The success of cell therapy for the treatment of myocardial infarction depends on finding novel approaches that can substantially implement the engraftment of the transplanted cells. In order to enhance cell engraftment, most studies have focused on the pretreatment of transplantable cells. Here we have considered an alternative approach that involves the preconditioning of infarcted heart tissue to reduce endogenous cell activity and thus provide an advantage to our exogenous cells. This treatment is routinely used in other tissues such as bone marrow and skeletal muscle to improve cell engraftment, but it has never been taken in cardiac tissue. To avoid long-term cardiotoxicity induced by full heart irradiation we developed a rat model of a catheter-based heart irradiation system to locally impact a delimited region of the infarcted cardiac tissue. As proof of concept, we transferred ZsGreen+ iPSCs in the infarcted heart, due to their ease of use and detection. We found a very significant increase in cell engraftment in preirradiated rats. In this study, we demonstrate for the first time that preconditioning the infarcted cardiac tissue with local irradiation can substantially enhance cell engraftment. 相似文献

The Smoothened (SMO) receptor is the most druggable target in the Hedgehog (HH) pathway for anticancer compounds. However, SMO antagonists such as vismodegib rapidly develop drug resistance. In this study, new SMO antagonists having the versatile purine ring as a scaffold were designed, synthesised, and biologically tested to provide an insight to their mechanism of action. Compound 4s was the most active and the best inhibitor of cell growth and selectively cytotoxic to cancer cells. 4s induced cell cycle arrest, apoptosis, a reduction in colony formation and downregulation of PTCH and GLI1 expression. BODIPY-cyclopamine displacement assays confirmed 4s is a SMO antagonist. In vivo, 4s strongly inhibited tumour relapse and metastasis of melanoma cells in mice. In vitro, 4s was more efficient than vismodegib to induce apoptosis in human cancer cells and that might be attributed to its dual ability to function as a SMO antagonist and apoptosis inducer. 相似文献

Linear dynamic viscoelastic properties have been used to evaluate the influence of heat processing on the microstructure of crayfish protein isolate (CFPI) in order to explore the potentials of crayfish in the production of surimi-like gel products. CFPI dispersions have been subjected to a temperature cycle that consisted of a constant heating rate temperature ramp and a rapid cooling step, following the transitions taking place in the system through the evolution of G′ and G″, under different CFPI concentrations and pH values. The influence of CFPI concentration and pH on linear viscoelasticity functions of CFPI aqueous systems before and after thermal processing has also been analysed. Occurrence of a gel-like behaviour has been found for CFPI dispersions. The mechanical spectra of CFPI gels have revealed a remarkable enhancement in gel strength by thermal processing. An apparent gel network enhancement has also been found by increasing the CFPI content or reducing the pH, excepting at the isoelectric point. The strong dependence of microstructure on pH found for thermally processed CFPI gels has been confirmed by Electron Microscopy. 相似文献

Low density polyethylene (LDPE) is a widely used thermoplastic. The dispersion of inorganic fillers in thermoplastic matrices such as polyethylene has been largely employed to improve some of its properties. However, interaction between both components is a major issue so the presence of a coupling agent is usually necessary to increase the interaction among the phases. In this study, LDPE chemically modified with vinyltriethoxysilane (VTES) was used as a coupling agent in glass fiber-reinforced LDPE. The composites were prepared in a mixing chamber and subsequently analyzed by tensile tests, rotational rheometry, and scanning electron microscopy (SEM). The mechanical properties were significantly increased by the use of small amounts of the coupling agent. Moreover, the rheological behavior and the SEM micrographs showed higher interaction between the matrix and the reinforcing phase in the composites containing LDPE modified with VTES, confirming the suitability of using this coupling agent in these systems. Thiocoraline is a thiodepsipeptide antitumor compound produced by two actinomycetes Micromonospora sp. ACM2-092 and Micromonospora sp. ML1, isolated from two marine invertebrates (a soft coral and a mollusc) found of the Indian Ocean coast of Mozambique. By using oligoprimers derived from nonribosomal peptide synthetase (NRPS) consensus sequences, six PCR fragments containing putative NRPS adenylation domains were amplified from the chromosome of Micromonospora sp. ML1. Insertional inactivation of each adenylation domain showed that two of them generated nonproducing mutants, thereby indicating that these domains were involved in thiocoraline biosynthesis. Sequencing of a 64.6 kbp DNA region revealed the presence of 36 complete open reading frames (ORFs) and two incomplete ones. Heterologous expression of a region of about 53 kbp, containing 26 of the ORFs, in Streptomyces albus and S. lividans led to the production of thiocoraline in these streptomycetes. Surprisingly, the identified gene cluster contains more NRPS modules than expected on the basis of the number of amino acids of thiocoraline. TioR and TioS would most probably constitute the NRPS involved in the biosynthesis of the thiocoraline backbone, according to the colinearity of the respective modules. It is proposed that two other NRPSs, TioY and TioZ, could be responsible for the biosynthesis of a small peptide molecule which could be involved in regulation of the biosynthesis of thicoraline in Micromonospora sp. ML1. In addition, a pathway is proposed for the biosynthesis of the unusual starter unit, 3-hydroxy-quinaldic acid. 相似文献

