A method for the production of bovine colostral F(ab′)2 fragments at pilot-plant scale was developed. Optimum yield of immunoglobulins in colostral whey was obtained after rennet treatment of first milking colostrum. Pepsin digestion was carried out directly on the colostral whey at pH 3·8 for 4 h, which led to the complete digestion of immunoglobulins into F(ab′)2 fragments. Elimination of β-lactoglobulin, the main immunogenic protein, was achieved by anion exchange on Duolite A560 at pH 6·0. The preparation was diafiltered with a 5000 Da membrane and the retentate spray-dried. The powder obtained contained approximately 34% F(ab′)2 fragments, with an antibody activity three times higher than the initial colostrum.  相似文献

A simple, fast and accurate Fourier transform mid-infrared spectroscopy method was developed for simultaneously determining sugar and organic acid contents in apricot fruit slurries using the attenuated total reflectance. The potential of this method coupled with chemometric techniques based on partial least squares was assessed by comparison with currently used enzymatic determination of sucrose, glucose, fructose, malic acid and citric acid. Fruits of eight contrasted cultivars harvested at different ripening stages were used in this study and randomly divided in a calibration set (505 apricots) and in a validation set (252 apricots). The most suitable region was found in the range between 1500 and 900 cm−1. Good prediction performances were obtained (R2 ? 0.74 and RMSEP ? 18%). Results concerning the prediction of other quality traits such as firmness, skin colour, ethylene production, soluble solids content and titratable acidity were discussed.  相似文献

Listeria monocytogenes can enter the food chain at virtually any point. However, food processing environments seem to be of particular importance. From an ecological point of view, food processing facilities are microbial habitats that are constantly disturbed by cleaning and sanitizing procedures. Although L. monocytogenes is considered ubiquitous in nature, it is important to recognize that not all L. monocytogenes strains appear to be equally distributed; the distribution of the organism seems to be related to certain habitats. Currently, no direct evidence exists that L. monocytogenes-associated biofilms have played a role in food contamination or foodborne outbreaks, likely because biofilm isolation and identification are not part of an outbreak investigation, or the definition of biofilm is unclear. Because L. monocytogenes is known to colonize surfaces, we suggest that contamination patterns may be studied in the context of how biofilm formation is influenced by the environment within food processing facilities. In this review, direct and indirect epidemiological and phenotypic evidence of lineage-related biofilm formation capacity to specific ecological niches will be discussed. A critical view on the development of the biofilm concept, focused on the practical implications, strengths, and weaknesses of the current definitions also is discussed. The idea that biofilm formation may be an alternative surrogate for microbial fitness is proposed. Furthermore, current research on the influence of environmental factors on biofilm formation is discussed.  相似文献

Milkfat was separated into major chainlength fractions by solid-phase extraction. The effect on thermal behavior and texture of replacing both saturated and monounsaturated long-chain triglycerides from milkfat by long-chain monounsaturated triglycerides with an unsaturated fatty acid in thesn-2 position is reported. Increasing proportions of cocoa butter were added to fractions of short-to medium-chain triglycerides (C22−C44) and medium- to long-chain triglycerides (C36−C48) isolated from milkfat. Thermal behavior and texture of the mixtures were measured. Results indicated that long-chain monounsaturated triglycerides from cocoa butter enhanced co-crystallization and co-operative melting and did not induce polymorphic transitions upon crystallization and melting of the fractions. At 4°C, they acted as texture builder if present in proportions of more than 30%, whereas below this level, they acted as texture softeners. The effect of the long-chain monounsaturated triglycerides on the texture of fractions that melt at low temperature could not be predicted from the proportion of solid fat at that temperature. Individual tubers from mature plants of cultivars Pentland Dell and Estima were harvested from experimental plots in two successive years and analysed for α‐solanine, α‐chaconine and total glycoalkaloid concentration (α‐solanine and α‐chaconine combined) using high‐performance liquid chromatography. Mean tuber glycoalkaloid concentration per plant was strongly affected by genotype and was much higher in plants of Pentland Dell (10.0 and 16.3 mg per 100 g fresh weight in 1994 and 1995, respectively) than Estima (4.0 and 4.3 mg per 100 g fresh weight in 1994 and 1995, respectively). The variation between plants in mean tuber glycoalkaloid concentration, expressed as coefficient of variation, was similar for the two cultivars in both years. Variation within plants was higher for cultivar Pentland Dell than Estima for both years but it was statistically significant only in 1995. Tuber position within a plant had no effect on glycoalkaloid concentrations in tubers of similar size. An inverse relationship between total glycoalkaloid concentration and tuber fresh weight of individual tubers was found for both cultivars and small tubers of Pentland Dell (20–40 g fresh weight) exceeded the safety limit of 20 mg per 100 g fresh weight. The pattern of glycoalkaloid accumulation differed between genotypes and appeared related to tuber growth. In a highly competitive market, reliable techniques for manufacturing quality control of electronic devices are demanded. Characterisation of modern microelectronic package integrity becomes more difficult due to the continued miniaturisation of electronic device and the complexity of advanced micro-assembling technologies such as chip-scale packages and 3D IC stacks. In this paper, sparse representations of acoustic signals are sought to improve the scanning acoustic microscopy (SAM), a common non-destructive tool for failure analysis of microelectronic packages. Sparse representation of an ultrasonic signal is obtained by decomposing it in an overcomplete dictionary. Detection and location of ultrasonic echoes are then performed on the basis of the resulting redundant representation. The method offers a solution to the deconvolution problem for restoration of the ultrasonic reflectivity function. It can restore closely space overlapping echoes beyond the resolution of the conventional SAM system. It also produces high resolution and accurate estimates for ultrasonic echo parameters, i.e., time-of-flight, amplitude, centre frequency, and bandwidth. These merits of the proposed method are explored in various potential applications for microelectronic package characterisation.  相似文献

