The interaction between sodium ascorbate and dopamine was investigated by three different parameters: radical intensity, prooxidant action, and cytotoxicity induction. Sodium ascorbate and dopamine produced the doublet and quartet ESR signals under alkaline conditions (pH 8.0-9.5), respectively. Addition of increasing concentrations of sodium ascorbate completely scavenged the dopamine radical and replaced the latter with its own radical. Similarly, dopamine slightly, but significantly reduced the radical intensity of sodium ascorbate. These two compounds stimulated the methionine oxidation and hydrogen peroxide generation in culture medium, but in combination, their stimulation activities were weakened. Both of these two compounds dose-dependently reduced the viable cell number of human oral squamous carcinoma HSC-4 cells, and their cytotoxic activity was significantly reduced by catalase. When these two compounds were mixed together before adding to HSC-4 cells, both of their cytotoxic activities were diminished. The present study demonstrates the interaction between sodium ascorbate and dopamine, which might modify their biological activities and generation of nerve disorders such as Parkinson's disease.  相似文献

We investigated the role of melatonin in the induction of gastric lesions induced by water immersion restraint stress or centrally administered thyrotropin-releasing hormone (TRH). Melatonin (0.1-1 ng) injected intracisternally (i.c) 30 min prior to stress dose-dependently inhibited the induction of gastric lesions by water immersion restraint stress, while 100 micrograms/kg, i.p. failed to protect the gastric mucosa. Preadministration of melatonin (1 ng, i.c.) significantly reduced (83%) the severity of gastric lesions induced by a TRH analogue (500 ng, i.c.). Serum melatonin concentrations 30 min after administration of 1 ng melatonin i.c. did not differ from those of rats receiving i.c. vehicle. These results suggest that melatonin plays a protective, anti-stress, role in the gastric mucosa via a mechanism involving the central nervous system.  相似文献

We performed X-ray diffraction and neutron scattering experiments on high quality single crystals of La1.85Sr0.15CuO4. We found a forbidden (1,0,0) peak, which develops below the HTT-LTO structural transition temperature, and an anomalous behavior of acoustic phonon at (2,2,0). These results indicate that the structural anomaly has a strong relation with an superconductivity.  相似文献

A pregnant woman was referred to our hospital at the 29th week of gestation with the symptom of polyhydramnios; diaphragmatic herniation of the fetus was suspected. Fetal chromosome anlaysis was performed using fibroblasts obtained by aminocentesis, and mosaicism of 46,XX and isochromosome of 12p were diagnosed. Out of 50 karyotyped cells, 19 (38.0%) showed the tetrasomy of the isochromosome of 12p. The mother vaginally delivered a baby girl who died just after delivery. The analysis of cord blood lymphocytes revealed only 0.5% incidence of tetrasomy of 12p. The incidence of tetrasomy was 8.0% for the placental chorionic villi, 48.0% for the fibroblasts obtained from the umbilical cord, and 70.0% for the skin fibroblasts. Thus, the diagnosis of Pallister-Killian Syndrome (PKS) is confirmed by mosaicism of i(12p), that is, the affected patients exhibit tissue-specific mosaicism, with the abnormal karyotype expression in limited lymphocytes, but marked in fibroblasts.  相似文献

Escherichia coli HKY56, which demonstrated resistance to various beta-lactams except carbapenems, was isolated from the throat swab of an inpatient in 1994. Conjugal transfer of cephem resistance from HKY56 to E. coli CSH2 was not successful. Three cefotaxime-resistant E. coli clones harboring plasmid pMRE001, pMRE002, or pMRE003, each of which carried a 3.4-, 5.8-, or 6.2-kb EcoRI fragment insert, respectively, were obtained from HKY56. Although restriction analysis suggested their different origins, these clones showed similar profiles of resistance to various beta-lactams. The sequence of 10 amino acid residues at the N terminus of beta-lactamase purified from E. coli HB101(pMRE001) was identical to that of Toho-1. This Toho-1-like beta-lactamase-1 (TLB-1) was able to hydrolyze cefoperazone and cefotaxime efficiently, but it failed to hydrolyze cephamycins. A Toho-1-specific DNA probe was hybridized with three distinct EcoRI fragments derived from the chromosomal DNA of strain HKY56, and these fragments corresponded to DNA inserts carried by pMRE001, pMRE002, and pMRE003, respectively. PCR and Southern hybridization analysis suggested that all six cephem-resistant E. coli strains, strains HKY273, HKY285, HKY288, HKY305, HKY316, and HKY335, which were isolated in 1996 at the same hospital where strain HKY56 had been isolated, also possessed multiple Toho-1-like beta-lactamase (TLB) genes, and the hybridization patterns obtained with the Toho-1-specific probe were quite similar among these six isolates. The DNA fingerprinting patterns observed by pulsed-field gel electrophoresis revealed that among the E. coli isolates tested, all isolates except HKY56 possessed a similar genetic background. These findings suggested that E. coli strains that carry chromosomally multiplied TLB genes may have been proliferating and transmitted among patients in the same hospital.  相似文献

A novel technique for the fabrication of photonic crystal (PC) nanocavities coupled with colloidal nanocrystals is presented. A waveguiding resist membrane embedding highly emitting dot-in-a-rod nanocrystals was patterned through e-beam lithography and released through wet etching process. The proposed approach makes the PC structure independent of fabrication imperfections induced by etching steps. Micro-photoluminescence spectra revealed degenerated resonant modes (Q-factor approximately 700) whose fabrication-induced spectral splitting is comparable to the full width at half-maximum of the peaks. Active nanocavities tunable from visible to infrared spectral range on GaAs or Si substrates can be easily implemented by this technique.  相似文献

We have investigated ultrasonic degradations of poly(ethylene oxide) (PEG) and poly(methyl methacrylate) (PMMA) in aqueous media by means of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS). The ultrasonic degradation of polymers was monitored as a function of ultrasonication duration to examine the structural details of ultrasonic degradation polymers. PEG solution ultrasonication produced five types of oligomers (M approximately 1000 Da) with different end groups, irrespective of the initial average molecular masses (M=2, 6, 20, and 2000 kDa). Several degradation pathways with free radical reactions have been suggested to explain these degradation products: the ultrasonic degradation of PEG is initiated by breaking of the C-O bond in the PEG chain, generating polymeric radicals with two terminal groups, i.e., X*( approximately CH2CH2*) and Y*( approximately CH2CH2O*), followed by termination with extraction or release of a hydrogen atom. However, PMMA (M=1630 Da) ultrasonication generated only one type of degradation oligomer, which has a hydrogen group at both ends, the same as that of the original oligomer. It has been suggested that the presence of the radical terminal groups X*( approximately CH2*) and Y*( approximately (CH3)CCOO(CH3)C*) is due to selective C-C bond breaking in the chain during the ultrasonic degradation of PMMA. The MALDI-TOFMS combined with the ultrasonic degradation technique (UD/MALDI-TOFMS) developed in this study could be extended to the analysis of synthetic polymer structures with high molecular weights.  相似文献
