The design and development of a small molecule named NPB [3-{(4(2,3-dichlorophenyl)piperazin-1-yl}{2-hydroxyphenyl)methyl}-N-cyclopentylbenzamide], which specifically inhibited the phosphorylation of BAD at Ser99 in human carcinoma cells has been previously reported. Herein, the synthesis, characterization, and effect on cancer cell viability of NPB analogs, and the single-crystal X-ray crystallographic studies of an example compound (4r), which was grown via slow-solvent evaporation technique is reported. Screening for loss of viability in mammary carcinoma cells revealed that compounds such as 2[(4(2,3-dichlorophenyl)piperazin-1-yl][naphthalen-1-yl]methyl)phenol (4e), 5[(4(2,3-dichlorophenyl)piperazin-1-yl][2-hydroxyphenyl)methyl)uran-2-carbaldehyde (4f), 3[(2-hydroxyphenyl][4(p-tolyl)piperazin-1-yl)methyl)benzaldehyde (4i), and NPB inhibited the viability of MCF-7 cells with IC50 values of 5.90, 3.11, 7.68, and 6.5 µM, respectively. The loss of cell viability was enhanced by the NPB analogs synthesized by adding newer rings such as naphthalene and furan-2-carbaldehyde in place of N-cyclopentyl-benzamide of NPB. Furthermore, these compounds decreased Ser99 phosphorylation of hBAD. Additional in silico density functional theory calculations suggested possibilities for other analogs of NPB that may be more suitable for further development. 相似文献

Bovine pancreatic lipase was isolated in pure form by lyophilization of fresh bovine pancreas, extraction of the enzyme with sucrose solution, fractional precipitation with ammonium sulfate and acetone, followed by chromatography on Sephadex G-100. The specific activity of the purest lipase fraction was 1750 micromoles fatty acid, liberated in 30 min per milligram of protein, indicating a purification of approximately 473-fold, with an overall yield of about 42%. Homogeneity of the enzyme was confirmed by rechromatography on Sephadex G-100 as well as with the gel electrophoretic and ultracentrifugal techniques. The purified enzyme gave a typical protein ultraviolet absorption spectrum with maximum absorption at 276 nm and minimum at 252 nm. The purified enzyme exhibited a single pH optimum of 8.8 and an isoelectric point near pH 5.5. Its optimum temperature was 37 C, and its optimum substrate concentration was 10%. The aim of this study was to examine the synthesis of a graft copolymer of chitosan and methacrylic acid (MAA) by free‐radical polymerization with a potassium peroxymonosulfate/cyclohexanone (CY) redox system in an inert atmosphere. The optimum reaction conditions affording maximum grafting ratio (%G), grafting efficiency (%E), add on (%A), and conversion (%C) were determined. The grafting parameters were found to increase with increasing concentration of MAA up to 24 × 10?2 mol/dm3, but thereafter, these parameters decreased. With increasing concentration of peroxymonosulfate from 0.6 × 10?2 to 1.2 × 10?2 mol/dm3, %G, %A, and %E increased continuously. All of these grafting parameters increased with increasing concentration of CY up to 1.2 × 10?2 mol/dm3, but beyond this concentration, the grafting parameters decreased. With various concentrations of chitosan from 0.6 to 1.4 g/dm3, the maximum %G, %A, and %E were obtained at 1.4 g/dm3. %G, %A, and %C decreased continuously with various concentrations of hydrogen ions from 2 × 10?3 to 6 × 10?3 mol/dm3. The grafting parameters increased with increasing temperature up to 35°C, but thereafter, these parameters decreased. With increasing time period of reaction from 60 to 180 min, %G, %A, and %E increased up to 120 min, but thereafter, these parameters decreased. Agar–gelatin-based co-hydrogels were prepared with different compositions of the agar and the gelatin fractions. The intermolecular hydrogen bonding was higher in the co-hydrogels as compared to the gelatin hydrogel. Swelling studies indicated diffusion-mediated swelling. The electrical stability of the co-hydrogels was higher as compared to the gelatin hydrogel. Though the firmness of the co-hydrogels was higher, Weichert model of viscoelasticity indicated that the inherent mechanical stability of the gelatin hydrogel was superior. The release of ciprofloxacin hydrochloride was predominately Fickian diffusion-mediated. In gist, the co-hydrogels can be tried as polymeric constructs for controlled drug delivery applications. 相似文献

