São João cheeses with varying curd dry salting treatments were made with decreasing levels of salt (sodium chloride): 4 (control), 3, 2 and 1% (w/w), along with the salt‐free version. The cheeses were ripened at 11 °C over a 40‐day period, and the effect of lowering salt on the physicochemical, microbiological and sensory properties of the cheese was studied. Reduced salt resulted in a concomitant moisture decrease with protein increase, ash and sodium reduction among experimental cheeses at the same ripening day, but there were no significant differences in pH, acidity and fat, or in the microbiological quality. Triangle tests indicated perceptible differences between test and control cheeses at the level of 2% NaCl (w/w) or less, but not with the cheese salted with 3 g NaCl/100 g. Considering the sensory, the physicochemical and the microbiological results, the cheese formulated at 3% NaCl (w/w) (presenting a reduction of 25% in salt) is feasible on an industrial scale, being indistinguishable by the regular consumer.  相似文献

Smart thin coatings using a recombinant elastin‐like polymer (ELP) containing the cell attachment sequence arginine–glycine–(aspartic acid) (RGD) are fabricated for the first time through simple deposition of the ELP dissolved in aqueous‐based solutions. The biopolymer is produced and characterized using electrophoresis and mass spectroscopy. The temperature and pH responsiveness are assessed by aggregate size measurements and differential scanning calorimetry. The deposition of the studied ELP onto chitosan is followed in situ with a quartz‐crystal microbalance with dissipation monitoring (QCM‐D). Contact angle measurements are performed at room temperature and at 50 °C, showing reversible changes from a moderate hydrophobic behavior to an extremely wettable surface. AFM analysis performed at room temperature reveals a smooth surface and no organized structure. At 50 °C, the surface presents spherical nanometer‐sized structures of collapsed biopolymer chains. Such results suggest that the ELP chains, when collapsed, aggregate into micelle‐like structures at the surface of the substrate, increasing its water affinity. Cell adhesion tests on the developed coatings are conducted using a SaOS‐2 cell line. Enhanced cell adhesion could be observed in the H‐RGD6‐coated surfaces, as compared with the original chitosan monolayer. An intermediate behavior is found in chitosan coated with the corresponding ELP without the RGD sequence. Therefore, the developed films have great potential as biomimetic coatings of biomaterials for different biomedical applications, including tissue engineering and controlled delivery of bioactive agents. Their thermo‐responsive behavior can also be exploited for tunable cell adhesion and controlled protein adsorption.  相似文献

A pigment network is one of the most important dermoscopic structures. This paper describes an automatic system that performs its detection in dermoscopy images. The proposed system involves a set of sequential steps. First, a preprocessing algorithm is applied to the dermoscopy image. Then, a bank of directional filters and a connected component analysis are used in order to detect the "lines" of the pigment network. Finally, features are extracted from the detected network and used to train an AdaBoost algorithm to classify each lesion regarding the presence of the pigment network. The algorithm was tested on a dataset of 200 medically annotated images from the database of Hospital Pedro Hispano (Matosinhos), achieving a sensitivity = 91.1% and a specificity = 82.1%.  相似文献

