The formation of acrylamide during heating of certain foodstuffs constitutes a potential health hazard. The health risk assessment should be based on knowledge about the relation between dietary exposure to acrylamide and internal doses of acrylamide and its genotoxic metabolite glycidamide. The primary aim of this study in mice was to measure these relationships at low levels of acrylamide intake through the diet. A secondary aim was to clarify which extraction method should be used when analyzing acrylamide in food in order to obtain a correct measure of the acrylamide that is available for absorption. In the analysis procedure, alkaline extraction has earlier shown much higher measured acrylamide levels in certain foods compared to water extraction. In this subcronic study the administered diets were composed to give five levels of acrylamide intakes between 3 and 50 mug/kg body weight per day (calculated on figures obtained after water extraction). Internal doses of acrylamide and glycidamide were measured through hemoglobin (Hb)-adducts. The results showed linear relationships between the exposure of acrylamide and Hb-adduct levels from both acrylamide and glycidamide at these low exposure levels. The study also showed that the "extra" acrylamide measured with alkaline extraction does not correspond to bioavailable acrylamide.  相似文献

Current breeding tools aiming to improve feed efficiency use definitions based on total dry matter intake (DMI); for example, residual feed intake or feed saved. This research aimed to define alternative traits using existing data that differentiate between feed intake capacity and roughage or concentrate intake, and to investigate the phenotypic and genetic relationships among these traits. The data set contained 39,017 weekly milk yield, live weight, and DMI records of 3,164 cows. The 4 defined traits were as follows: (1) Feed intake capacity (FIC), defined as the difference between how much a cow ate and how much she was expected to eat based on diet satiety value and status of the cow (parity and lactation stage); (2) feed saved (FS), defined as the difference between the measured and the predicted DMI, based on the regression of DMI on milk components within experiment; (3) residual roughage intake (RRI), defined as the difference between the measured and the predicted roughage intake, based on the regression of roughage intake on milk components and concentrate intake within experiment; and (4) residual concentrate intake (RCI), defined as the difference between the measured and the predicted concentrate intake, based on the regression of concentrate intake on milk components and roughage intake within experiment. The phenotypic correlations were ?0.72 between FIC and FS, ?0.84 between FS and RRI, and ?0.53 between FS and RCI. Heritability of FIC, FS, RRI, and RCI were estimated to be 0.21, 0.12, 0.15, and 0.03, respectively. The genetic correlations were ?0.81 between FS and FIC, ?0.96 between FS and RRI, and ?0.25 between FS and RCI. Concentrate intake and RCI had low heritability. Genetic correlation between DMI and FIC was 0.98. Although the defined traits had moderate phenotypic correlations, the genetic correlations between DMI, FS, FIC, and RRI were above 0.79 (in absolute terms), suggesting that these traits are genetically similar. Therefore, selecting for FIC is expected to simply increase DMI and RRI, and there seems to be little advantage in separating concentrate and roughage intake in the genetic evaluation, because measured concentrate intake was determined by the feeding system in our data and not by the genetics of the cow.  相似文献

The ability of the pediocin AcH producing Lactobacillus plantarum DDEN 11007 and its non-producing plasmid-cured isogenic variant, DDEN 12305 to prevent the persistence and growth of Listeria monocytogenes EP2 in two gastrointestinal (GI) tract models was examined. In vitro studies conducted in a two-stage continuous flow system showed that L. plantarum DDEN 11007 inhibited L. monocytogenes EP2 under these conditions, while less effect was seen of the non-bacteriocin producing variant. The inhibitory effect was more pronounced at pH 5 than at pH 7. No effect on persistence of L. monocytogenes in the GI tract was seen in gnotobiotic rats colonized with either the pediocin AcH producing or the non-bacteriocin producing variant of L. plantarum when compared to rats inoculated with L. monocytogenes EP2 alone. Surprisingly, inoculation of the gnotobiotic animals with either of the L. plantarum strains prior to inoculation with L. monocytogenes EP2 resulted in increased occurrence of L. monocytogenes in liver and spleen when compared to the animals inoculated with L. monocytogenes EP2 alone. Our results indicate that the presence of L. plantarum in the gut of gnotobiotes facilitates L. monocytogenes invasion by an unknown mechanism. This observation is however not necessarily specifically related to L. plantarum, and should not be interpreted as the expected effect in animals carrying a conventional intestinal microflora.  相似文献

The accuracy of genomic prediction determines response to selection. It has been hypothesized that accuracy of genomic breeding values can be increased by a higher density of variants. We used imputed whole-genome sequence data and various single nucleotide polymorphism (SNP) selection criteria to estimate genomic breeding values in Brown Swiss cattle. The extreme scenarios were 50K SNP chip data and whole-genome sequence data with intermediate scenarios using linkage disequilibrium-pruned whole-genome sequence variants, only variants predicted to be missense, or the top 50K variants from genome-wide association studies. We estimated genomic breeding values for 3 traits (somatic cell score, nonreturn rate in heifers, and stature) and found differences in accuracy levels between traits. However, among different SNP sets, accuracy was very similar. In our analyses, sequence data led to a marginal increase in accuracy for 1 trait and was lower than 50K for the other traits. We concluded that the inclusion of imputed whole-genome sequence data does not lead to increased accuracy of genomic prediction with the methods.  相似文献

