Parthenium (Parthenium hysterophorus L.) leaf residue (LP, leaf powder) inhibited salvinia (Salvinia molesta Mitchell) biomass and the number of healthy fronds at 0.25% (w/v) and killed the treated plants at and above 0.75% (w/v) in about 5–15 days, depending on the quantity of the residue. At the lethal dose, the LP caused an abrupt desiccation of above-water plant parts, probably due mainly to root dysfunction. This was concurrent with the loss of dehydrogenase activity in, and an increase in solute leakage from, the roots and loss of chlorophylla, b, and total chlorophyll contents in the fronds, resulting in death of the treated plants. The LP appears inhibitory to salvinia through affecting macromolecules—proteins, lipids, and nucleic acids. The inhibitory activity of LP at the lethal dose suspended in water was completely lost when allowed to stand for 30 days under outdoor conditions and promoted growth of the salvinia plants placed in it. The standard allelochemicals, including those present in parthenium LP, except parthenin andp-hydroxybenzoic acid, did not inhibit growth up to 100 ppm. However, parthenin andp-hydroxybenzoic acid killed salvinia plants at 100 and 50 ppm, respectively. Sincep-hydroxybenzoic acid is unlikely to be present at such a high concentration, parthenin appears to be one of the main allelochemicals responsible for the inhibitory effect of parthenium leaf residue on salvinia.A portion of this work was presented at the International Symposium on Weed Management for Sustainable Agriculture held at C.C.S. Starting from the system of linearized kinetic equations, the exact expressions for the spin and mass diffusion coefficients and thermal diffusion ratio in quantum mixtures of helium isotopes with any degree of degeneration is obtained. From the general expression the various limiting expressions have been obtained, which are determined by various relations between collision rates of quasiparticles. The results of the calculations are compared with available experimental data. It is shown that in the low-temperature region, where an impuriton gas is one-component, the relaxation of concentration of 3 He in superfluid 3 He– 4 He mixtures is determined by the acoustic and dissipative collective modes with an effective diffusion coefficient. From the general relation, the expression for a spin diffusion coefficient in solid 3 He– 4 He quantum mixtures is derived. The comparison of the results obtained with the experimental data makes it possible to find a numerical value for the width of the impuriton energy zone.  相似文献

The hydrodynamics of the self propulsion of a spermatozoa, swimming through the mucus filling the cervical channel, is investigated. The mucus is modeled as a micropolar fluid and the spermatozoa as a 2-dimensional sheet swimming at low Reynolds number between two rigid walls. The wavelengths of the propulsive waves passing down the sheet are assumed to be very large compared to the channel spacing, but the amplitude of the propulsive waves is arbitrary. Expressions for the propulsive velocity and the energy expended by the swimming sheet are obtained in terms of various parameters involved. The results are elaborated through graphs. It is found that both the propulsive velocity and the rate of working by the sheet increase as the value of the micropolar parameters N increases and that of L decreases.  相似文献

In a grave illness the homeostasis of some elements may be upset either by the disease process per se, or by the therapy adopted. Patients with pyogenic, viral and tubercular meningo-encephalitis were targets of interest. 108 subjects' serum and cerebrospinal fluid (CSF) compartments were assayed for magnesium, calcium, zinc and copper, by atomic absorption spectrometry. Results showed that the serum pool was more intensely altered than the CSF pool in the case of magnesium and calcium but the reverse occurred for zinc and copper. No organism-dependent change was noted, but comatosed patients were the worst effected. These findings are discussed with respect to the relative brain and CSF content of each element and the patho-physiology which could alter the compartments studied.  相似文献

Bovine pancreatic lipase was isolated in pure form by lyophilization of fresh bovine pancreas, extraction of the enzyme with sucrose solution, fractional precipitation with ammonium sulfate and acetone, followed by chromatography on Sephadex G-100. The specific activity of the purest lipase fraction was 1750 micromoles fatty acid, liberated in 30 min per milligram of protein, indicating a purification of approximately 473-fold, with an overall yield of about 42%. Homogeneity of the enzyme was confirmed by rechromatography on Sephadex G-100 as well as with the gel electrophoretic and ultracentrifugal techniques. The purified enzyme gave a typical protein ultraviolet absorption spectrum with maximum absorption at 276 nm and minimum at 252 nm. The purified enzyme exhibited a single pH optimum of 8.8 and an isoelectric point near pH 5.5. Its optimum temperature was 37 C, and its optimum substrate concentration was 10%. These properties resembled those of milk lipase.  相似文献

