We report a new, bioconjugate approach to performing highly multiplexed single-base extension (SBE) assays, which we demonstrate by genotyping a large panel of point mutants in exons 5-9 of the p53 gene. A series of monodisperse polyamide "drag-tags" was created using both chemical and biological synthesis and used to achieve the high-resolution separation of genotyping reaction products by microchannel electrophoresis without a polymeric sieving matrix. A highly multiplexed SBE reaction was performed in which 16 unique drag-tagged primers simultaneously probe 16 p53 gene loci, with an abbreviated thermal cycling protocol of only 9 min. The drag-tagged SBE products were rapidly separated by free-solution conjugate electrophoresis (FSCE) in both capillaries and microfluidic chips with genotyping accuracy in excess of 96%. The separation requires less than 70 s in a glass microfluidic chip, or about 20 min in a commercial capillary array sequencing instrument. Compared to gel electrophoresis, FSCE offers greater freedom in the design of SBE primers by essentially decoupling the length of the primer and the electrophoretic mobility of the genotyping products. FSCE also presents new possibilities for the facile implementation of SBE on integrated microfluidic electrophoresis devices for rapid, high-throughput genetic mutation detection or SNP scoring.  相似文献

One important feature of the baroreflex loop is its strong preference for oscillations around 0.1 Hz. In this study, we investigated heart rate intervals, arterial blood pressure (BP), and prefrontal oxyhemoglobin changes during 5 min rest and during brisk finger movements in 19 healthy subjects. We analyzed the phase coupling around 0.1 Hz between cardiovascular and (de)oxyhemoglobin oscillations, using the cross-spectral method. The analyses revealed phase shifts for slow oscillations in BP and heart rate intervals between -10° and -118° (BP always leading). These phase shifts increased significantly (p<0.01) in the movement session. The coupling between cardiovascular and oxyhemoglobin oscillations was less clear. Only 12 subjects demonstrated a phase coupling (COH(2) ≥ 0.5) between oxyhemoglobin and BP oscillations. This may be explained by an overwhelming proportion of nonlinearity in cardiovascular and hemodynamic systems. The phase shifts between slow cardiovascular and hemodynamic oscillations are relatively stable subject-specific biometric features and could be of interest for person identification in addition to other biometric data. Slow BP-coupled oscillations in prefrontal oxyhemoglobin changes can seriously impair the detection of mentally induced hemodynamic changes in an optical brain-computer interface, a novel nonmuscular communication system.  相似文献

Lipid metabolites regulate fatty acid and glucose homeostasis. The intention of the current study is to identify circulating lipid species, which are altered in rodent obesity and strongly correlate with the classically measured metabolites glucose, triglycerides, and cholesterol. Mice fed a high fat diet (HFD) for 14 weeks have increased body weight and fasting glucose. Serum triglycerides are not altered, while cholesterol tends to be increased. Accordingly, major cholesteryl ester (CE) species and free cholesterol are not significantly raised in obesity while minor metabolites, including CE 20:3 and CE 18:3, are increased or reduced, respectively. Distinct sphingomyelin (SM) species are elevated while ceramides are not raised. Phosphatidylinositol (PI) species, including PI 34:1, are raised while others are decreased. PI 34:1 strongly correlates with fasting glucose and proinsulin levels. Phosphatidylcholine (PC) 26:0, 40:2, and 40:5, which are induced in obesity, correlate with cholesterol. PC 38:4 and PC 40:6 are also raised in fat fed mice and positively correlate with fasting glucose. Lysophosphatidylcholine (LPC) species are also changed in obesity and the already shown reduction of LPC 16:1 has been confirmed. LPC 22:4, which is increased, correlates with serum cholesterol. The data indicate that circulating levels of various lipid species are changed in the obesity model studied and some of them are strongly associated with classically measured metabolites.  相似文献

Aggregation behavior and reduced locomotory activity in the German cockroach is known to be caused by chemical compounds in the feces. The attractive and/or arrestant efficacy of three relevant substances was tested in first instars by a two-choice aggregation test and in adults with a locomotion compensator apparatus that allows quantification of taste-directed orientation and walking speed as a function of antennal stimulation. The three substances tested were a feces crude extract; a mixture of six carboxylic acids (mix G) out of a total of 29 that were identified in the feces extract and tested as single compounds and in various combinations; and a steroid glucoside denoted as blattellastanoside A, which has been suggested as an aggregation arrestant pheromone in Blattella germanica. With both of our test methods, feces extract and mix G proved to be very attractive, whereas the effects of blattellastanoside A were, if anything, very poor. Possible reasons for discrepancies are discussed.  相似文献

There is an urgent and compelling need to develop innovative and more effective ways to integrate sustainable renewable energy solutions into the already existing systems or, better yet, create new systems that all together make use of renewable energy. This study aims to establish the optimum working conditions of a geothermal preheater in a power plant that makes use of both renewable and nonrenewable energy resources, where renewable (geothermal) energy is used to boost the power output in an environmentally sustainable way. Hence, two models, one, a simplified model of a Rankine cycle with single reheat and regeneration, and another, with a geothermal preheater substituting the low-pressure feedwater heater (LPFWH), were compared. The Engineering Equations Solver software was used to perform an analysis of the thermodynamic performance of the two models designed. An analysis was done to evaluate the energetic and exergetic effects of replacing a LPFWH with a geothermal preheater sourcing heat from a low temperature geothermal resource (100°C-160°C). Results from the thermodynamic analysis reveal that the hybridization boosts the power output by approximately 4% and it is superior in terms of the second law. Entropy generation minimization analysis was then employed to establish optimal working conditions of the hybrid system (ie, the geothermal preheater modeled as a downhole coaxial heat exchanger).  相似文献