Moscatel wines from Setúbal were analyzed for their total phenolic (mean value 1,243 mg gallic acid equivalents/L), and total flavonoid (mean value 248 mg catechin/L) composition by spectrophotometric and chromatographic methods were used to quantify phenolic compounds as benzoic acids, cinnamic acids, stilbens, and some flavonoids. Antioxidant activity of the wines was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH; mean value 70.7% inhibition), ferric reducing antioxidant power (FRAP; mean value 3,098 mg of Trolox equivalents/L) and oxygen radical absorbance capacity (ORAC; mean value 10,724 μmol/L) assays. Results were analyzed using principal component analysis which allowed samples to be grouped in terms of both winemaking producer and vintage. By plotting correlation loadings, it was possible to understand which variables were responsible for sample distribution. The correlation between results obtained for variables show that, in general, total flavonoid content is a better estimation of antioxidant activity in Moscatel samples (r ORAC/flavonoids = 0.832, r FRAP/flavonoids = 0.677) than total phenolic content (r ORAC/phenolics = 0.680, r FRAP/phenolics = 0.372). No major correlations were detected for DPPH assay results (r DPPH/flavonoids = 0.283, r DPPH/phenolics = 0.271). Chromatographic profiles showed important differences among Moscatel wines. Gallic acid contents and results of antioxidant activity tests were strongly correlated (r values in the range 0.74–0.92). Correlations of the results obtained for antioxidant activity tests with contents of other phenolic compounds such as ethyl caffeate, ethyl gallate, caffeic acid, protocatechuic acid, and t-caftaric acid depend on sample and type of test employed. Intervertebral disc (IVD) degeneration involves a complex cascade of events, including degradation of the native extracellular matrix, loss of water content, and decreased cell numbers. Cell recruitment strategies for the IVD have been increasingly explored, aiming to recruit either endogenous or transplanted cells. This study evaluates the IVD therapeutic potential of a chemoattractant delivery system (HAPSDF5) that combines a hyaluronan-based thermoreversible hydrogel (HAP) and the chemokine stromal cell derived factor-1 (SDF-1). HAPSDF5 was injected into the IVD and was combined with an intravenous injection of mesenchymal stem/stromal cells (MSCs) in a pre-clinical in vivo IVD lesion model. The local and systemic effects were evaluated two weeks after treatment. The hydrogel by itself (HAP) did not elicit any adverse effect, showing potential to be administrated by intradiscal injection. HAPSDF5 induced higher cell numbers, but no evidence of IVD regeneration was observed. MSCs systemic injection seemed to exert a role in IVD regeneration to some extent through a paracrine effect, but no synergies were observed when HAPSDF5 was combined with MSCs. Overall, this study shows that although the injection of chemoattractant hydrogels and MSC recruitment are feasible approaches for IVD, IVD regeneration using this strategy needs to be further explored before successful clinical translation.  相似文献

Background and Aims

The history of Vitis vinifera L. cv. Verdelho has been studied, however, its geographic origin remains unknown. Although not widespread in mainland Portugal, Verdelho has gained a reputation as a Madeira wine, which has led most authors to attribute its origin to the Madeira Islands. The objective of this work was to determine the origin of Verdelho.

Methods and Results

An integrated strategy was followed, combining the molecular analysis of 28 nuclear and three chloroplast DNA microsatellites with an historical literature search. Historical records of the presence of Verdelho in the Azores Islands were found and these preceded those of the Madeira Islands by about 200 years. Moreover, the genetic analysis supports Verdelho as the progenitor of the Azores cultivars, Arinto dos Açores and Terrantez do Pico, and as the progeny of cv. Savagnin.

Conclusions

Verdelho is a key cultivar in the Azorean genetic diversity pool, sharing first‐degree relationships with the two most important autochthonous grape cultivars, and reinforcing the historical data of its ancestry in the region.

Significance of the Study

The results presented here, both historical and genetic, shed new light on the origin of Verdelho, and suggest that the Azores Islands is the site where this cultivar was likely to have originated.  相似文献

Fast-scan cyclic voltammetry (FSCV) with carbon-fiber microelectrodes has been successfully used to detect catecholamine release in vivo. Generally, waveforms with anodic voltage limits of 1.0 or 1.3 V (vs Ag/AgCl) are used for detection. The 1.0 V excursion provides good temporal resolution but suffers from a lack of sensitivity. The 1.3 V excursion increases sensitivity but also increases response time, which can blur the detection of neurochemical events. Here, the scan rate was increased to improve the sensitivity of the 1.0 V excursion while maintaining the rapid temporal response. However, increasing scan rate increases both the desired faradaic current response and the already large charging current associated with the voltage sweep. Analog background subtraction was used to prevent the analog-to-digital converter from saturating from the high currents generated with increasing scan rate by neutralizing some of the charging current. In vitro results with the 1.0 V waveform showed approximately a 4-fold increase in signal-to-noise ratio with maintenance of the desired faster response time by increasing scan rate up to 2400 V/s. In vivo, stable stimulated release was detected with an approximate 4-fold increase in peak current. The scan rate of the 1.3 V waveform was also increased, but the signal was unstable with time in vitro and in vivo. Adapting the 1.3 V triangular wave into a sawhorse design prevented signal decay and increased the faradaic response. The use of the 1.3 V sawhorse waveform decreased the detection limit of dopamine with FSCV to 0.96 ± 0.08 nM in vitro and showed improved performance in vivo without affecting the neuronal environment. Electron microscopy showed dopamine sensitivity is in a quasi-steady state with carbon-fiber microelectrodes scanned to potentials above 1.0 V.  相似文献