A new chip-based method to identify protein-protein interactions was developed using the guanine nucleotide exchange factor GRF2 and two interacting proteins, Ras and calmodulin, as model proteins. A generic immobilization strategy for FLAG-tagged bait proteins on a protein-repellent streptavidin chip surface was implemented by presentation of an oriented anti-FLAG antibody. A flow cell device, integrating different chip surfaces, was developed, and the interaction of immobilized GRF2 with the two analytes was verified by fluorescence assays. On-chip tryptic digest assays were then performed on the capture surface and analyzed by microLC-MS/MS. The interaction of GRF2 with calmodulin and Ras was demonstrated, and the lower limit of detection was determined. We also implemented an on-chip immunoprecipitation assay to identify GRF2-binding partners from complex protein mixtures. Cells overexpressing FLAG-GRF2 were lysed and then incubated with the anti-FLAG chip. In addition to detecting GRF2, we also identified calmodulin, demonstrating that this technique can successfully identify endogenous levels of proteins, bound to recombinant bait proteins. This chip-based method has the advantage that no subsequent gel separations of protein complexes prior to LC-MS analysis are required and is therefore amenable to miniaturized high-throughput determination of protein-protein interactions.  相似文献

Phosphorylation is a common form of protein modification. To understand its biological role, the site of phosphorylation has to be determined. Generally, only limited amounts of phosphorylated proteins are present in a cell, thus demanding highly sensitive procedures for phosphorylation site determination. Here, a novel method is introduced which enables the localization of tyrosine phosphorylation in gel-separated proteins in the femtomol range. The method utilizes the immonium ion of phosphotyrosine at m/z 216.043 for positive ion mode precursor ion scanning combined with the recently introduced Q2-pulsing function on quadrupole TOF mass spectrometers. The high resolving power of the quadrupole TOF instrument enables the selective detection of phosphotyrosine immonium ions without interference from other peptide fragments of the same nominal mass. Performing precursor ion scans in the positive ion mode facilitates sequencing, because there is a no need for polarity switching or changing pH of the spraying solvent. Similar limits of detection were obtained in this approach when compared to triple-quadrupole mass spectrometers but with significantly better selectivity, owing to the high accuracy of the fragment ion selection. Synthetic phosphopeptides could be detected at 1 fmol/microL, and 100 fmol of a tyrosine phosphorylated protein in gel was sufficient for the detection of the phosphorylated peptide in the unseparated digestion mixture and for unambiguous phosphorylation site determination. The new method can be applied to unknown protein samples, because the identification and localization of the modification is performed on the same sample.  相似文献

Micromechanical structures that have squeeze-film damping as the dominant energy dissipation mechanism are of interest in this paper. For such structures with narrow air gap, the Reynolds equation is used for calculating squeeze-film damping, which is generally solved with trivial pressure boundary conditions on the side walls. This procedure, however, fails to give satisfactory results for structures under two important conditions: 1) for an air gap thickness comparable to the lateral dimensions of the microstructure and 2) for nontrivial pressure boundary conditions such as fully open boundaries on an extended substrate or partially blocked boundaries that provide side clearance to the fluid flow. Several formulas exist to account for simple boundary conditions. In practice, however, there are many micromechanical structures such as torsional microelectromechanical system (MEMS) structures that have nontrivial boundary conditions arising from partially blocked boundaries. Such boundaries usually have clearance parameters that can vary due to fabrication. These parameters, however, can also be used as design parameters if we understand their role on the dynamics of the structure. We take a MEMS torsion mirror as an example device that has large air gap and partially blocked boundaries due to static frames. We actuate the device and experimentally determine the quality factor Q from the response measurements. Next, we model the same structure in ANSYS and carry out computational fluid dynamics analysis to evaluate the stiffness constant K, the damping constant D, and the quality factor Q due to the squeeze film. We compare the computational results with experimental results and show that without taking care of the partially blocked boundaries properly in the computational model, we get unacceptably large errors.  相似文献

The authors examined whether a form of implicit memory that has been unambiguously dissociated from conscious awareness--learning of spatial context on the contextual cuing task introduced by M. M. Chun and Y. Jiang (1998)--is mature in childhood as predicted by an evolutionary view of cognition. School-aged children did not show reliable learning relative to adults who performed the same version of the task or another version that slowed responses to match those of children. Thus, unreliable learning in childhood was mediated by immature implicit representations of spatial context rather than by slower baseline response speed. The present finding is inconsistent with the prediction of the evolutionary view of cognition but consistent with incomplete maturation of medial temporal lobes known to mediate contextual learning. Diamond-like carbon (DLC) films were deposited by microwave assisted chemical vapour deposition system using d.c. bias voltage ranging from −100 V to −300 V. These films were characterized by X-ray photoelectron spectroscopy (XPS) and spectroscopic ellipsometry techniques for estimating sp 3/sp 2 ratio. The sp 3/sp 2 ratio obtained by XPS is found to have an opposite trend to that obtained by spectroscopic ellipsometry. These results are explained using sub-plantation picture of DLC growth. Our results clearly indicate that the film is composed of two different layers, having entirely different properties in terms of void percentage and sp 3/sp 2 ratio. The upper layer is relatively thinner as compared to the bottom layer.  相似文献
