The molecular mechanism of entry of herpes viruses requires a multicomponent fusion system. Virus entry and cell-cell fusion of Herpes simplex virus (HSV) requires four glycoproteins: gD, gB and gH/gL. The role of gB remained elusive until recently, when the crystal structure of HSV-1 gB became available. Glycoprotein B homologues represent the most highly conserved group of herpes virus glycoproteins; however, despite the high degree of sequence and structural conservation, differences in post-translational processing are observed for different members of this virus family. Whereas gB of HSV is not proteolytically processed after oligomerization, most other gB homologues are cleaved by a cellular protease into subunits that remain linked through disulfide bonds. Proteolytic cleavage is common for activation of many other viral fusion proteins, so it remains difficult to envisage a common role for different herpes virus gB structures in the fusion mechanism. We selected bovine herpes virus type 1 (BoHV-1) and herpes simplex virus type 1 (HSV-1) as representative viruses expressing cleaved and uncleaved gBs, and have screened their amino acid sequences for regions of highly interfacial hydrophobicity. Synthetic peptides corresponding to such regions were tested for their ability to induce the fusion of large unilamellar vesicles and to inhibit herpes virus infection. These results underline that several regions of the gB protein are involved in the mechanism of membrane interaction.  相似文献

Air samples were collected simultaneously at three sites downwind of Lake Ontario and at a control site near Lake Erie from March to July of 1999. The Lake Erie site (Stockton, NY) had PCB concentrations similar to rural Integrated Atmospheric Deposition Network (IADN) sampling sites across the Great Lakes, exhibited limited seasonal variation, and approximates regional background. Samples taken along Lake Ontario's southeastern shore (Rice Creek and Sterling, NY) had elevated PCB concentrations averaging approximately 1 ng/m3 and were more chlorinated than air collected at IADN sites and at Stockton. Air samples from Potsdam (approximately 75 km inland) had similar concentrations but were less chlorinated. Clausius-Clapeyron plots revealed a strong correlation between PCB fugacity and temperature near Lake Ontario; however, the extent of chlorination of the air samples rules out volatilization from the lake as a major source. It is hypothesized that volatilization from local surfaces, enriched in higher chlorinated congeners by meteorological or geographic factors, drives both the concentration and composition of airborne PCBs along Lake Ontario's southeastern shore.  相似文献

This study falls in the areas of both differential psychology and twin psychology. Using the EFT and the WCST (computerized version), we examined 11 MZFF pairs between 18 and 35 years of age. The aim was to establish the genetic and/or environmental determination of global-analytical cognitive style as well as some characteristics of conceptualization linked to field dependence. The research strategy consisted of introducing three other groups of the same size to control the weight of environmental factors different from those determined by subject selection. The results seem to support the hypothesis of genetic determination of field dependence of the MZFFs, probably linked to the XX chromosome combination. The "couple effect" and the attitude of parents and others toward two identical female subjects may contribute to full expression of the genome. The characteristics of conceptualization revealed by the WCST show that MZFFs persevere in errors typical of a global approach to experience.  相似文献

Fat is the second most abundant component of the nutrient composition of the mealworm Tenebrio molitor (Coleoptera: Tenebrionidae) that represents also an interesting source of PUFA, especially n-6 and n-3 fatty acids, involved in prevention of cardiovascular diseases. This study investigated the possibility of modifying the fat content and the FA composition of yellow mealworms through feeding and how this would be influenced by developmental stages, pupal sex, and generation with the future aim of applying this coleopteran as a diet supplement for human health. Growth rate and cumulative mortality percentage on the different feeding substrates were also evaluated to select the optimal conditions for a mass-raising of this insect species. Despite the different fat content in the six different breeding substrates used, T. molitor larvae and pupae contained a constant fat percentage (>34% in larvae and >30% in pupae). A similar total fat content was found comparing larvae and male and female pupae of the second generation to those of the first generation. On the contrary, FA composition differed both in larvae and pupae reared on the different feeding substrates. However, the exemplars reared on the diets based on 100% bread and 100% oat flour showed SFA, PUFA percentages, and an n-6/n-3 ratio more suitable for human consumption; the diet based on beer yeast, wheat flour, and oat flour resulted in a contemporary diet that most satisfied the balance between a fat composition of high quality and favorable growth conditions.  相似文献

Nonalcoholic fatty liver disease (NAFLD) is characterized by the accumulation of lipids in the liver. Given the high prevalence of NAFLD, its evolution to nonalcoholic steatohepatitis (NASH) and hepatocellular carcinoma (HCC) is of global concern. Therapies for managing NASH-driven HCC can benefit from targeting factors that play a continuous role in NAFLD evolution to HCC. Recent work has shown that postprandial liver translation exacerbates lipid accumulation through the activity of a translation factor, eukaryotic initiation factor 6 (eIF6). Here, we test the effect of eIF6 inhibition on the progression of HCC. Mice heterozygous for eIF6 express half the level of eIF6 compared to wt mice and are resistant to the formation of HCC nodules upon exposure to a high fat/high sugar diet combined with liver damage. Histology showed that nodules in eIF6 het mice were smaller with reduced proliferation compared to wt nodules. By using an in vitro model of human HCC, we confirm that eIF6 depletion reduces the growth of HCC spheroids. We also tested three pharmacological inhibitors of eIF6 activity—eIFsixty-1, eIFsixty-4, and eIFsixty-6—and all three reduced eIF6 binding to 60S ribosomes and limited the growth of HCC spheroids. Thus, inhibition of eIF6 activity is feasible and limits HCC formation.  相似文献
