Ion channels are pore-forming proteins that allow ions to flow across plasma membranes and intracellular organelles in both excitable and non-excitable cells. They are involved in the regulation of several biological processes (i.e., proliferation, cell volume and shape, differentiation, migration, and apoptosis). Recently, the aberrant expression of ion channels has emerged as an important step of malignant transformation, tumor progression, and drug resistance, leading to the idea of “onco-channelopathy”. Here, we review the contribution of ion channels and transporters in multiple myeloma (MM), a hematological neoplasia characterized by the expansion of tumor plasma cells (MM cells) in the bone marrow (BM). Deregulation of ion channels sustains MM progression by modulating intracellular pathways that promote MM cells’ survival, proliferation, and drug resistance. Finally, we focus on the promising role of ion channels as therapeutic targets for the treatment of MM patients in a combination strategy with currently used anti-MM drugs to improve their cytotoxic activity and reduce adverse effects.  相似文献

Schottky and Ohmic contacts–based electronics play an important role in highly sensitive detection of biomolecules and neural electric impulses, respectively. The reversible conversion between these two contacts appears especially important for multifunctional sensing by just one biosensor. Here, Schottky barrier height (SBH) is successfully tuned by triboelectric nanogenerator (TENG) and the same device is made to achieve reversible conversion between Schottky contact and Ohmic contact. In the same Schottky to Ohmic reversible (SOR) biosensor, highly sensitive detections of biomolecule (i.e., neurotransmitter) and neural electric signal are achieved at different contact states. The SOR biosensor reveals the feasibility of using one device to realize multifunctional detection. This work proposes a simple and significant method to achieve reversible tuning between the Schottky contact and Ohmic contact on one device by TENG, which exhibits great potential in developing multifunctional and high‐sensitivity biosensors, rectifiers, and other functional electronic devices.  相似文献

As previously demonstrated, contact‐electrification (CE) is strongly dependent on temperature, however the highest temperature in which a triboelectric nanogenerator (TENG) can still function is unknown. Here, by designing and preparing a rotating free‐standing mode Ti/SiO2 TENG, the relationship between CE and temperature is revealed. It is found that the dominant deterring factor of CE at high temperatures is the electron thermionic emission. Although it is normally difficult for CE to occur at temperatures higher than 583 K, the working temperature of the rotating TENG can be raised to 673 K when thermionic emission is prevented by direct physical contact of the two materials via preannealing. The surface states model is proposed for explaining the experimental phenomenon. Moreover, the developed electron cloud‐potential well model accounts for the CE mechanism with temperature effects for all types of materials. The model indicates that besides thermionic emission of electrons, the atomic thermal vibration also influences CE. This study is fundamentally important for understanding triboelectrification, which will impact the design and improve the TENG for practical applications in a high temperature environment.  相似文献