The top‐down, micromolding technique, referred to as Particle Replication in Nonwetting Templates (PRINT®), affords a new opportunity for the generation of inhalation therapeutics. Powders were fabricated with predetermined particle size and shape; when dispersed with a collision jet nebulizer, these particles resulted in monodisperse aerosols with geometric standard deviations well below 1.2. Dynamic shape factors for this novel set of uniformly shaped particles were determined by correcting the drag of nonspherical particles in the ultra‐Stokesian flow conditions of the aerodynamic particle sizer (APS). This convenient approach for shape factor determination agreed well with current literature approaches and allowed for the correction of APS results for particles with known volumes. Determined shape factor values of PRINT geometries were used to estimate the theoretical median aerodynamic diameters of individual aerosols, which were then compared to actual inhalation powders. © 2013 American Institute of Chemical Engineers AIChE J, 59: 3184–3194, 2013  相似文献

Compounds that simultaneously activate peroxisome proliferator‐activated receptor (PPAR) subtypes α and γ have the potential to effectively treat dyslipidemia and type 2 diabetes (T2D) in a single pharmaceutically active molecule. The frequently observed side effects of selective PPARγ agonists, such as edema and weight gain, were expected to be overcome by using additive PPARα activity, leading to dual PPARα/γ agonists with balanced activity for both subtypes. Herein we report the discovery, synthesis, and optimization of a new series of α‐ethoxyphenylpropionic acid bearing 5‐ or 6‐substituted indoles. The incorporation of oxime ethers on the carbonyl portion of the benzoyl group can bring the PPARα/γ potency ratio equal to or slightly greater than one, as is the case for compounds 20 c and 21 a . Compound 20 c shows high efficacy in an ob/ob mouse model of T2D and dyslipidemia, similar to that of rosiglitazone and tesaglitazar, but with a significant increase in body weight gain. In contrast, compound 21 a , less potent as a dual PPARα/γ activator than 20 c , showed an interesting pharmacological profile, as it elicits a decrease in body weight relative to reference compounds.  相似文献

The aim of this study was to investigate soy protein recovery feasibility after lab- and pilot-plant scale oleosome isolation. The proteins were isolated by isoelectric precipitation and by ultrafiltration. The functional properties of the recovered proteins were compared to soy protein isolate produced in our laboratory. The residual lipid content in the aqueous supernatant affected the protein recovery yields and purities. Ethanol precipitation and ultrafiltration resulted in the best protein yields, which were 25 and 26% greater than protein yield obtained by isoelectric precipitation with distilled water dilution. The protein content of the isoelectric precipitated pilot-plant supernatant was higher (98%) than the protein content of ethanol-precipitated proteins (80%). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed similar peptide profiles for laboratory and pilot-plant supernatants. Protein solubility curves between pH 3 and 8 were typical for soy protein isolate with higher solubilities for proteins obtained from pilot-plant supernatant. The soy protein isolate and ethanol-precipitated protein had the highest emulsification capacity on a dry-weight basis. These desirable functional properties of proteins recovered as co-products after oleosome isolation suggest they are highly suitable for industrial application as food ingredients and their recovery would contribute to the economics of the overall oleosome fractionation process.  相似文献
