Although obesity and high levels of low-density lipoprotein (LDL) are well-known risk factors for cardiovascular disease, the precise role(s) of different LDL constituents in obesity has not been explored. In the present study, we compared the LDL proteome of healthy control adults (body mass index<25) and obese subjects (body mass index>30). LDL was isolated by density-gradient ultracentrifugation and proteins were separated with 2-D PAGE, quantified, and identified by peptide mass fingerprinting using MALDI-TOF MS. A new LDL-associated protein was identified as transthyretin and found to be significantly more abundant in LDL from the obese subjects. In addition, LDL from the obese subjects contained relatively more α(1) -antitrypsin, apo J, apo C-II, than LDL from controls, and also more of an acidic isoform (pI/Mr; 5.2/23 100) of apo A-I. On the other hand, the relative amounts of apo A-IV and the major isoform of apo A-I (pI/Mr; 5.3/23 100) were significantly less in LDL from the obese subjects. Apo E was less and non-sialylated apo C-III more abundant in LDL from obese men than control men, while there were no such differences between LDL from obese and control women. These findings illustrate that obesity is not only associated with increased LDL-cholesterol levels but also with alterations in the LDL protein composition. The presence of transthyretin in LDL from obese subjects may reflect over-nutrition and affect the lipid metabolism in obesity.  相似文献

As in every engineering discipline, metrics play an important role in software development, with the difference that almost all software projects need the customization of metrics used. In other engineering disciplines, the notion of a measurement system (i.e. a tool used to collect, calculate, and report quantitative data) is well known and defined, whereas it is not as widely used in software engineering. In this paper we present a framework for developing custom measurement systems and its industrial evaluation in a software development unit within Ericsson. The results include the framework for designing measurement systems and its evaluation in real life projects at the company. The results show that with the help of ISO/IEC standards, measurement systems can be effectively used in software industry and that the presented framework improves the way of working with metrics. This paper contributes with the presentation of how automation of metrics collection and processing can be successfully introduced into a large organization and shows the benefits of it: increased efficiency of metrics collection, increased adoption of metrics in the organization, independence from individuals and standardized nomenclature for metrics in the organization.  相似文献

Computationally tractable planning problems reported in the literature so far have almost exclusively been defined by syntactical restrictions. To better exploit the inherent structure in problems, it is probably necessary to study also structural restrictions on the underlying state-transition graph. The exponential size of this graph, though, makes such restrictions costly to test. Hence, we propose an intermediate approach, using a state-variable model for planning and defining restrictions on the separate state-transition graphs for each state variable. We identify such restrictions which can tractably be tested and we present a planning algorithm which is correct and runs in polynomial time under these restrictions. The algorithm has been implemented and it outperforms Graphplan on a number of test instances. In addition, we present an exhaustive map of the complexity results for planning under all combinations of four previously studied syntactical restrictions and our five new structural restrictions. This complexity map considers both the optimal and non-optimal plan generation problem.  相似文献

We present a class, 3S, of planning instances such that the plan existence problem is tractable while plan generation is provably intractable for instances of this class. The class is defined by simple structural restrictions, all of them testable in polynomial‐time. Furthermore, we show that plan generation can be carried out in time bounded by a polynomial in the size of the input and the size of the generated solution. For this class, we propose a provably sound and complete incremental planner, i.e., a planner that can usually output an executable prefix of the final plan before it has generated the whole plan. Many moth pheromones are composed of mixtures of acetates of long‐chain (≥10 carbon) fatty alcohols. Moth pheromone precursors such as fatty acids and fatty alcohols can be produced in yeast by the heterologous expression of genes involved in insect pheromone production. Acetyltransferases that subsequently catalyze the formation of acetates by transfer of the acetate unit from acetyl‐CoA to a fatty alcohol have been postulated in pheromone biosynthesis. However, so far no fatty alcohol acetyltransferases responsible for the production of straight chain alkyl acetate pheromone components in insects have been identified. In search for a non‐insect acetyltransferase alternative, we expressed a plant‐derived diacylglycerol acetyltransferase (EaDAcT) (EC 2.3.1.20) cloned from the seed of the burning bush (Euonymus alatus) in a yeast system. EaDAcT transformed various fatty alcohol insect pheromone precursors into acetates but we also found high background acetylation activities. Only one enzyme in yeast was shown to be responsible for the majority of that background activity, the acetyltransferase ATF1 (EC 2.3.1.84). We further investigated the usefulness of ATF1 for the conversion of moth pheromone alcohols into acetates in comparison with EaDAcT. Overexpression of ATF1 revealed that it was capable of acetylating these fatty alcohols with chain lengths from 10 to 18 carbons with up to 27‐ and 10‐fold higher in vivo and in vitro efficiency, respectively, compared to EaDAcT. The ATF1 enzyme thus has the potential to serve as the missing enzyme in the reconstruction of the biosynthetic pathway of insect acetate pheromones from precursor fatty acids in yeast.  相似文献

Forest cover decline is one of the most important environmental issues in the tropics. The present study was carried out in Burkina Faso, West Africa, and aimed at assessing the trajectories of forest cover change and measuring landscape metrics of the trajectory classes in order to better understand the processes of change. Landsat and ASTER images acquired over a period of 30 years were used for cover change detection and the Fragstats package was used to compute landscape metrics with five unifying change classes. Results showed a substantial increase in cropland with concurrent decline in forest cover. Deforestation represented 63% of the Percentage of Landscape (PLAND) in 2006, while reforestation accounted for only 28%. Both of these classes had high Normalized Landscape Shape Index (NLSI) values, indicating that they were present as scattered small patches. The old cultivation (30-year permanent cropland) was aggregated (IJI ≈ 0) while deforestation exhibited highly interspersed patches. The old forest and old cultivation presented lower Area Weighted Fractal Dimension Index (FRAC_AM), but deforestation and reforestation had the higher FRAC_AM. These results confirmed that there was a high level of deforestation and fragmentation in southern Burkina Faso and justify the need for a proper management plan to ensure the sustainable use of forest resources.  相似文献
