The β2 subunit of Na+, K+-ATPase was originally identified as the adhesion molecule on glia (AMOG) that mediates the adhesion of astrocytes to neurons in the central nervous system and that is implicated in the regulation of neurite outgrowth and neuronal migration. While β1 isoform have been shown to trans-interact in a species-specific mode with the β1 subunit on the epithelial neighboring cell, the β2 subunit has been shown to act as a recognition molecule on the glia. Nevertheless, none of the works have identified the binding partner of β2 or described its adhesion mechanism. Until now, the interactions pronounced for β2/AMOG are heterophilic cis-interactions. In the present report we designed experiments that would clarify whether β2 is a cell–cell homophilic adhesion molecule. For this purpose, we performed protein docking analysis, cell–cell aggregation, and protein–protein interaction assays. We observed that the glycosylated extracellular domain of β2/AMOG can make an energetically stable trans-interacting dimer. We show that CHO (Chinese Hamster Ovary) fibroblasts transfected with the human β2 subunit become more adhesive and make large aggregates. The treatment with Tunicamycin in vivo reduced cell aggregation, suggesting the participation of N-glycans in that process. Protein–protein interaction assay in vivo with MDCK (Madin-Darby canine kidney) or CHO cells expressing a recombinant β2 subunit show that the β2 subunits on the cell surface of the transfected cell lines interact with each other. Overall, our results suggest that the human β2 subunit can form trans-dimers between neighboring cells when expressed in non-astrocytic cells, such as fibroblasts (CHO) and epithelial cells (MDCK). 相似文献

Mass spectrometry (MS)-based techniques can be a powerful tool to identify neuropsychiatric disorder biomarkers, improving prediction and diagnosis ability. Here, we evaluate the efficacy of MS proteomics applied to human peripheral fluids of schizophrenia (SCZ) patients to identify disease biomarkers and relevant networks of biological pathways. Following PRISMA guidelines, a search was performed for studies that used MS proteomics approaches to identify proteomic differences between SCZ patients and healthy control groups (PROSPERO database: CRD42021274183). Nineteen articles fulfilled the inclusion criteria, allowing the identification of 217 differentially expressed proteins. Gene ontology analysis identified lipid metabolism, complement and coagulation cascades, and immune response as the main enriched biological pathways. Meta-analysis results suggest the upregulation of FCN3 and downregulation of APO1, APOA2, APOC1, and APOC3 in SCZ patients. Despite the proven ability of MS proteomics to characterize SCZ, several confounding factors contribute to the heterogeneity of the findings. In the future, we encourage the scientific community to perform studies with more extensive sampling and validation cohorts, integrating omics with bioinformatics tools to provide additional comprehension of differentially expressed proteins. The produced information could harbor potential proteomic biomarkers of SCZ, contributing to individualized prognosis and stratification strategies, besides aiding in the differential diagnosis. 相似文献

Nowadays, people do not only navigate the web, but they also contribute contents to the Internet. Among other things, they write their thoughts and opinions in review sites, forums, social networks, blogs and other websites. These opinions constitute a valuable resource for businesses, governments and consumers. In the last years, some researchers have proposed opinion extraction systems, mostly domain-independent ones, to automatically extract structured representations of opinions contained in those texts. In this work, we tackle this task in a domain-oriented approach, defining a set of domain-specific resources which capture valuable knowledge about how people express opinions on a given domain. These resources are automatically induced from a set of annotated documents. Some experiments were carried out on three different domains (user-generated reviews of headphones, hotels and cars), comparing our approach to other state-of-the-art, domain-independent techniques. The results confirm the importance of the domain in order to build accurate opinion extraction systems. Some experiments on the influence of the dataset size and an example of aggregation and visualization of the extracted opinions are also shown. 相似文献

Extracts obtained by supercritical carbon dioxide extraction of cones and leaves of hop (Humulus lupulus L) at different combinations of temperature and pressure were analysed for their α- and β-acids and volatiles, using HPLC and GC respectively. The yield and composition of the mixtures of bitter compounds from the cones were largely influenced by the temperature and pressure applied during the extraction. Bitter compounds could not be detected in the extracts from the leaves. The most important volatile components identified were β-myrcene, β-caryophyllene and α-humulene. The extraction parameters also influenced the composition of the mixture of volatiles from the cones and leaves, 40°C and 20.0 MPa being optimum for the extraction of both bitter compounds and volatiles, which are thought to be important for the aroma of beer. 相似文献

Management in wireless backhaul networks is a challenging task, especially in rural and isolated environments. In these scenarios, the backhaul network usually consists of a set of heterogeneous wireless links that provide limited and variable bandwidth to the access networks, often 3G/4G small cells. Because of the highly constrained nature of this type of backhaul network, intelligent and joint management in both the backhaul network and the access network is crucial in order to avoid performance degradation caused by traffic congestion. In order to avoid the saturation in the backhaul network, access networks should consider the backhaul state when taking decisions in the admission control and scheduling procedures. However, no standardized mechanisms currently exist for sharing management information between both networks. In this work, we propose to use the Explicit Congestion Notification (ECN) bit in the outer IP headers present in the Iuh 3GPP IPSec‐enabled interfaces in order to notify the backhaul congestion state to the access network. We analyze for the referred scenario, compatibility and security details, validating our approach by running numerically simulations and implementing the notification mechanism. Our low complexity approach offers 2% accuracy and backhaul update latency lower than 10 ms during 80% of the time, which makes the solution appropriate for admission control and scheduling intervals in small cells. 相似文献

Previously, we presented automated methods for thoraco-abdominal asynchrony estimation and movement artifact detection in respiratory inductance plethysmography (RIP) signals. This paper combines and improves these methods to give a method for the automated, off-line detection of pause, movement artifact, and asynchrony. Simulation studies demonstrated that the new combined method is accurate and robust in the presence of noise. The new procedure was successfully applied to cardiorespiratory signals acquired postoperatively from infants in the recovery room. A comparison of the events detected with the automated method to those visually scored by an expert clinician demonstrated a higher agreement (κ = 0.52) than that amongst several human scorers (κ = 0.31) in a clinical study . The method provides the following advantages: first, it is fully automated; second, it is more efficient than visual scoring; third, the analysis is repeatable and standardized; fourth, it provides greater agreement with an expert scorer compared to the agreement between trained scorers; fifth, it is amenable to online detection; and lastly, it is applicable to uncalibrated RIP signals. The cement industry is the third largest carbon emitting industrial sector in the EU. This article presents the foreseeable technological evolution of the cement industry under business as usual circumstances, and examines the effects on the sector of carbon trading. For those purposes a global dynamic simulation model of the cement industry (CEMSIM) has been developed. The model is composed of a series of interconnected modules on cement consumption and production, international trade and capacity planning. This study quantifies the benefits achieved from emission trading in different markets (EU15, EU27 and Annex B), derived both from the revenues of emission trading, and from the lower compliance costs.