The effect of different operational strategies and inoculum structure (granules and disaggregated granules) during the start-up of four up-flow anaerobic sludge blanket hydrogenogenic reactors was investigated. The more stable volumetric hydrogen production rates obtained were 0.38 and 0.36 L H2/L-d, in reactors operated with a constant organic loading rate (OLR) with both inoculum structures, whereas in reactors operated with an increasing OLR methane started to be produced earliest in time. Specific hydrogenogenic activity results proved that the disaggregated inoculum produced a more active biomass than the granular one, but not granule formation was evident. The methane hydrogenotrophic activity was the main limitation of the systems evaluated. In the reactors inoculated with disaggregated sludge the start-up strategy did not influence the bacterial DGGE fingerprint, in contrast to the reactors started-up with granular sludge; members of the Clostridium genus were always present. The results demonstrated that operational conditions during the start-up period are crucial for the production of hydrogenogenic biomass. 相似文献

This work presents experimental performance results for a 50 cm2 Polymer Electrolyte Membrane (PEM) Fuel Cell, including polarization curves and Electrochemical Impedance Spectroscopy (EIS) analysis of the Fuel Cell. EIS results were used for the determination of the cell ohmic resistance as well as charge transfer resistances under different operating conditions. Different combinations of operating conditions and bipolar plate designs were analysed. In particular, the effect of the cathode oxygen concentration, reactant gases humidification, and bipolar plate (BP) design were assessed. Butler–Volmer (BV) kinetic parameters such as the charge transfer coefficient were also determined from Tafel plots. The electronic contact resistances were measured for both Bipolar Plate designs, and the membrane protonic resistances were calculated. Its dependence on the BP flow field design and operating conditions is addressed. The results obtained in this work are aimed both at gaining insight into the fundamental processes determining the fuel cell performance, and at determining parameters needed for Computational Fuel Cell Dynamics (CFCD) numerical simulations. 相似文献

The alternative technologies discussed in this review are able to eliminate pathogenic bacteria while retaining bioactive compounds such as anthocyanins, phenols, pigments, and vitamins, improve the extraction processes of fruit or vegetable juices. The use of nonthermal processing technologies in beverages might provide a higher quality value to the final product and consumers tended to prefer more natural products with minimum preservative substances. This review is analyze and discuss the advantages and disadvantages of alternative technologies applied in beverages, evaluating the parameters that can optimize beverage processing such as time, temperature, pressure, frequency, tension, among others. Additionally, finding solutions to beverage processing issues will be considered analyzing the results obtained with other applied methods, and an overview of new strategies such as microfiltration or the combination of two alternative technologies to improve the quality of the final product will also be evaluated.
Practical applications
The applications of alternative technologies mentioned in this article, are a feasible option for the preservation of beverages in the area of industry. The capacity of each of them has been demonstrated, in addition to the use of microfiltration and ultrafiltration, but the combination between them sounds very encouraging to achieve the quality and safety of beverages, with an extended shelf life and greater protection of its main components. Corynebacterium bovis is a mastitis-causing microorganism responsible for economic losses related to decrease in milk production. The aim of the study was identify Corynebacterium spp. strains recovered from milk samples of subclinical mastitis by using matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). Samples were collected during a 10-mo mastitis-monitoring program in a high-production dairy farm. In this study, 80 strains were analyzed; from these 54 (67.5%) were identified at species level as Corynebacterium bovis, 24 (31.2%) isolates were identified at the genus level as Corynebacterium spp., and only 1 (1.35%) isolated had unreliable identification. Results demonstrated that MALDI-MS could be an important technique for the identification of Corynebacterium spp. in milk. 相似文献
