SARS-CoV-2 variants raise concern because of their high transmissibility and their ability to evade neutralizing antibodies elicited by prior infection or by vaccination. Here, we compared the neutralizing abilities of sera from 70 unvaccinated COVID-19 patients infected before the emergence of variants of concern (VOCs) and of 16 vaccine breakthrough infection (BTI) cases infected with Gamma or Delta against the ancestral B.1 strain, the Gamma, Delta and Omicron BA.1 VOCs using live virus. We further determined antibody levels against the Nucleocapsid (N) and full Spike proteins, the receptor-binding domain (RBD) and the N-terminal domain (NTD) of the Spike protein. Convalescent sera featured considerable variability in the neutralization of B.1 and in the cross-neutralization of different strains. Their neutralizing capacity moderately correlated with antibody levels against the Spike protein and the RBD. All but one convalescent serum failed to neutralize Omicron BA.1. Overall, convalescent sera from patients with moderate disease had higher antibody levels and displayed a higher neutralizing ability against all strains than patients with mild or severe forms of the disease. The sera from BTI cases fell into one of two categories: half the sera had a high neutralizing activity against the ancestral B.1 strain as well as against the infecting strain, while the other half had no or a very low neutralizing activity against all strains. Although antibody levels against the spike protein and the RBD were lower in BTI sera than in unvaccinated convalescent sera, most neutralizing sera also retained partial neutralizing activity against Omicron BA.1, suggestive of a better cross-neutralization and higher affinity of vaccine-elicited antibodies over virus-induced antibodies. Accordingly, the IC50: antibody level ratios were comparable for BTI and convalescent sera, but remained lower in the neutralizing convalescent sera from patients with moderate disease than in BTI sera. The neutralizing activity of BTI sera was strongly correlated with antibodies against the Spike protein and the RBD. Together, these findings highlight qualitative differences in antibody responses elicited by infection in vaccinated and unvaccinated individuals. They further indicate that breakthrough infection with a pre-Omicron variant boosts immunity and induces cross-neutralizing antibodies against different strains, including Omicron BA.1.  相似文献

Phospholipids represent a crucial component for the structure of cell membranes. Phosphatidylcholine and phosphatidylethanolamine are two phospholipids that comprise the majority of cell membranes. De novo biosynthesis of phosphatidylcholine and phosphatidylethanolamine occurs via the Kennedy pathway, and perturbations in the regulation of this pathway are linked to a variety of human diseases, including cancer. Altered phosphatidylcholine and phosphatidylethanolamine membrane content, phospholipid metabolite levels, and fatty acid profiles are frequently identified as hallmarks of cancer development and progression. This review summarizes the research on how phospholipid metabolism changes over oncogenic transformation, and how phospholipid profiling can differentiate between human cancer and healthy tissues, with a focus on colorectal cancer, breast cancer, and non-small cell lung cancer. The potential for phospholipids to serve as biomarkers for diagnostics, or as anticancer therapy targets, is also discussed.  相似文献

Ultrathin films of poly(thiophene) (PT) and poly(bithiophene) (PBT) were prepared by electrochemical route using ionic liquid (BFEE) as medium and electrolyte. Distinct morphologies and electrical properties were observed in these materials. To evaluate its response in photovoltaics, these films were used as active layer in bilayer geometry solar cells with the electron acceptor molecule C60. The best performance was observed for PT films. In order to probe the differences in molecular dynamics and structural order, ultrafast electron dynamics in the low-femtosecond regime was evaluated by resonant Auger spectroscopy using the core–hole clock method at the sulfur K absorption edge. Electron delocalization times for the different polymeric films were derived as a function of the excitation energy. Photoabsorption measurements were conducted and molecular orientation derived. These results corroborated with the morphology found for these films and thus the performance of PT and PBT in the devices, and with the proposed conduction mechanism.  相似文献

This study examined the effects of O3 and hot water treatments on the epidermis of Golden papaya fruit. Heat treatments were applied in a hot water brushing (HWB) system. Papayas were brushed under a pressurized hot water rinse stage at 45, 55 and 65 °C for 60 s. In the HWB treatment, 4 ppm ozone was applied to the papayas for 1 or 2 min. The results show that ozone applications did not affect the fruit's cuticular surface, while heat treatments allowed natural fissures on the fruit epidermis to recover. Several crystalloid forms were identified on the epidermis of the papayas after the heat treatments. The predominant crystalloid forms on papayas are tubular and there is a positive response to temperature; the higher the temperature, the larger and more frequent the tubular crystalloids.  相似文献