Epoxy [50:50 mixture of Di‐Glycidyl Ether of Bis‐Phenol A (DGEBA) and Epoxidized Novolac (EPN)] was solution blended with Vinyl Acetate‐2‐ Ethylhexylacrylate (VAc‐EHA) resin in aqueous medium, in varying weight fractions, with Hexamethoxymethylmelamine (HMMM) as a crosslinker and data was compared with a control. The present work was aimed to optimize the tensile strength, dynamic mechanical strength, impact strength, and toughness by preparing a blend followed by jute composites of a semi‐ and full interpenetrating network (IPN). In control experiments epoxy alone was crosslinked (semi‐IPN), whereas the DGEBA‐EPN and VAc‐EHA/HMMM were crosslinked separately (full‐IPN), using jute as the substrate for making composites. Composites of full‐IPN systems of epoxy/VAc‐EHA system had higher moduli and UTS than the semi‐IPN systems. Dynamic mechanical study showed that full‐IPN systems have higher Tg values than semi‐IPN systems. Variation of ultrasonic attenuation and velocities with temperature have been evaluated in the temperature range 5-50 K due to electron-phonon interaction mechanism in transition metals vanadium, niobium and tantalum for longitudinal and shear waves. For this evaluation, we have also computed the second order elastic constants using Morse potential. Behaviour of acoustical phonons in these bcc metals is different from other normal metals, intermetallics, semimetallics and alloys. The aim of the present work is to get an insight of the phenomena behind the low-pressure low-power transients that occur during startup of a natural circulation boiling system. A RELAP5 model developed for a test facility and its prototype is used to record additional system parameters that were not included in the data obtained from experiments. The flow oscillations observed during experimental and numerical studies are analyzed and classified. It is inferred that the low amplitude oscillations are not condensation induced geysering instabilities, but a density wave instability supported by flashing. The similarity between the nature of startup transients observed in the test facility and the prototype is also examined. The effect of flashing is more pronounced in the prototype due to the strong variation of saturation temperature as the length scale is 4 times that of the model. The time series data obtained from experimental observations and numerical simulations are analyzed to identify the structural nature of flow oscillations. The power spectral density estimated using fast Fourier transform (FFT) algorithm illustrates the chaotic nature of the signals. The nonlinear time series analysis (TISEAN) package has been used for the estimation of Lyapunov exponent and the Poincaré section. The Poincaré section and the Lyapunov exponent confirm the chaotic nature of the flow oscillations. 相似文献

Ion conducting polymer electrolyte, poly(N-methyl 4-vinylpyridine iodide) (PVPI) is synthesized for dye-sensitized solar cell (DSSC) application. A new solid polymer electrolyte composite containing low viscosity ionic liquid (IL) 1-ethyl 3-methylimidazolium dicyanamide (EMImDCN) doped PVPI is developed and its structural, electrical and photoelectrochemical studies are presented in detail. Fourier transform infrared (FTIR) spectroscopy, proton NMR and atomic force microscopy (AFM) affirms the modified polymer and its composite nature with porous surface morphology. The developed solid polymer electrolyte shows enhancement in ionic conductivity (σ) due to IL doping. The maximum σ value of 9.12 × 10?6 S cm?1 was obtained at 40 wt% IL concentration. The redox behavior of the electrolyte has been verified by the cyclic voltammetry studies. For device application, we have fabricated a DSSC using this solid polymer–IL electrolyte system which shows energy conversion efficiency of the solid-state cell as 0.65% under irradiation of simulated sunlight (AM 1.5, 100 mW cm?2). 相似文献