Occurrence and importance of psychrotolerant histamine producing bacteria in chilled fresh tuna were demonstrated in the present study. The objective was to evaluate microbial formation of histamine and biogenic amines in chilled fresh tuna from the Indian Ocean and stored either vacuum-packed (VP) or modified atmosphere-packed (MAP). Firstly, biogenic amines and the dominating microbiota were determined in VP tuna involved in an outbreak of histamine fish poisoning in Denmark. Secondly, the microbiota of fresh MAP tuna was evaluated at the time of processing in Sri Lanka and chemical, microbial and sensory changes were evaluated during storage at 1-3 degrees C. To explain the results obtained with naturally contaminated tuna the effect of VP and MAP on biogenic amine formation by psychrotolerant bacteria was evaluated in challenge tests at 2 degrees C and 10 degrees C. The VP tuna that caused histamine fish poisoning had a histamine concentration of >7000 mg/kg and this high concentration was most likely produced by psychrotolerant Morganella morganii-like bacteria or by Photobacterium phosphoreum. Similar psychrotolerant M. morganii-like bacteria dominated the spoilage microbiota of fresh MAP tuna with 60% CO2/40% N2 and formed >5000 mg/kg of histamine after 24 days at 1.7 degrees C. These psychrotolerant bacteria were biochemically similar to M. morganii subsp. morganii and their 16S rDNA (1495 bp) showed >98% sequence similarity to the type strain of this species. Toxic concentrations of histamine were produced at 2.1 degrees C in inoculated VP tuna by both the psychrotolerant M. morganii-like bacteria (7400+/-1050 mg/kg) and P. phosphoreum (4250+/-2050 mg/kg). Interestingly, MAP with 40% CO2/60% O2, in challenge tests, had a strong inhibitory effect on growth and histamine formation by both the psychrotolerant M. morganii-like bacteria and P. phosphoreum. In agreement with this, no formation of histamine was found in naturally contaminated fresh MAP tuna with 40% CO2/60% O2 during 28 days of storage at 1.0 degrees C. To reduce current problems with histamine fish poisoning due to VP tuna it is suggested, for lean tuna loins, to replace vacuum packaging with MAP containing approximately 40% CO2 and approximately 60% O2.  相似文献

White tea (WT) is rich in flavan‐3‐ols as green tea (GT) and might provide health protective effects due to the strong antioxidant properties of flavan‐3‐ols. Since intervention studies with WT are lacking, we evaluated the effects of WT consumption on antioxidant status, antioxidant capacity and biomarkers of oxidative stress compared to water and GT. After an overnight fast, 70 healthy non‐smokers were randomized to consume 600 mL of WT, GT or water (control). Plasma (epi‐)catechin and epi(gallo)catechingallate, antioxidant capacity (Folin assay, trolox equivalent antioxidant capacity test), 8‐iso‐prostaglandin F, ascorbic acid and uric acid were determined before and several times within 8 h after consumption. DNA strand breaks were measured in vivo and ex vivo (H2O2 stimulation) in leukocytes. Plasma flavan‐3‐ols significantly increased after WT and GT ingestion. Trolox equivalent antioxidant capacity was lower after 5 h in controls versus WT (p=0.031) and GT (p=0.005). Folin‐Ciocalteu reducing capacity, ascorbic and uric acid as well as markers of oxidative stress (8‐iso‐prostaglandin‐F, DNA strand breaks) were not affected by the beverages. A short‐term increase of catechins does not change plasma antioxidant capacity in healthy subjects. Conclusions with respect to health protective effects of WT and GT on the basis of these biomarkers can, thus, not be drawn.  相似文献

This is the first account of PCDDs, PCDFs, and non-ortho PCBs in Canadian Arctic seabirds. Livers and eggs of thick-billed murres, northern fulmars, and black-legged kittiwakes were collected in 1975 and 1993 from Prince Leopold Island in Lancaster Sound, Canada. Detectable concentrations of PCDDs, PCDFs, and non-ortho PCBs were found in all the Arctic seabird samples analyzed. Of the PCDD congeners assayed, only 2,3,7,8-substituted PCDDs were detected in the samples, whereas non-2,3,7,8-substituted PCDFs were found in addition to 2,3,7,8-substituted PCDFs in some of the samples. The predominant PCDD/F congener found in the livers of all three species was 2,3,4,7,8-PnCDF, both in 1975 and 1993. Concentrations of most dioxins and furans decreased in the fulmars and kittiwakes between 1975 and 1993 but increased in the murres. Of the non-ortho PCBs measured, PCB-126 occurred in the highest concentrations and contributed the majority of the non-ortho PCB-TEQ in all three species in both years. The highest concentrations of dioxins and furans as well as the highest TEQ values were found in the northern fulmar livers in both 1975 and 1993. Concentrations of some of the PCDDs and PCDFs are among the highest reported for Canadian Arctic biota.  相似文献

Fourier-transformed infrared spectroscopy has been established for analysing most of the physical–chemical parameters of honey. Additionally this technique can be used for determination of the botanical origin of a honey sample by comparison of the mid-infrared spectra. In this study calibrations for authentification of the main regional honey types should be developed to be able to measure simultaneously physical–chemical properties and the botanical origin at a minimum of time and at low costs. Honey samples from local beekeepers were collected and characterized by standard methods. The most common honey types were used for calibration. We recorded the mid infrared spectrum from each honey sample. Classification models were achieved by PCA-calibration and validated with samples from various botanical origins. Honey of Brassica spp., Robinia pseudoacacia, Calluna vulgaris, Trifolium spp., Tilia spp., Helianthus annuus, Centaurea cyanus and honeydew honey was used for calibration. Afterwards the calibration models were improved during routine analysis. Most of the honey samples from rape, false acacia, heather and honeydew can be classified correctly by the FTIR in consideration of the physical–chemical and sensorial properties.  相似文献
