Magnesium silicide (Mg2Si) thick films with (110) orientation were fabricated on (001) sapphire substrate using radiofrequency magnetron sputtering. Stoichiometric Mg2Si films with composition Si/(Mg + Si) = 0.33 were achieved over a range of vacuum from 10 mTorr to 140 mTorr and 300°C. On postannealing the film at 500°C, the out-of-plane lattice parameter shifted to lower values and the electrical conductivity increased by two orders of magnitude. A room-temperature Seebeck coefficient of 517 μV K?1 was observed and found to decrease with increasing temperature; the Seebeck coefficient remained at a constant positive value of 212 μV K?1 at 500°C. This can be related to the possibility of p-type conduction in this temperature range.  相似文献

The lipids extracted from the kernels of pumpkin (Cucurbita spp) seeds of three cultivars were classified by thin layer chromatography into six fractions: steryl esters (SEs, 0.5–1.2%), triacylglycerols (TAGs, 92.7–93.4%), free fatty acids (FFAs, 2.9–3.5%), sn‐1,3‐diacylglycerols (1,3‐DAGs, 0.4–0.9%), sn‐1,2‐diacylglycerols (1,2‐DAGs, 0.7–0.9%) and phospholipids (PLs, 1.5%). Fatty acids derivatised as methyl esters were analysed by gas chromatography with flame ionisation detection. Molecular species and fatty acid distributions of TAGs, isolated from the total lipids in the kernels, were analysed by a combination of argentation thin layer chromatography (TLC) and gas chromatography. A modified argentation TLC procedure, developed to optimise the separation of the complex mixture of total TAGs, provided 11 different groups of TAGs, based on both the degree of unsaturation and the total chain length of fatty acid groups. With a few exceptions, SM2 (5.8–20.1%), S2D (8.8–11.2%), M3 (6.7–24.8%), SMD (6.8–16.7%), M2D (16.7–23.6%), SD2 (4.6–15.1%) and MD2 (4.9–18.6%) were the main TAG components. These results suggest that there are significant differences (P < 0.05) not only in fatty acid distributions of acyl lipids but also in molecular species of TAGs among the three cultivars. The differences in pumpkin cultivars could be appreciable, based on the distribution of molecular species in TAGs. The subcellular localization of biomolecules at high resolution has traditionally been investigated by combining transmission electron microscopy (TEM) and chemical staining with heavy metals or immuno-based labeling with gold-conjugated antibodies. Here, we employ genetically encoded tags to examine the localization of proteins in transfected cultured cells by TEM. We purified a fusion protein of postsynaptic density-95 (PSD-95) coupled to three tandem repeats of metallothionein (MT) (PDS-95-3MT) from COS7 cells grown in the presence of Cd2+. PSD-95-3MT was detected as black particles by TEM. To visualize the subcellular localization of PSD-95-3MT, expression constructs encoding this fusion protein were transfected into primary hippocampal neurons cultured in medium containing Cd2+. The subcellular accumulation of PSD-95-3MT and Cd2+ provided excellent contrast in TEM micrographs. To address if genetically encoded tags affect the function of the target proteins, we found that the conjugation of 3MT to PSD-95 did not alter its association with known binding partners. These results demonstrate that 3MT coordinating Cd2+ is a valuable genetically encoded tag to study the localization of proteins by TEM.  相似文献

A detection of a defect of a helical heating tube installed in the fast breeder reactor “Monju” in Japan is done by a feeding of an eddy current testing (ECT) probe with magnetic sensor into the tube. An undesirable vibration of the ECT probe always happens under a certain condition and makes the inspection difficult. Several characteristics of the vibration have been made clear by some experiments using a mock-up, but the essential factor of the vibration is still unclear. In this paper, a numerical simulation of the vibration is implemented on the assumption that the vibration is caused by Coulomb friction. An analytical model, which is obtained as a lumped mass model, is a large-scale non-linear vibration system and many computational costs are ordinarily required to carry out the simulations. The Transfer Influence Coefficient Method is applied so that the simulation is efficiently carried out. The results of simulation qualitatively agree well with the experimental results. It confirms the validity of the assumption that the vibration is caused by Coulomb friction.  相似文献

Poly(ADP-ribosyl)ation is a post-translational modification of proteins by transferring poly(ADP-ribose) (PAR) to acceptor proteins by the action of poly(ADP-ribose) polymerase (PARP). Two tankyrase (TNKS) isoforms, TNK1 and TNK2 (TNKS1/2), are ubiquitously expressed in mammalian cells and participate in diverse cellular functions, including wnt/β-catenin signaling, telomere maintenance, glucose metabolism and mitosis regulation. For wnt/β-catenin signaling, TNKS1/2 catalyze poly(ADP-ribosyl)ation of Axin, a key component of the β-catenin degradation complex, which allows Axin’s ubiquitination and subsequent degradation, thereby activating β-catenin signaling. In the present study, we focused on the functions of TNKS1/2 in neuronal development. In primary hippocampal neurons, TNKS1/2 were detected in the soma and neurites, where they co-localized with PAR signals. Treatment with XAV939, a selective TNKS1/2 inhibitor, suppressed neurite outgrowth and synapse formation. In addition, XAV939 also suppressed norepinephrine uptake in PC12 cells, a rat pheochromocytoma cell line. These effects likely resulted from the inhibition of β-catenin signaling through the stabilization of Axin, which suggests TNKS1/2 enhance Axin degradation by modifying its poly(ADP-ribosyl)ation, thereby stabilizing wnt/β-catenin signaling and, in turn, promoting neurite outgrowth and synapse formation.  相似文献

