Identical distribution of C2- and Cx-cellulase activities of enzyme complexes produced by Trichoderma lignorum on a medium with lactose, a soluble "inductor", and on a medium with cellulose was found by means of disc elestrophoresis in polyacrylamide gel. The maximum rate of synthesis of cellulases on the medium with lactose was registered during the highest deceleration, and even complete cessation, of the fungal growth. During this phase, only one electrophoretically homogeneous cellulase component with Rf of 0.44 possessing all types of the cellulase activity is present in the cultural broth. In the course of growth of the fungus on cellulose after 48 hours, also only one electrophoretically homogeneous component with Rf of 0.44 was found in the cultural broth when the rate of the substrate degradation was highest. The appearance of minor protein components with the activity of cellulase at later stages of cultivation after cessation of the fungal growth is supposed to be caused by modification of the main cellulase component with Rf of 0.44 by the growth medium.  相似文献

γ-Alumina extrudates for chemical-looping combustion in fluidized bed reactors were shaped by varying acetic acid concentrations between 0.07 and 3.76 M. Influence of pseudo-boehmite peptization on structural properties, microstructure, chemical phases and attrition resistance was determined. With addition of acetic acid, the d90 of boehmite agglomerates after 1 h kneading decreased from 134 to 40 μm at pH 4. Due to this, the extrusion diameter was reduced from 1500 to 200 μm, as well as median pore radii (from 30.1 to 5.3 nm). Porosity was about 70%. Addition of more than 1.87 M acid lead to a slight increase in mesopore sizes caused by some pore blocking caused by the formation of aluminium acetate salts. A small micropore surface was determined with t-layer model from Harkins and Jura. Higher attrition resistance was observed for samples peptized with lower acid concentration because of the closer contact between particles after decomposition.  相似文献

A method is developed to generate a top of the atmosphere clear reflectance from the Global Vegetation Index (GVI) product. Our goal is to use this dataset as a threshold to be applied to the forthcoming POLDER observations, for operational cloud detection. The method is based on the hypothesis that clouds add a high frequency signal to the slow variations of the surface reflectance in clear conditions. The validity of our algorithm is verified through an analysis of the temporal profile of the reflectance that it generates. We show that these profiles are better than those resulting from the simpler Maximum Value Composite (MVC) method. The method is applied to five years of GVI products and the results are used to derive a reference database which accounts for the interannual variability of the surface reflectance.  相似文献

The aim of this study was to discuss the influence of different filter surface chemistries on the properties of foam filters. For reliable results, it is essential to ensure comparable structural properties (cell size and strut thickness) for all different surface chemistries (Al2O3, MgAl2O4, 3Al2O3·2SiO2, SiO2, and TiO2) possess the same structural properties (cell size and strut thickness). Filters made of 100% of the investigated materials and alumina skeletons coated with the investigated materials were prepared. The coated alumina samples were sintered in one and two steps. The processing route with two sintering steps resulted in improved mechanical properties and comparable shrinkage and strut thickness. The 100% bulk foams possessed different pore sizes due to the differences of the material shrinkage. In this study, a comparison of the experimental investigated properties of the ceramic foam filters and the theoretically calculated values for foam materials derived from the bulk material properties is established.  相似文献

The potential for broadband and data services is large and continues to expand for the foreseeable future. Consideration of the cost-effective delivery and support of both new, advanced services, as well as existing services, has led to the driver for multiservice platforms (MSPs). The cost effectiveness results from the MSP potential to share network bandwidth between services, offer a potential simplification of the operational support systems (OSS), and enable new service features and functions. A generic ATM-based MSP is described in this paper where issues of service support, service interworking, operations and maintenance, and network architecture are discussed. Master Logic Diagram (MLD), a method for identifying events initiating accidents in chemical installations, is presented. MLD is a logic diagram that resembles a fault tree but without the formal mathematical properties of the latter. MLD starts with a Top Event "Loss of Containment" and decomposes it into simpler contributing events. A generic MLD has been developed which may be applied to all chemical installations storing toxic and/or flammable substances. A methodology for estimating the risk owing to the phenomenon of boiling liquid expanding vapour explosion (BLEVE) in the presence of uncertainties both in the model and in the parameters of the models is presented. BLEVE takes place when a tank containing liquefied petroleum gas (LPG) is exposed to fire and fails catastrophically. Two models have been used in the estimation of the intensity of thermal radiation from the resulting fireball, namely the solid-flame model assuming an emission power independent of the combustion mass and the point-source model that estimates the emissive power as a function of the combustion mass. Three measures of the BLEVE consequences, the intensity of thermal radiation, the dose of thermal radiation and the probability of loss of life as a result of the exposure to the thermal radiation and as a function of the distance from the center of the tank have been considered. Uncertainties in the exact values of the parameters of the models have been quantified and the resulting uncertainties in the three consequence measures have been assessed. A sensitivity analysis on the relative contribution of the uncertainty in each of the input variables to the uncertainties of the consequence measures has been performed. One conclusion is that the uncertainties in the probability of loss of life are mainly due to the uncertainties in the model of the physical phenomenon rather than to the uncertainties of the dose-response model.  相似文献

Galphao, the most abundant G protein in mammalian brain, occurs at least in two subforms, i.e., Galphao1 and Galphao2, derived by alternative splicing of the mRNA. A third Galphao1-related isoform, Galphao3, has been purified, representing about 30% of total Go in brain. Initial studies revealed distinct biochemical properties of Galphao3 as compared with other Galphao isoforms. In matrix-assisted laser desorption/ionization peptide mass mapping of gel-isolated Galphao1 and Galphao3, C-terminal peptides showed a difference of +1 Da for Galphao3. Nanoelectrospray tandem mass spectrometry sequencing revealed an Asp instead of an Asn at position 346 of Galphao3. Gel electrophoretic analysis of recombinant Galphao3 showed the same mobility as native Galphao3 but distinct to Galphao1. The conversion of 346Asn-->Asp changed the signaling properties, including the velocity of the basal guanine nucleotide-exchange reaction, which points to the involvement of the C terminus in basal guanosine 5'-[gamma-thio]triphosphate binding. No cDNA coding for Galphao3 was detected, suggesting an enzymatic deamidation of Galphao1 by a yet-unidentified activity. Therefore, Galpha heterogeneity is generated not only at the DNA or RNA levels, but also at the protein level. The relative amount of Galphao1 and Galphao3 differed from cell type to cell type, indicating an additional principle of G protein regulation.  相似文献
