aqueous solution of 0.39 × 103 M−1, 2.81 × 103 M−1 and 49 × 103 M−1 for o-, m- and p-CA complexes, respectively, were determined by phase solubility tests. Different analytical techniques (IR, MS) in combination with different solvent washing procedures, were used for confirmation of the nature of the inclusion complexes. Dioxan was a suitable solvent for removal of only free CA and CA adsorbed on the surface of the βCD, while methanol removed absorbed, included and free CA from the complexes. In the case of o- and m-CA–βCD complexes, antioxidant activity was significantly increased while, in p-CA–βCD, it remained unchanged. The impact of complex structure on antioxidant activity of CA isomers was clarified.  相似文献

Intermittent and ephemeral streams in dryland environments support diverse assemblages of aquatic and terrestrial life. Understanding when and where water flows provide insights into the availability of water, its response to external controlling factors, and potential sensitivity to climate change and a host of human activities. Knowledge regarding the timing of drying/wetting cycles can also be useful to map critical habitats for species and ecosystems that rely on these temporary water sources. However, identifying the locations and monitoring the timing of streamflow and channel sediment moisture remains a challenging endeavor. In this paper, we analyzed daily conductivity from 37 sensors distributed along 10 streams across an arid mountain front in Arizona (United States) to assess spatiotemporal patterns in flow permanence, defined as the timing and extent of water in streams. Conductivity sensors provide information on surface flow and sediment moisture, supporting a stream classification based on seasonal flow dynamics. Our results provide insight into flow responses to seasonal rainfall, highlighting stream reaches very reactive to rainfall versus those demonstrating more stable streamflow. The strength of stream responses to precipitation are explored in the context of surficial geology. In summary, conductivity data can be used to map potential stream habitat for water-dependent species in both space and time, while also providing the basis upon which sensitivity to ongoing climate change can be evaluated.  相似文献