Transaction costs theory (TCT) has long been an important conceptual lens for examining International Business (IB) phenomena and perhaps especially relevant for the study of multinational corporations, entry mode choices and location selection. In this paper we examine the extent to which TCT been used and has impacted IB research. Methodologically, we conduct a bibliometric study of the articles published on nine top journals for publishing IB-related research. We use Jean-François Hennart’s research as the key marker for TCT in IB research given that Hennart’s work has been a hallmark in the discipline. On a sample of 377 articles published between 1982 and 2010, and using the works rather than the authors as the unit of analysis, we analyze citations, co-citations and a spatial visualization of the intellectual research themes delved into. Our analyses provide insights on the influence of Hennart but more broadly of TCT on IB research over the past three decades. We conclude that the TCT has a pervasive influence on a large array of IB research and that Hennart’s work is boundary-spanning, connecting several research themes.  相似文献

Clotrimazole is a common choice for the treatment of vulvovaginal infections, but its low solubility and some side effects pose a challenge to its application. This work evaluated the feasibility to formulate clotrimazole-loaded cationic nanocapsules using Eudragit® RS100 and medium chain triglycerides as polymer and oily core, respectively, by the method of interfacial deposition of a preformed polymer. The physicochemical characteristics of nanocapsule formulations were evaluated at 0 day and 60 days after preparation. Particle size, zeta potential, polydispersity index, pH and drug content were stable during this period. In addition, nanocapsules were able to protect clotrimazole from photodegradation under UV radiation. By the dialysis bag diffusion technique, the nanosized formulations showed prolonged release of clotrimazole by anomalous transport and first order kinetics. A microbiological study was carried out by the microdilution method and showed that nanocapsules (mean size: 144 nm; zeta potential: + 12 mV) maintained the antifungal activity of clotrimazole against Candida albicans and Candida glabrata strains susceptible and resistant to fluconazole.  相似文献

Optimal operation models for a hydropower system using partial constraint satisfaction (PCS) approaches are proposed and developed in this study. The models use mixed integer nonlinear programming (MINLP) formulations with binary variables. The models also integrate a turbine unit commitment formulation along with water quality constraints used for evaluation of reservoir downstream water quality impairment. New PCS-based models for hydropower optimization formulations are developed using binary and continuous evaluator functions to maximize the constraint satisfaction. The models are applied to a real-life hydropower reservoir system in Brazil. Genetic Algorithms (GAs) are used to solve the optimization formulations. Decision maker's preferences towards power production targets and water quality improvements are incorporated using partial satisfaction constraints to obtain compromise operating rules for a multi-objective reservoir operation problem dominated by conflicting goals of energy production, water quality and consumptive water uses.  相似文献

For stationary sequences, under general dependence restrictions, any limiting point process for time normalized upcrossings of high levels is a compound Poisson process, i.e., there is a clustering of high upcrossings, where the underlying Poisson points represent cluster positions and the multiplicities correspond to cluster sizes. For such classes of stationary sequences, there exists the upcrossings index η, 0≤η≤1, which is directly related to the extremal index θ, 0≤θ≤1, for suitable high levels. In this paper, we consider the problem of estimating the upcrossings index η for a class of stationary sequences satisfying a mild oscillation restriction. For the proposed estimator, properties such as consistency and asymptotic normality are studied. Finally, the performance of the estimator is assessed through simulation studies for autoregressive processes and case studies in the fields of environment and finance. Comparisons with other estimators derived from well known estimators of the extremal index are also presented.  相似文献

In this article, novel FIFO and RAM-based Synchronization Modules to keep synchronism throughout the input channels of a Data Acquisition Electronics (DAE) system are proposed. DAE is a main component of a Medical Imaging System, namely, a Positron Emission Mammography (PEM) system. DAE input data comes from a scanner constituted by an array of scintillating crystals. The scanner captures radiation generated by human cells injected with a radioactive substance and converts it into electrical signals. The corresponding digital information is sent to the DAE. In order to deal with the huge amount of data, flowing at high data rates, point-to-point (p2p) communication channels are used between the scanner and the DAE. Propagation delays associated with the different communication channels may change differently. Additionally, differences among channel delays may exceed one clock period. Keeping synchronism in these circumstances requires more than the classical asynchronous FIFO solution. All these aspects motivate the work proposed in this article. The PEM DAE system is a multi-board, multi-FPGA, multi-clock domain system. Therefore, the DAE architecture follows a Globally Asynchronous, Locally Synchronous (GALS) design style. The novel Synchronization Modules proposed in this article are implemented in the DAE. The effectiveness of these new structures is validated through simulation and laboratorial test.
