The focus of this research was to investigate the efficacy of icing as a postharvest treatment for reduction of the levels of Vibrio vulnificus and Vibrio parahaemolyticus in commercial quantities of shellstock oysters. The experiments were conducted in June and August of 2006 and consisted of the following treatments: (i) on-board icing immediately after harvest; (ii) dockside icing approximately 1 to 2 h prior to shipment; and (iii) no icing (control). Changes in the levels of pathogenic Vibrio spp. during wholesale and retail handling for 2 weeks postharvest were also monitored. On-board icing achieved temperature reductions in all sacks in accordance with the National Shellfish Sanitation Program standard, but dockside icing did not meet this standard. Based on one-way analysis of variance, the only statistically significant relationship between Vibrio levels and treatment occurred for samples harvested in August; in this case, the levels of V. vulnificus in the noniced oysters were significantly higher (P < 0.05) than were the levels in the samples iced on-board. When analyzing counts over the 14-day storage period, using factorial analysis, there were statistically significant differences in V. vulnificus and V. parahaemolyticus levels by sample date and/or treatment (P < 0.05), but these relationships were not consistent. Treated (iced) oysters had significantly higher gaping (approximately 20%) after 1 week in cold storage than did noniced oysters (approximately 10%) and gaping increased significantly by day 14 of commercial storage. On-board and dockside icing did not predictably reduce the levels of V. vulnificus or V. parahaemolyticus in oysters, and icing negatively impacted oyster survival during subsequent cold storage.  相似文献

ABSTRACT Pretreated carrot discs were thermally processed (90 °C to 110 °C) in closed containers and the resulting textural characteristics were analyzed. The pretreatment conditions used include conventional high‐temperature blanching (90 °C, 4 min), low‐temperature blanching (LTB = 60 °C, 40 min), LTB combined with 0.5% calcium chloride soaking, LTB combined with 2% sodium chloride soaking, high pressure pretreatment (HP = 400 MPa, 60 °C, 15 min), HP combined with 0.5% calcium chloride soaking, and control (non‐pretreated sample). Alcohol insoluble residues (AIR) from the pretreated carrot discs were characterized in terms of degree of methoxylation (DM). The AIR samples were further subjected to fractionation into water‐soluble pectin (WSP), chelator‐soluble pectin (CSP), and sodium carbonate‐soluble pectin (NSP). Heat depolymerization patterns and β‐elimination kinetics were investigated on the different pectin fractions. Thermal texture degradation was strongly influenced by the pretreatment condition used and the processing temperature during subsequent thermal treatment. Pretreatment conditions that showed a significant reduction in DM exhibited decreased WSP content, reduced β‐elimination, and consequently superior textural characteristics. β‐elimination was markedly pronounced in the highly methoxylated WSP fractions. CSP and NSP fractions were insensitive to β‐elimination. A strong correlation (r> 0.95) between thermal texture loss of carrots and β‐elimination kinetics exists. Overall, the benefits of controlled pectinmethylesterase activity in carrot processing were pointed out.  相似文献

Strawberry samples of two varieties (Camarosa and Elsanta) were dehydrated using different osmotic solutions (60% glucose, fructose, sucrose and raftilose) and subsequently frozen by rapid and high-pressure shift freezing (HPSF). The effect of pectinmethylesterase (PME) and calcium (Ca++) added to the osmotic solutions on several compositional parameters and the textural/structural quality of dehydrated and osmodehydrofrozen-then-thawed samples was studied. Due to the presence of PME and Ca++ in the osmotic solutions, weight reduction upon dehydration was slightly decreased, which was correlated to a small positive effect on the net uptake of sugars and depression of the initial freezing point. Except for the Camarosa samples treated with sucrose, PME and Ca++ in osmotic sugar solutions positively affected the relative hardness of dehydrated fruits, which was ascribed to the effect of PME and Ca++ on the cell wall strength of the tissue. No cell wall damage and tissue particle alterations were observed upon dehydration. The effect of osmotic dehydration (OD) using different sugar solutions without PME and Ca++ on the texture and structure of frozen-then-thawed samples was limited and sometimes negative. The added PME and Ca++ however positively influenced the volume and shape of the thawed samples, which could be related to slightly higher relative hardness values and, for the Elsanta strawberry fruits, also to the reduced (up to 81%) drip loss upon thawing. Upon freezing the dehydrated fruits, no cell wall disruption was observed. Tissue distortion caused by freezing and indicated by a decrease in particle size, convexity and roundness, was compensated by the use of PME and Ca++ during OD.  相似文献

