Resilin is a rubber-like protein found in the exoskeleton of arthropods. It often contributes large proportions to the material of certain structures in movement systems. Accordingly, the knowledge of the presence and distribution of resilin is essential for the understanding of the functional morphology of these systems. Because of its specific autofluorescence, resilin can be effectively visualized using fluorescence microscopy. However, the respective excitation maximum is in the UV range, which is not covered by the lasers available in most of the modern commercial confocal laser scanning microscopes. The goal of this study was to test the potential of confocal laser scanning microscopy (CLSM) in combination with a 405 nm laser to visualize and analyse the presence and distribution of resilin in arthropod exoskeletons. The results clearly show that all resilin-dominated structures, which were visualized successfully using wide-field fluorescence microscopy (WFM) and a 'classical' UV excitation, could also be visualized efficiently with the proposed CLSM method. Furthermore, with the application of additional laser lines CLSM turned out to be very appropriate for studying differences in the material composition within arthropod exoskeletons in great detail. As CLSM has several advantages over WFM with respect to detailed morphological imaging, the application of the proposed CLSM method may reveal new information about the micromorphology and material composition of resilin-dominated exoskeleton structures leading to new insights into the functional morphology and biomechanics of arthropods.  相似文献

Amyotrophic lateral sclerosis (ALS) is an incurable and lethal neurodegenerative disease in which progressive motor neuron loss and associated inflammation represent major pathology hallmarks. Both the prevention of neuronal loss and neuro-destructive inflammation are still unmet challenges. Medical ozone, an ozonized oxygen mixture (O3/O2), has been shown to elicit profound immunomodulatory effects in peripheral organs, and beneficial effects in the aging brain. We investigated, in a preclinical drug testing approach, the therapeutic potential of a five-day O3/O2 i.p. treatment regime at the beginning of the symptomatic disease phase in the superoxide dismutase (SOD1G93A) ALS mouse model. Clinical assessment of SOD1G93A mice revealed no benefit of medical ozone treatment over sham with respect to gross body weight, motor performance, disease duration, or survival. In the brainstem of end stage SOD1G93A mice, however, neurodegeneration was found decelerated, and SOD1-related vacuolization was reduced in the motor trigeminal nucleus in the O3/O2 treatment group when compared to sham-treated mice. In addition, microglia proliferation was less pronounced in the brainstem, while the hypertrophy of astroglia remained largely unaffected. Finally, monocyte numbers were reduced in the blood, spleen, and mesenteric lymph nodes at postnatal day 60 in SOD1G93A mice. A further decrease in monocyte numbers seen in mesenteric lymph nodes from sham-treated SOD1G93A mice at an advanced disease stage, however, was prevented by medical ozone treatment. Collectively, our study revealed a select neuroprotective and possibly anti-inflammatory capacity for medical ozone when applied as a therapeutic agent in SOD1G93A ALS mice.  相似文献

The environmental and health risks posed by emerging engineered nanoparticles (ENPs) released into aquatic environments are largely dependent on their aggregation, transport, and deposition behavior. Herein, laboratory-scale columns were used to examine the mobility of polyacrylic acid (PAA)-coated cerium dioxide nanoparticles (nCeO2) and an analogous nanosized polymeric capsule (nCAP) in water saturated quartz sand or loamy sand. The influence of solution ionic strength (IS) and cation type (Na+, Ca2+, or Mg2+) on the transport potential of these ENPs was examined in both granular matrices and results were also compared to measurements obtained using a natural groundwater. ENP suspensions were characterized using dynamic light scattering and nanoparticle tracking analysis to establish aggregate size, and laser Doppler electrophoresis to determine ENP electrophoretic mobility. Regardless of IS, virtually all nCeO2 particles suspended in NaNO3 eluted from the quartz sand-packed columns. In contrast, heightened nCeO2 and nCAP particle retention and dynamic (time-dependent) transport behavior was observed with increasing concentrations of the divalent salts and in the presence of natural groundwater. Enhanced particle retention was also observed in loamy sand in comparison to the quartz sand, emphasizing the need to consider the nature of the aqueous matrix and granular medium in evaluating contamination risks associated with the release of ENPs in natural and engineered aquatic environments.  相似文献