A two‐step grafting procedure was applied to cellulose/chitin (CC) mix fibers, namely: activation under the action of cold plasma discharges, followed by reaction with N‐isopropylacrylamide (NIPAAm) and poly(N‐isopropylacrylamide) (PNIPAAm) to obtain fibers with responsiveness to external stimuli. The graft samples were characterized using attenuated total reflection Fourier transform infrared spectroscopy, X‐ray photoelectron spectroscopy (XPS), X‐ray diffraction, scanning electron microscopy coupled with energy‐dispersive X‐ray microanalysis and antimicrobial testing. All obtained results confirm the morphological and structural changes after plasma treatment which determine the modification of cellulose fiber properties. It was estimated from XPS data that the degrees of modification/grafting were about 23% for CC/NIPAAm and 13% for CC/PNIPAAm. Such treatment could be transferred to practical technologies, particularly in textile applications and special applications in the medical field. This investigation aimed to adapt the total focusing method (TFM) algorithm (originated from the synthetic aperture focusing technique in digital signal processing) to accommodate a circular array of piezoelectric sensors (PZT) and characterise defects using guided wave signals for the development of a structural health monitoring system. This research presents the initial results of a broader study focusing on the development of a structural health monitoring (SHM) guided wave system for advance carbon fibre reinforced plastic (CFRP) composite materials. The current material investigated was an isotropic (aluminium) square plate with 16 transducers operating successively as emitter or sensor in pitch and catch configuration enabling the collection of 240 signals per assessment. The Lamb wave signals collected were tuned on the symmetric fundamental mode with a wavelength of 17 mm, by setting the excitation frequency to 300 kHz. The initial condition for the imaging system, such as wave speed and transducer position, were determined with post processing of the baseline signals through a method involving the identification of the waves reflected from the free edge of the plate. The imaging algorithm was adapted to accommodate multiple transmitting transducers in random positions. A circular defect of 10 mm in diameter was drilled in the plate, which is similar to the delamination size introduced by a low velocity impact event in a composite plate. Images were obtained by applying the TFM to the baseline signals, Test 1 data (corresponding to the signals obtained after introduction of the defect) and to the data derived from the subtraction of the baseline to the Test 1 signals. The result shows that despite the damage diameter being 40 % smaller than the wavelength, the image (of the subtracted baseline data) demonstrated that the system can locate where the waves were reflected from the defect boundary. In other words, the contour of the damaged area was highlighted enabling its size and position to be determined.  相似文献

The article describes some properties of cellulose acetates (CAs) with different substitution degrees. The hydrophilic/hydrophobic properties, morphological aspects, and interface properties with red blood cells and platelets are affected by the substitution degree, synthesis conditions, history of the formed films from solutions in acetone/water nonsolvent/nonsolvent mixtures, and low pressure plasma treatment. The results obtained are useful in biomedical applications, including evaluation of bacterial adhesion onto surfaces, or utilization of CA for semipermeable membranes. © 2012 Wiley Periodicals, Inc. (1) Background: Biomarkers might play a significant role in predicting the clinical outcomes of patients with ovarian cancer. By analyzing lipid metabolism genes, future perspectives may be uncovered; (2) Methods: RNA-seq data for serous ovarian cancer were downloaded from The Cancer Genome Atlas and Gene Expression Omnibus databases. The non-negative matrix factorization package in programming language R was used to classify molecular subtypes of lipid metabolism genes and the limma package in R was performed for functional enrichment analysis. Through lasso regression, we constructed a multi-gene prognosis model; (3) Results: Two molecular subtypes were obtained and an 11-gene signature was constructed (PI3, RGS, ADORA3, CH25H, CCDC80, PTGER3, MATK, KLRB1, CCL19, CXCL9 and CXCL10). Our prognostic model shows a good independent prognostic ability in ovarian cancer. In a nomogram, the predictive efficiency was notably superior to that of traditional clinical features. Related to known models in ovarian cancer with a comparable amount of genes, ours has the highest concordance index; (4) Conclusions: We propose an 11-gene signature prognosis prediction model based on lipid metabolism genes in serous ovarian cancer.  相似文献

A new and efficient route using inexpensive reagents such as fumed silica and tetrapropylammonium bromide is proposed for the synthesis of titanium silicalite-1. High crystalline titanium silicalite-1 was obtained in alkaline medium, using sodium hydroxide as HO? ion source required for the crystallization process. Hydrolysis of titanium source with formation of insoluble oxide species was prevented by titanium complexation with before structuring gel. The final solids were fully characterized by powder X-ray diffraction, scanning electron microscopy, Fourier transform infrared, ultraviolet-visible diffuse reflectance, Raman and atomic absorption spectroscopies, as well as nitrogen sorption analysis. It was found that a molar ratio Ti:Si of about 0.04 in the initial reaction mixture is the upper limit to which well formed titanium silicalite-1 with channels free of crystalline or amorphous material can be obtained. Above this value, solids with MFI type structure containing both Ti isomorphously substituted in the network and extralattice anatase nanoparticles inside of channels is formed.  相似文献