This study was conducted to determine plasmid colocalization and transferability of both erm(B) and tet(M) genes in Enterococcus faecalis isolates from abattoir pigs in Canada. A total of 124 E. faecalis isolates from cecal contents of abattoir pigs were examined for antibiotic susceptibility. High percentages of resistance to macrolides and tetracyclines were found. Two predominant multiresistance patterns of E. faecalis were examined by PCR and sequencing for the presence of genes encoding antibiotic resistance. Various combinations of antibiotic resistance genes were detected; erm(B) and tet(M) were the most common genes. Plasmid profiling and hybridization revealed that both genes were colocated on a ~9-kb transferable plasmid in six strains with the two predominant multiresistant patterns. Plasmid colocalization and cotransfer of tet(M) and erm(B) genes in porcine E. faecalis isolates indicates that antibiotic coselection and transferability could occur via this single genetic element. To our knowledge, this is the first report on plasmid colocalization and transferability of erm(B) and tet(M) genes in E. faecalis on a mobile genetic element of ~9 kb. Physical linkage between important antibiotic resistance determinants in enterococci is of interest for predicting potential transfer to other bacterial genera.  相似文献

Background

Engineered carbon nanotubes are currently used in many consumer and industrial products such as paints, sunscreens, cosmetics, toiletries, electronic processes and industrial lubricants. Carbon nanotubes are among the more widely used nanoparticles and come in two major commercial forms, single-walled carbon nanotubes (SWCNT) and the more rigid, multi-walled carbon nanotubes (MWCNT). The low density and small size of these particles makes respiratory exposures likely. Many of the potential health hazards have not been investigated, including their potential for carcinogenicity. We, therefore, utilized a two stage initiation/promotion protocol to determine whether inhaled MWCNT act as a complete carcinogen and/or promote the growth of cells with existing DNA damage. Six week old, male, B6C3F1 mice received a single intraperitoneal (ip) injection of either the initiator methylcholanthrene(MCA, 10 μg/g BW, i.p.), or vehicle (corn oil). One week after i.p. injections, mice were exposed by inhalation to MWCNT (5 mg/m3, 5 hours/day, 5 days/week) or filtered air (controls) for a total of 15 days. At 17 months post-exposure, mice were euthanized and examined for lung tumor formation.

Results

Twenty-three percent of the filtered air controls, 26.5% of the MWCNT-exposed, and 51.9% of the MCA-exposed mice, had lung bronchiolo-alveolar adenomas and lung adenocarcinomas. The average number of tumors per mouse was 0.25, 0.81 and 0.38 respectively. By contrast, 90.5% of the mice which received MCA followed by MWCNT had bronchiolo-alveolar adenomas and adenocarcinomas with an average of 2.9 tumors per mouse 17months after exposure. Indeed, 62% of the mice exposed to MCA followed by MWCNT had bronchiolo-alveolar adenocarcinomas compared to 13% of the mice that received filtered air, 22% of the MCA-exposed, or 14% of the MWCNT-exposed. Mice with early morbidity resulting in euthanasia had the highest rate of metastatic disease. Three mice exposed to both MCA and MWCNT that were euthanized early had lung adenocarcinoma with evidence of metastasis (5.5%). Five mice (9%) exposed to MCA and MWCNT and 1 (1.6%) exposed to MCA developed serosal tumors morphologically consistent with sarcomatous mesotheliomas, whereas mice administered MWCNT or air alone did not develop similar neoplasms.

Conclusions

These data demonstrate that some MWCNT exposures promote the growth and neoplastic progression of initiated lung cells in B6C3F1 mice. In this study, the mouse MWCNT lung burden of 31.2 μg/mouse approximates feasible human occupational exposures. Therefore, the results of this study indicate that caution should be used to limit human exposures to MWCNT.  相似文献

