Recombinant porcine myoglobin has been produced in Escherichiacoli using the cII fusion expression system of Nagai and Th?gersen[Nature, 309, 810–812 (1984)]. After processing and reconstitutionwith haem, the protein is gel-electrophoretically and spectrophotometricallyindistinguishable from native pig myoglobin. Large crystalsof both native and recombinant porcine myoglobin were grownfrom 50 mM sodium phosphate, pH 7.1, 80% ammonium sulphate.The crystals belong to space group C2 (a = 156.9 ?, b = 42.0?, c = 92.2 ?, ß = 127.9?) and diffract to a nominal2.5 ? resolution. We plan to explore apomyoglobin as a bindingsurface in studies combining site-directed mutagenesis and X-rayanalysis. These experiments will be extended by studying thebinding of haem analogues to the mutant apoproteins.  相似文献

Aerobic mineralization of several different paperboard materials under laboratory‐exposure conditions was studied. A novel respirometer was used to titrimetrically determine the kinetics of gas evolution and the rate of biodegradation was quantified by an empirical rate coefficient k. Various bleached paperboard materials studied showed about the same rate (k = 0.14–0.16 days−1) and a 50–58 weight percent conversion of substrate carbon into carbon dioxide under the conditions used in the study. © 1999 John Wiley & Sons, Inc. The rising level of atmospheric CO2 has stimulated several recent studies attempting to predict the effects of increased CO2 on ecological communities. However, most of these studies have been conducted in the benign conditions of the laboratory and in the absence of herbivores. In the current study, we utilized large octagonal chambers, which enclosed portions of an intact scrub-oak community to investigate the interactive effects of CO2 and insect herbivory on myrtle oak, Quercus myrtifolia. Specifically, we assessed the effects of ambient and elevated CO2 (2x current concentrations) on percent foliar nitrogen, C:N ratio, total relative foliar tannin content, and the presence of leaf damage caused by leaf mining and leaf chewing insects that feed on myrtle oak. Total foliar N declined and C:N ratios increased significantly in oaks in elevated CO2 chambers. The percentages of leaves damaged by either leafminers or leaf chewers tended to be lower in elevated compared to ambient chambers, but they co-occurred on leaves less than expected, regardless of CO2 treatment. Leaves that had been either mined or chewed exhibited a similar wounding or defensive response; they had an average of 25 and 21% higher protein binding ability, which is correlated with tannin concentration, compared to nondamaged control leaves, respectively. While the protein-binding ability (expressed as total percent tannin) of leaves from elevated CO2 was slightly higher than from leaves grown in ambient chambers, this difference was not significant.  相似文献

This paper discusses the discovery of uncommon taste or trigeminal active compounds and their associated sensory analysis using human tasting panels with the aim of enhancing the overall taste experience whilst reducing where possible the sugar and salt content of foods. The first example outlines the discovery of the sensory quality attributes of (R)-2-(carboxymethylamino)propanoic acid, named (R)-strombine, as assessed by a panel of 47 subjects to confirm its contribution to the typical taste of scallop muscle. The second example discusses the pungency and trigeminal effect of polygodial, which is compared with piperine and capsaicin, as well as the elucidation of a new structure eliciting a trigeminal effect, (+/-)-trans-2,3,3a,7a-tetrahydro-1 H-indene-4-carbaldehyde, discovered in Amomum tsao-ko. Finally, the time intensity trigeminal effect of (-)-menthol is compared with (1R,2RS,4RS)-1-isopropyl-4-methylbicyclo[3.1.0]hexan-2-ol, named dihydroumbellulol, a new cooling compound obtained by hemi-synthesis from umbellulone extracted from Umbellularia californica Nutt.  相似文献

We assessed the polyphenolic contents and antioxidant potential of the aqueous, ethanol and methanol stem bark extracts of Jatropha curcas. The total phenol, flavonoids, flavonols and proanthocyanidin contents of the extracts were evaluated to determine their effect on the antioxidant property of this plant, using standard phytochemical methods. The antioxidant and free radical scavenging activity of ethanol, methanol and aqueous extracts of the plant were also assessed against 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), ferric reducing, nitric oxide (NO), superoxide anion, (O(2) (-)) and hydrogen peroxide (H(2)O(2)) using spectroscopic methods and results were compared with that of butylated hydroxyl toluene (BHT) and ascorbic acid as standards. The concentrations of different classes of phenolic compounds were higher in methanol and ethanol extracts compared to aqueous extracts. There was correlation between total phenol, total flavonoids, total flavonol and total proanthocyanidins (r = 0.996, 0.978, 0.908, and 0.985) respectively. There was correlations between the amount of phenolic compounds and percentage inhibition of DPPH radicals scavenging activity of the extract (r = 0.98). Findings from the present study indicated that J. curcas is a potential source of natural antioxidants and may be a good candidate for pharmaceutical plant based products.  相似文献

