We report the crystal structure of a thermophilic “ene” reductase (TOYE) isolated from Thermoanaerobacter pseudethanolicus E39. The crystal structure reveals a tetrameric enzyme and an active site that is relatively large compared to most other structurally determined and related Old Yellow Enzymes. The enzyme adopts higher order oligomeric states (octamers and dodecamers) in solution, as revealed by sedimentation velocity and multiangle laser light scattering. Bead modelling indicates that the solution structure is consistent with the basic tetrameric structure observed in crystallographic studies and electron microscopy. TOYE is stable at high temperatures (Tm>70 °C) and shows increased resistance to denaturation in water‐miscible organic solvents compared to the mesophilic Old Yellow Enzyme family member, pentaerythritol tetranitrate reductase. TOYE has typical ene‐reductase properties of the Old Yellow Enzyme family. There is currently major interest in using Old Yellow Enzyme family members in the preparative biocatalysis of a number of activated alkenes. The increased stability of TOYE in organic solvents is advantageous for biotransformations in which water‐miscible organic solvents and biphasic reaction conditions are required to both deliver novel substrates and minimize product racemisation.  相似文献

Calcium cobaltite Ca3Co4O9, abbreviated Co349, is a promising thermoelectric material for high-temperature applications in air. Its anisotropic properties can be assigned to polycrystalline parts by texturing. Tape casting and pressure-assisted sintering (PAS) are a possible future way for a cost-effective mass-production of thermoelectric generators. This study examines the influence of pressure and dwell time during PAS at 900°C of tape-cast Co349 on texture and thermoelectric properties. Tape casting aligns lentoid Co349. PAS results in a textured Co349 microstructure with the thermoelectrically favorable ab-direction perpendicular to the pressing direction. By pressure variation during sintering, the microstructure of Co349 can be tailored either toward a maximum figure of merit as required for energy harvesting or toward a maximum power factor as required for energy harvesting. Moderate pressure of 2.5 MPa results in 25% porosity and a textured microstructure with a figure of merit of 0.13 at 700°C, two times higher than the dry-pressed, pressureless-sintered reference. A pressure of 7.5 MPa leads to 94% density and a high power factor of 326 µW/mK2 at 800°C, which is 11 times higher than the dry-pressed reference (30 MPa) from the same powder.  相似文献

Protein concentrates prepared by expanded bed adsorption (EBA) chromatography from industrial potato juice (PJ) were analysed for chemical composition, color, enzyme activities, thermal properties and selected functional properties (solubility and emulsifying stability). Two EBA multi-modal resins, MIMO I-45 and MIMO 1300 (UpFront Chromatography), were employed under various pH conditions resulting in four potato protein concentrates, A-D. Concentrate B contained an electrophoretically pure protease inhibitor fraction (20-21 kDa), whereas concentrate A, C and D contained both patatin (41 kDa) and protease inhibitors. The potato protein concentrates were explored for the presence of transitions from native to denatured states using differential scanning calorimetry (DSC). Concentrate C had lower heat of transition (ΔH) and T-onset than the other concentrates. The concentrate containing protease inhibitors exhibited the highest denaturation temperature and enthalpy. All concentrates differed significantly (P < 0.05) in color brightness, with concentrate B and D emerging as the brightest. The solubility of the concentrates was evaluated at pH 6 and pH 4.5. All concentrates had lower solubility at pH 4.5 than at pH 6 (70-80%). The stability of emulsions (1% protein, 20% oil, 0.08% xanthan gum) against creaming was analysed with a new method based on the Single electrode Capacitance Probe (SeCaP) technology. Small differences among concentrates were observed by the new SeCaP method.  相似文献

This paper reports on a laboratory experiment concerning frost formation and moisture condensation in fibrous insulation based on stone–wool. Frost formation in samples of stone–wool open to air was noted in cases when temperature field over the specimen was between +20 and ?20 °C and air on the warm side was saturated with moisture.Frost accumulated with time in the part of the specimen facing the cold air. In the part of the specimen facing the warm humid air condense formation occurred. In this part the material had moisture content considerably higher than what could be anticipated from data such as moisture isotherms.Border between frost and liquid condensate was quite sharp in the specimens of higher density. Moisture content mass by mass has an upward trend with decreasing density of the material sample. Moisture resistance factor was found to be quite high at these circumstances. A model has been developed for studying the effect of three hypothetical approaches in reducing electricity and energy demand in the Norwegian building stock: wide diffusion of thermal carriers, heat pumps and conservation measures, respectively. Combinations of these are also considered. The model has a demand side perspective, considers both residential and service sectors, and calculates energy flows from net to delivered energy. Energy demand is given by the product of activity and intensity matrices. The activity levels are defined for the stock and the new construction, renovation and demolition flows. The intensity properties are defined in archetypes, and are the result of different energy class and heating carriers share options. The scenarios are shaped by combining the activity flows with different archetypes. The results show that adopting conservation measures on a large scale does allow reducing both electricity and total energy demand from present day levels while the building stock keeps growing. The results also highlight the importance of making a clear distinction between the assumptions on intensity and activity levels.  相似文献

The paper investigates an online version of the vehicle routing problem with time windows, in which additionally arriving requests cause the revision of so far followed routes and schedules. An extended online optimization framework is proposed, which automatically adapts to problem variations and enables the explicit consideration of up-to-date knowledge about the current performance of the controlled system. Actually, we use the mean punctuality observed in the transportation system to adjust the objective function utilized for solving the next decision problem instance. The search is guided toward least cost solutions coming along with high punctuality. We prove the applicability of this approach within comprehensive numerical experiments.  相似文献

We present a method for accurate image registration and motion compensation in multidimensional signals, such as two-dimensional (2-D) X-ray images and three-dimensional (3-D) computed tomography/magnetic resonance imaging volumes. The method is based on phase from quadrature filters, which makes it robust to noise and temporal intensity variations. The method is equally applicable to signals of two, three or higher number of dimensions. We use parametric models, e.g., affine models, finite elements or local affine models with global regularization. Experimental results show high accuracy for 2-D and 3-D motion compensation.  相似文献
