We present a fluorescence lifetime imaging technique with simultaneous spectral and temporal resolution. The technique is fully compatible with the commonly used multiphoton microscopes and nondescanned (direct) detection. An image of the back-aperture of the microscope lens is projected on the input of a fiber bundle. The input of the fiber bundle is circular, and the output is flattened to match the input slit of a spectrograph. The spectrum at the output of the spectrograph is projected on a 16-anode PMT module. For each detected photon, the encoding logics of the PMT module deliver a timing pulse and the number of the PMT channel in which the photon was detected. The photons are accumulated by a multidimensional time-correlated single photon counting (TCSPC) process. The recording process builds up a four-dimensional photon distribution over the times of the photons in the excitation pulse period, the wavelengths of the photons, and the coordinates of the scan area. The method delivers a near-ideal counting efficiency and is capable of resolving double-exponential decay functions. Notes that electrolytic lesions of the centrum-medianum and parafascicularis complex of the thalamus produce, in the rat, large deficits of both acquisition and retention of active-avoidance responses, especially in the 2-way shuttle-box test. The effect of such lesions on 26 male cats and 6 male and 2 female macaques was examined. It was found that lesions in the cat did not produce significant effects, either with acoustical or visual CS, during postoperative relearning. Moreover, rate of extinction was not affected. Negative results were also found in monkeys. These lesions did not modify the threshold of flinch or jump responses to electric shock in either species. (20 ref.) In the case study presented in this paper we consider early development phases of a mechanical product. We want to evaluate different concepts and decide which one(s) to pursue. A problem in early phases is that usually no test runs are available. In our case study, based on a standard, there are ways to compute the lifetime distributions of the components of the different concepts. Some parameters needed for these computations are not known precisely. Unfortunately, the lifetime distributions of the components are highly sensitive to these parameters. Our approach is to equip these parameters with distributions. These distributions would be called prior distributions in Bayesian terminology, but no update is possible since no test runs are available. Our approach implies that the distribution of the system lifetime for each concept is random, i.e. we get random elements in the space of lifetime distributions. Using Monte-Carlo simulations, we demonstrate several ways to compare the random lifetime distributions of the concepts. Some of these comparisons use stochastic orderings. We also introduce a new stochastic ordering which is particularly suitable for reliability purposes. Our case study, consisting of three scenarios, allows us to demonstrate some conclusions that can be reached. 相似文献
Mensacarcin is a potent cytotoxic agent isolated from Streptomyces bottropensis. It possesses a high content of oxygen atoms and two epoxide groups, and shows cytostatic and cytotoxic activity comparable to that of doxorubicin, a widely used drug for antitumor therapy. Another natural compound, rishirilide A, was also isolated from the fermentation broth of S. bottropensis. Screening a cosmid library of S. bottropensis with minimal PKS-gene-specific primers revealed the presence of three different type II polyketide synthase (PKS) gene clusters in this strain: the msn cluster (mensacarcin biosynthesis), the rsl cluster (rishirilide biosynthesis), and the mec cluster (putative spore pigment biosynthesis). Interestingly, luciferase-like oxygenases, which are very rare in Streptomyces species, are enriched in both the msn cluster and the rsl cluster. Three cosmids, cos2 (containing the major part of the msn cluster), cos3 (harboring the mec cluster), and cos4 (spanning probably the whole rsl cluster) were introduced into the general heterologous host Streptomyces albus by intergeneric conjugation. Expression of cos2 and cos4 in S. albus led to the production of didesmethylmensacarcin (DDMM, a precursor of mensacarcin) and the production of rishirilide A and B (a precursor of rishirilide A), respectively. However, no product was detected from the expression of cos3. In addition, based on the results of isotope-feeding experiments in S. bottropensis, a putative biosynthesis pathway for mensacarcin is proposed. 相似文献
Being the most proliferative journal of oncology a cancer research of the past decade, the Open Access journal Oncotarget had reached more than 20,000 publications and a relatively high impact factor score in the past years. In 2018, the journal citation report decided to withdraw the status of an impact factor journal. Since there was a large discussion in the scientific community and specific reasons for the withdrawal were not stated, this bibliometric analysis was performed to assess if Oncotarget exhibits any differences in its bibliometric structure compared to other journals. For this purpose, we used the “New Quality and Quantity Indices in Sciences” platform and analyzed 20,000 Oncotarget articles. Density equalizing mapping technique helps to construct maps of cancer research in Oncotarget and shows that it has led to a unique global landscape which is not asymmetrically dominated by the Western hemisphere but exhibits a publishing architecture with a pronounced emphasis on Chinese articles.
The major intent of this article was to describe the design principles of the drug-therapy documentation module of the Patient Data Management System (PDMS) ICUData, in routine use at the intensive care unit (ICU) of the Department of Anesthesiology and Intensive Care Medicine at the University Hospital of Giessen, Germany, since February 1999. The new drug management system has been in routine use since March 2000. Until 8 January 2001, 1140 patients have been documented using this approach. It could be demonstrated that it was possible to transform the formerly unstructured text-based documentation into a detailed and structured model. The mediated benefit resulted in the automatic calculation of fluid balance. Further, detailed statistical analyses of therapeutic behavior in drug administration are now possible. 相似文献
The adsorption of ethyl pyruvate on Pt(111) has been studied by in situ XANES measurements in the presence and absence of
hydrogen. Depending on the hydrogen and ethyl pyruvate pressure, the C and O K‐edge spectra exhibit distinctly different angular
dependence. Without hydrogen ethyl pyruvate is oriented preferentially perpendicular to the surface, indicating bonding via
the O lone pairs. In the presence of hydrogen the mean orientation is more tilted towards the surface. Likely, ethyl pyruvate
also interacts with Pt via its π system under these conditions. The observed angle‐dependent shift of the energy of the π*
and σ* resonances indicates the coexistence of differently adsorbed ethyl pyruvate species. The experimental findings demonstrate
the importance of the in situ approach for unraveling the adsorption mode of ethyl pyruvate in the enantioselective hydrogenation
over cinchona‐alkaloid‐modified Pt.

In interactive theorem proving, tactics and tacticals have been introduced to automate proof search. In this scenario, user interaction traditionally is restricted to the mode in which the user decides which tactic to apply on the top-level, without being able to interact with the tactic once it has begun running.We propose a technique to allow the implementation of derivational analogy in tactical theorem proving. Instead of replaying tactics including backtracked dead ends our framework makes choice points in tactics explicit and thus avoids dead ends when reusing tactics. Additionally users can override choices a tactic has made or add additional steps to a derivation without terminating the tactic. The technique depends on an efficient replay of tactic executions without repeating search that the original computation may have involved. 相似文献
A filtration system for keeping the analytics free of particles is usually required for inline characterization of particle‐laden process gasses. Additionally gas pumps may be needed for overcoming the pressure drop of filter element and filter cake. Consecutive reactions of process gas and filter cake may be disadvantageous for the quality of the gas analysis. A new probing apparatus is presented for extraction of particle‐free gas samples from particle‐laden process gasses. The probe combines the functionalities of a gas pump and a particle separator. While the gas is transported from process to analysis, particles are separated according to the principle of air classification. 相似文献