Poultry farming generates large quantities of waste. The current practice is to spread this waste onto farmland as fertilizer. However, as the factory farms for poultry grow both in numbers and size, the amount of waste generated has increased significantly in recent years. In consequence, excessive application of poultry wastes on farmland is resulting in more and more nutrients entering the surface water. One of the options being considered is the use of poultry waste as power plant fuel. Since poultry-derived fuel (PDF) is biomass, its co-firing will have the added advantage of reducing greenhouse gas emissions from power generation. To evaluate the combustion characteristics of co-firing PDF with coal, combustion tests were conducted in CanmetENERGY's pilot-scale circulating fluidized bed combustor (CFBC). The goal of this program was to verify that PDF can be co-fired with coal and, more importantly, that emissions from the combustion process are not adversely affected by the presence of PDF in the feed. The test results were very promising and support the idea that co-firing in an existing CFBC boiler firing coal is a good way to utilize PDF, resolving a potential waste disposal problem while reducing the amount of CO2 released from the boiler.  相似文献

To develop and validate meshes for computational fluid dynamics (CFD) simulations of transport in fixed beds, a single particle is often used as a test case. We present results for drag coefficient (CD) and heat transfer Nusselt number (Nu) for flow past a sphere, focusing on high flow rates typical of industrial steam reformers (400 < Re < 20,000). Over this range, good predictions of CD were obtained using large eddy simulation (LES) to capture vortex shedding and wake dynamics, with a mesh refined downstream from the sphere. The small time-steps and high cell count required make this too expensive for fixed beds. Nu can be accurately calculated using a Reynolds-averaged Navier-Stokes (RANS) method with shear-stress transport (SST) k-ω closure provided the mesh at the particle surface is fine enough and covers most of the boundary layer. Single sphere simulations of heat transfer are more useful for fixed bed mesh development than drag coefficient calculations.  相似文献

A versatile methodology for the synthesis of 6/8-halogenated 7-aminocoumarins from the corresponding 7-hydroxy analogs using Pd-catalyzed amination reaction as the key step is presented. Further readily conversion into 7-azidocoumarins was performed and the resulting aryl azides proved higher stability and reactivity than the corresponding non-halogenated parent compound. These new compounds may thus constitute attractive scaffolds for designing novel photoaffinity reagents for various challenging bio-labeling applications.  相似文献

The aim of this study was to investigate the potential of using the resistivity and deposition rate data from Crossflow Sampler-Modified Fouling Index Ultrafiltration (CFS-MFIUF) measurements to determine the coupled effects of colloidal fouling and cake enhanced osmotic pressure (CEOP) effect. Cake filtration derived from CFS-MFIUF was combined with a CEOP model to predict the crossflow RO fouling profile under constant flux filtration. The prediction based on resistivity, I′ from CFS-MFIUF measurement alone was found to underestimate the RO fouling for high salinity solutions. However, when incorporating the mass information from the CFS-MFIUF test to account for the CEOP effect, the prediction showed good agreement with the TMP profile of the RO system. The results indicated that the CFS-MFIUF test which includes the CEOP effect is a very promising technique to provide an estimation of the RO colloidal fouling profile. When the changes of cake thickness and porosity throughout the filtration were considered, the predicted TMP profile based on the model clearly indicated a two-stage of fouling profile which agreed well with the experimental data. Additional studies on the effects of cake thickness and porosity on CEOP highlighted the important influence of cake structure on CEOP.  相似文献

The modification of silver paste electrodes with a combination of dodecylbenzenesulfonic acid and KCl has been shown to lead to significant enhancements of the electrochemical reduction of hydrogen peroxide. The catalytic enhancement was shown to be dependent on the concentration of the surfactant/salt solution, which resulted in increases of some 80-fold in amperometric response to hydrogen peroxide at −0.1 V vs Ag/AgCl, pH 6.8 over unmodified silver paste. Physical analysis showed modifications to both the surface morphology and chemical composition of the silver paste electrode surface. However, BET and electrochemical analysis revealed no significant change in surface area. It is suggested that the enhanced catalysis may result from the formation of stabilised surfactant/salt structures at the metal electrode surface. The electrode was also shown to be suitable for the amperometric detection of hydrogen peroxide with a limit of detection of 1.1 × 10−6 M (S/N = 3).  相似文献