Fucosylation is an oligosaccharide modification that plays an important role in immune response and malignancy, and specific fucosyltransferases (FUTs) catalyze the three types of fucosylations: core-type, Lewis type, and H type. FUTs regulate cancer proliferation, invasiveness, and resistance to chemotherapy by modifying the glycosylation of signaling receptors. Oligosaccharides on PD-1/PD-L1 proteins are specifically fucosylated, leading to functional modifications. Expression of FUTs is upregulated in renal cell carcinoma, bladder cancer, and prostate cancer. Aberrant fucosylation in prostate-specific antigen (PSA) could be used as a novel biomarker for prostate cancer. Furthermore, elucidation of the biological function of fucosylation could result in the development of novel therapeutic targets. Further studies are needed in the field of fucosylation glycobiology in urological malignancies.  相似文献

Soybean embryonic axes were separated from other tissues, i.e., the cotyledons and seed coat. The molecular species and FA distribution of TAG isolated from total lipids in the embryonic axes were analyzed by a combination of argentation-TLC and GC, and were investigated in relation to their tocopherol distribution, which was determined by HPLC. The dominant components were γ-tocopherols, with much smaller amounts of α-, β-, and δ-tocopherols. A modified argentation-TLC procedure, developed to optimize the separation of the complex mixture of total TAG, provided 16 different groups of TAG, based on both the degree of unsaturation and the total acyl-chain length of FA groups. With a few exceptions, the major TAG components were S2D (6.8–10.3%), SMD (6.9–11.2%), SD2 (7.2–9.8%), SMT (3.2–7.4%), SDT (11.5–19.5%), D3 (3.5–8.3%), MDT (4.5–7.7%), D2T (11.1–20.6%), and DT2 (8.2–15.7%) (where S denotes saturated FA, M denotes monoenes, D denotes dienes, and T denotes trienes). These results indicate that there were significant differences (P<0.05) not only in tocopherol distribution but also in the molecular species of TAG among the four cultivars. Therefore, these tissues should be made available as raw materials for soybean-germ oil or soy milk, based on the differences in the distributions of tocopherol homologs and the molecular species of TAG within the embryonic axes.  相似文献

Regiospecific distributions of fatty acids of triacylglycerols (TAG) and phospholipids (PL) separated from broad beans (Vicia faba) of four cultivars (Minpo, Sanuki, Nintoku and Sanren) were investigated. The major lipid components were PL (47.5–50.5 wt‐%) and TAG (47.7–50.1 wt‐%), while steryl esters, hydrocarbons, free fatty acids, diacylglycerols and monoacylglycerols were present in minor proportions (1.6–2.4 wt‐%). The PL components isolated from the four cultivars were phosphatidylcholine (56.4–58.4 wt‐%), phosphatidylethanolamine (20.3–21.7 wt‐%) and phosphatidylinositol (16.6–18.6 wt‐%). Phosphatidylinositol was unique in that it had the highest saturated fatty acid content among these PL. The principal characteristics of the fatty acid distribution in the TAG and PL were evident in the beans: Unsaturated fatty acids were predominantly concentrated in the sn‐2 position while saturated fatty acids primarily occupied the sn‐1 or sn‐3 position in these lipids. The lipid components and fatty acid distributions were almost the same in the four cultivars and were not influenced by genetic variability and planting location. These results could be useful information to both consumers and producers for the manufacture of traditional broad bean foods in Japan.  相似文献

Extracted lipids obtained from Jack beans (white and red) were fractionated by TLC into nine subfractions. The major components were TAGs (TAG: 43.8–45.7 wt%) and phospholipids (PL: 46.7–47.0 wt%), while other components were also present in minor proportions (0.3–2.7 wt%). The principal fatty acids (FA) are generally palmitic (18.8–28.8%), stearic (0.7–6.8%), oleic (42.0–51.8%), linoleic (16.2–22.8%), and α‐linolenic (3.0–8.2%) acids, the distribution of which differs according to these lipid classes. There were no significant differences (p>0.05) in the positional distribution of FA in the TAG; unsaturated FA (97.5%) were predominantly concentrated in the sn‐2 position while saturated FA (33.3%) primarily occupied the sn‐1 position or sn‐3 position. However, significant differences (p<0.05) in FA distribution existed when the individual PL were compared between the white and red beans. Based on the FA composition of these lipids, it seems that the two cultivars of Jack beans are very similar to each other with a few exceptions. The results could be useful to both producers and consumers for our daily diet to improve value of the Japanese diet. Practical applications : The lipid composition suggests that these beans could be a good source of nutraceuticals with providing heath benefits. The white and red beans may be well incorporated into our daily Japanese diets to improve nutritional value. The data obtained in this study provide valuable information for manufacturing functional drinks such as Jack bean tea in Japan.  相似文献