Carleton University’s experimental atrium and tunnel facilities share a fan chamber and three large exhaust fans. Using oxygen consumption calorimetry, the Heat Release Rates (HRR) of fires in either of these facilities can be calculated. This paper focuses on the design of the instrumentation in the fan chamber, which was carried out using the Fire Dynamics Simulator (FDS) and manual velocity measurements. Due to a high amount of mixing and turbulence and a long travel distance, the temperature and gas concentration profiles are relatively uniform. However, for the same reason, the velocity profile had to be looked at very closely to find an optimum combination of bi-directional probes. The analysis indicated that acceptable HRR measurements can be obtained using an array of six thermocouples, four bi-directional velocity probes and a gas sampling grid. Results from the preliminary tests are presented. The system shows a reasonable estimate of the HRR as compared to the propane calibration burner.  相似文献

We report on the rheological properties of semidilute carbon nanotube (CNT)/epoxy suspensions with varying filler content prepared with different dispersion methods. We show that dispersion techniques can be distinguished by means of the rheological response. It was found that the suspensions undergo a lasting dispersion changes during the first shear sweep measurement. Shear‐induced changes within the suspension can also be visualized optically. Our results demonstrate that the rheological properties are strongly influenced by the measurement procedure, which has to be known in order to guarantee reproducible and comparable results. Furan is formed in a variety of heat‐treated foods through thermal degradation of natural food constituents. Relatively high levels of furan contamination are found in ground roasted coffee, instant coffee, and processed baby foods. European exposure estimates suggest that mean dietary exposure to furan may be as high as 1.23 and 1.01 μg/kg bw/day for adults and 3‐ to 12‐month‐old infants, respectively. Furan is a potent hepatotoxin and hepatocarcinogen in rodents, causing hepatocellular adenomas and carcinomas in rats and mice, and high incidences of cholangiocarcinomas in rats at doses ≥2 mg/kg bw. There is therefore a relatively low margin of exposure between estimated human exposure and doses that cause a high tumor incidence in rodents. Since a genotoxic mode of action cannot be excluded for furan‐induced tumor formation, the present exposures may indicate a risk to human health and need for mitigation. This review summarizes the current knowledge on mechanisms of furan formation in food, human dietary exposure to furan, and furan toxicity, and highlights the need to establish the risk resulting from the genotoxic and carcinogenic properties of furan at doses lower than 2 mg/kg bw.  相似文献

In close relationships, the association between negative beliefs about a partner and loving was found to be moderated by how that negative knowledge was organized. In general, evaluatively integrative organization (i.e., categorizing positive and negative beliefs together) was associated with more positive thoughts, feelings, and behaviors toward the partner when the negative content of beliefs was high and relationships were relatively long. Additional evidence suggests that compartmentalization (i.e., organizing positive and negative beliefs in separate categories) may be an effective strategy for handling negative beliefs about a partner in a new relationship. Findings for behavioral closeness raise the possibility that the nature of shared activities, as well as an individual's cognitive processes, may influence how knowledge about a relationship partner is organized. Intergenomic evolutionary conflicts increase biological diversity. In sexual conflict, female defence against males is generally assumed to be resistance, which, however, often leads to trait exaggeration but not diversification. Here, we address whether tolerance, a female defence mechanism known from interspecific conflicts, exists in sexual conflict. We examined the traumatic insemination of female bed bugs via cuticle penetration by males, a textbook example of sexual conflict. Confocal laser scanning microscopy revealed large proportions of the soft and elastic protein resilin in the cuticle of the spermalege, the female defence organ. Reduced tissue damage and haemolymph loss were identified as adaptive female benefits from resilin. These did not arise from resistance because microindentation showed that the penetration force necessary to breach the cuticle was significantly lower at the resilin-rich spermalege than at other cuticle sites. Furthermore, a male survival analysis indicated that the spermalege did not impose antagonistic selection on males. Our findings suggest that the specific spermalege material composition evolved to tolerate the traumatic cuticle penetration. They demonstrate the importance of tolerance in sexual conflict and genitalia evolution, extend fundamental coevolution and speciation models and contribute to explaining the evolution of complexity. Establishing biological synthesis of nanoparticles is increasing nowadays in the field of nanotechnology. The search for an optimal source with durability, stability, capacity to withstand higher environmental conditions with excellent characteristics is yet to meet. Consequently, there is need to create an eco‐friendly strategy for metal nanoparticle synthesis. One approach investigated in this review is the use of biosurfactants to enhance the synthesis biologically. In comparison with the other technologies, biosurfactants are less toxic and exhibit higher properties. This method is different from the conventional practice like physical and chemical methods. Several research studies represented that the biosurfactant influences the production of nanoparticles about 2–50 nm. In this manner, the research towards the biosurfactant has raised. This review also addressed the feasibility of biosurfactant and their benefits in the synthesis of metallic nanoparticles.
