We have developed a microscope system for real-time single sperm tracking with an automated laser tweezers escape power assay. Phase contrast images of swimming sperm are digitized to the computer at video rate. The custom algorithm creates a region of interest centered about a sperm in response to a mouse click and performs all subsequent tasks autonomously. Microscope stage movement responds to feedback from video analysis of swimming sperm to center the sperm with respect to the field of view. For escape power assays, sperm are automatically relocated to the laser trap focus where they are held for a user-defined duration at fixed power, or held as laser power is gradually reduced. The sperm's position is automatically monitored to measure the laser power at which the sperm escapes the trap. Sperm are tracked for extended durations before and after laser trap experiments. Motility measurements including the curvilinear velocity and the absolute position of the sperm relative to the cell chamber are calculated and written to the hard drive at video rate. Experimental throughput is increased over 30 times compared to off-line data analysis. The efficacy of the "track and trap" algorithm is validated through examples and comparisons with the manually collected data.  相似文献

We present a memetic approach for multi-objective improvement of robustness influencing features (called robustness objectives) in airline schedules. Improvement of the objectives is obtained by simultaneous flight retiming and aircraft rerouting, subject to a fixed fleet assignment. Approximations of the Pareto optimal front are obtained by applying a multi-meme memetic algorithm. We investigate biased meme selection to encourage exploration of the boundaries of the search space and compare it with random meme selection. An external population of high quality solutions is maintained using the adaptive grid archiving algorithm.The presented approach is applied to investigate simultaneous improvement of reliability and flexibility in real world schedules from KLM Royal Dutch Airlines. Experimental results show that the approach enables us to obtain schedules with significant improvements for the considered objectives. A large scale simulation study was undertaken to quantify the influence of the robustness objectives on the operational performance of the schedules. Rigorous sensitivity analysis of the results shows that the influence of the schedule reliability is dominant and that increased schedule flexibility could improve the operational performance.  相似文献

Offset surfaces play an important role in various CAD/CAM applications. Given a set of oriented points, we propose a hierarchical method in this paper to fit both the zero-level surface and its offset surface with a single implicit function. The implicit function is formed by compactly supported radial basis functions (CSRBFs). Different from other existing methods in literature, our approach reconstructs an implicit function which interpolates or approximates both the zero-level surface and the offset surface of a given point set, simultaneously. Employing locally supported functions leads to an efficient computational procedure, while the coarse-to-fine hierarchy makes our approach insensitive to the density of scattered data and allows us to reconstruct large parts of missing data. The performance of our method is demonstrated by a number of examples and the application of adaptive slicing hollowed models in rapid prototyping.  相似文献

To further our understanding of the role interests play in the development of adult intellect, this study examines the relationship between intraindividual differences in interest and knowledge profiles. On the basis of a sample of 330,154 individuals, results show a moderate, positive average intraindividual correlation between interest and knowledge that increases as individuals age and is stronger for men than for women. Consistent with recent theories regarding the development of adult intelligence, expertise, and talent, these results demonstrate that noncognitive intraindividual differences can add to one's understanding of the acquisition of knowledge and skills. The current findings regarding the intraindividual convergence of interests and knowledge substantiate K. Murphy's (1998) call for renewed research on interests and point to the usefulness of an intraindividual perspective in applied psychology. Meat products can be contaminated with carcinogenic N-nitrosamines, which is ascribed to the reaction between a nitrosating agent, originating from nitrite or smoke, and a secondary amine, derived from protein and lipid degradation. Although in model systems it is demonstrated that many amine containing compounds can be converted to N-nitrosamines, the yield is dependent of reaction conditions (e.g., low pH and high temperature). In this article, the influence of the composition of the meat products (e.g., pH, aw, spices) and processing (e.g., ageing, ripening, fermentation, smoking, heat treatment and storage) on the presence and availability of the amine precursors and the N-nitrosamine formation mechanism is discussed. In addition, this article explores the current N-nitrosamine mitigation strategies in order to obtain healthier and more natural meat products.  相似文献

The flow behavior and recrystallization response of a 9310 steel alloy deformed in the ferrite temperature range were studied in this work. Samples were compressed under various conditions of strain (0.6, 0.8 and multi-axial), strain rate (10?4 seconds?1 to 10?1 seconds?1) and temperature [811 K to 1033 K (538 °C to 760 °C)] using a Gleeble thermo-mechanical simulator. Deformation was characterized by both qualitative and quantitative means, using standard microscopy, electron backscatter diffraction (EBSD) analysis and flow stress modeling. The results indicate that deformation is primarily accommodated through dynamic recovery in sub-grain formation. EBSD analysis shows a continuous increase in sub-grain boundary misorientation with increasing strain, ultimately producing recrystallized grains from the sub-grains at high strains. This suggests that a sub-grain rotation recrystallization mechanism predominates in this temperature range. Analyses of the results reveal a decreasing mean dynamically recrystallized grain size with increasing Zener-Hollomon parameter, and an increasing recrystallized fraction with increasing strain.  相似文献

BACKGROUND: High erucic acid oilseed rape (HEAR) was tested as a source crop for estimates of regional geneflow. Two methods to detect HEAR in low erucic acid oilseed rape (LEAR) were compared: real‐time quantitative PCR (qPCR) and gas chromatography (GC). RESULTS: Fields (2.5 ha) of a LEAR variety (0.028% EA) in Tayside and Hertfordshire were juxtaposed adjacent to and 1 km distant from a HEAR (44% EA) field. The LEAR variety was a varietal association to ensure high cross‐pollination (CP). The methods were highly correlated, measuring between 30% and 0.5% CP. However, the qPCR method became unreliable below 0.5% CP, whereas GC was robust enough to detect raised EA equivalent to one F1 seed in 500 (0.2%). A statistical mixture model was fitted to the distributions of EA in samples in order to assign a CP value to each 500‐seed sample. Declines of CP from 30% to < 1%, and EA from 5% to 0.2%, with distance up to 150 m in the near fields was best fitted with a power function. The combined mean EA for both far fields was 0.11%, well above the background LEAR value of 0.028%, and mean CP was 0.36%. CONCLUSIONS: The GC method of detection raised %EA should be a reliable and high‐throughput means of estimating %CP between fields, provided the %EA of single F1 seed in receptor fields is measured to confirm the presence of F1 seed. We estimated the mass budget for mercury (Hg) seasonally deposited into the Florida Everglades and investigated seasonality of Hg cycling by analyzing data obtained for water, soil, flocculent detrital material (floc), periphyton, and mosquitofish collected throughout the Everglades freshwater marshes in the 2005 dry and wet seasons. Higher wet season total Hg (THg) in soil, floc, and periphyton agreed with greater Hg amounts entering these compartments during the wet season, probably owing to substantially greater Hg deposition in the wet season than in the dry season. Seasonal differences were absent for THg in surface water. Methylmercury (MeHg) showed mixed seasonal patterns, with higher water and soil MeHg and lower periphyton MeHg in the dry season but no seasonality for floc MeHg. Seasonal variations in Hg deposition, MeHg production and transport, and mass of ecosystem compartments could be responsible for the seasonality of MeHg cycling. Higher mosquitofish THg, higher bioaccumulation factors, and higher biomagnification factors from periphyton to mosquitofish were observed in the wet season than in the dry season, indicating that the wet season is more favorable for Hg bioaccumulation.