Modelling activated sludge systems has gained an increasing momentum after the introduction of activated sludge models (ASMs) in 1987. Application of dynamic models for full-scale systems requires essentially a calibration of the chosen ASM to the case under study. Numerous full-scale model applications have been performed so far which were mostly based on ad hoc approaches and expert knowledge. Further, each modelling study has followed a different calibration approach: e.g. different influent wastewater characterization methods, different kinetic parameter estimation methods, different selection of parameters to be calibrated, different priorities within the calibration steps, etc. In short, there was no standard approach in performing the calibration study, which makes it difficult, if not impossible, to (1) compare different calibrations of ASMs with each other and (2) perform internal quality checks for each calibration study. To address these concerns, systematic calibration protocols have recently been proposed to bring guidance to the modeling of activated sludge systems and in particular to the calibration of full-scale models. In this contribution four existing calibration approaches (BIOMATH, HSG, STOWA and WERF) will be critically discussed using a SWOT (Strengths, Weaknesses, Opportunities, Threats) analysis. It will also be assessed in what way these approaches can be further developed in view of further improving the quality of ASM calibration. In this respect, the potential of automating some steps of the calibration procedure by use of mathematical algorithms is highlighted.  相似文献

The fatty acid specificity of four lipases towards eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) was evaluated when performing ethanolysis of squid oil. During the first part of ethanolysis, no DHA ethyl esters were detected when using the lipases from Thermomyces lanuginosus, Pseudomonas cepacia or Pseudomonas fluorescens (in the case of the second and third lipases, no EPA ethyl esters were detected either). This indicates that these three lipases could not catalyse the conversion of DHA located in a triacylglycerol to ethyl ester, and that the Pseudomonas lipases could not catalyse the conversion of EPA either. This pattern was not found for the lipase from Rhizomucor miehei. The lipase from Thermomyces lanuginosus showed the lowest specificity towards DHA and the highest DHA recovery during DHA enrichment in the acylglycerol fraction. It was thus used to catalyse the ethanolysis of squid oil on a larger scale. The ethyl esters formed were removed using short‐path distillation, resulting in a product containing mainly mono‐ and diacylglycerols. The product contained 34 mol‐% DHA and 17 mol‐% EPA, compared with 19 mol‐% DHA and 12 mol‐% EPA in the original squid oil.  相似文献

The understanding of how genetic information may be inherited through generations was established by Gregor Mendel in the 1860s when he developed the fundamental principles of inheritance. The science of genetics, however, began to flourish only during the mid-1940s when DNA was identified as the carrier of genetic information. The world has since then witnessed rapid development of genetic technologies, with the latest being genome-editing tools, which have revolutionized fields from medicine to agriculture. Cannabinoids exert anti-cancer actions; however, the underlying cytotoxic mechanisms and the cannabinoid receptors (CBRs) involved remain unclear. In this study, CBRs were characterized in several cancer cell lines. Radioligand binding screens surprisingly revealed specific binding only for the non-selective cannabinoid [3H]WIN-55,212-2, and not [3H]CP-55,940, indicating that the expressed CBRs exhibit atypical binding properties. Furthermore, [3H]WIN-55,212-2 bound to a single site in all cancer cells with high affinity and varying densities. CBR characteristics were next compared between human prostate cancer cell lines expressing low (PC-3) and high (DU-145) CBR density. Although mRNA for canonical CBRs was detected in both cell lines, only 5 out of 15 compounds with known high affinity for canonical CBRs displaced [3H]WIN-55,212-2 binding. Functional assays further established that CBRs in prostate cancer cells exhibit distinct signaling properties relative to canonical Gi/Go-coupled CBRs. Prostate cancer cells chronically exposed to both CBR agonists and antagonists/inverse agonists produced receptor downregulation, inconsistent with actions at canonical CBRs. Treatment of DU-145 cells with CBR ligands increased LDH-release, decreased ATP-dependent cell viability, and produced mitochondrial membrane potential depolarization. In summary, several cancer cell lines express CBRs with binding and signaling profiles dissimilar to canonical CBRs. Drugs selectively targeting these atypical CBRs might exhibit improved anti-cancer properties.  相似文献

An extensive literature opposes the provision of property titles for the residents of informal settlements. One concern is that titling leads to commodification and the market-driven displacement of the original inhabitants. Another is that it propagates the ideology of private ownership, undermines collective solidarity and demobilises social movements. This article, based on observations from Mexico City and Guadalajara, finds little evidence of displacement but highlights the importance of location. It supports the view that formalisation undermines resistance, but argues that titling does so by meeting rather than creating the desire of the urban poor for private property and homeownership.  相似文献
