The immune response to SARS-CoV-2 infection requires antibody recognition of the spike protein. In a study designed to examine the molecular features of anti-spike and anti-nucleocapsid antibodies, patient plasma proteins binding to pre-fusion stabilised complete spike and nucleocapsid proteins were isolated and analysed by matrix-assisted laser desorption ionisation–time of flight (MALDI-ToF) mass spectrometry. Amongst the immunoglobulins, a high affinity for human serum albumin was evident in the anti-spike preparations. Careful mass comparison revealed the preferential capture of advanced glycation end product (AGE) forms of glycated human serum albumin by the pre-fusion spike protein. The ability of bacteria and viruses to surround themselves with serum proteins is a recognised immune evasion and pathogenic process. The preference of SARS-CoV-2 for AGE forms of glycated serum albumin may in part explain the severity and pathology of acute respiratory distress and the bias towards the elderly and those with (pre)diabetic and atherosclerotic/metabolic disease.  相似文献

In the design process of transmissions, one major criterion is the resulting noise emission of the powertrain due to the gear excitation. Within the past years, a lot of investigations have shown that the noise emission can be correlated to the quasi-static transmission error. Therefore, the transmission error can be used as a characteristic value for quality assurance by experimental inspections as well as for a tooth contact analysis in the design process.The noise behavior of gearboxes is mainly caused by the excitation in the tooth mesh. The standardized design and calculation methods for gears concentrate on the reduction of the excitation level. But often the physical sound characteristics do not fit in with the human noise perception. Thus gear design rules are required that are able to rate the excitation according to the perception.The effect of the targeted topography scatter generally described is the reduction of the gear mesh amplitudes with an increase of the background noise. The objective of the report is to develop an understanding of the influence of different micro geometry scatters on the excitation behavior.Finally, it is the aim to design an aurally-accurate micro geometry scattering for the optimization of ground bevel gears in terms of tonality reduction and increased background noise. By means of a variant calculation and weighted target variables, psychoacoustic optimized micro geometry scattering is designed. At the same time, attention is also focused on a simple production of mixed topographies.  相似文献

Lysine-specific demethylase 1 (LSD1) has evolved as a promising therapeutic target for cancer treatment, especially in acute myeloid leukaemia (AML). To approach the challenge of site-specific LSD1 inhibition, we developed an enzyme-prodrug system with the bacterial nitroreductase NfsB (NTR) that was expressed in the virally transfected AML cell line THP1-NTR+. The cellular activity of the NTR was proven with a new luminescent NTR probe. We synthesised a diverse set of nitroaromatic prodrugs that by design do not affect LSD1 and are reduced by the NTR to release an active LSD1 inhibitor. The emerging side products were differentially analysed using negative controls, thereby revealing cytotoxic effects. The 2-nitroimidazolyl prodrug of a potent LSD1 inhibitor emerged as one of the best prodrug candidates with a pronounced selectivity window between wild-type and transfected THP1 cells. Our prodrugs are selectively activated and release the LSD1 inhibitor locally, proving their suitability for future targeting approaches.  相似文献

The suitability of phenyl-based deep eutectic solvents (DESs) as absorbents for toluene absorption was investigated by means of thermodynamic modeling and molecular dynamics (MD). The thermodynamic models perturbed-chain statistical associating fluid theory (PC-SAFT) and conductor-like screening model for real solvents (COSMO-RS) were used to predict the vapor–liquid equilibrium of DES–toluene systems. PC-SAFT yielded quantitative results even without using any binary fitting parameters. Among the five DESs studied in this work, [TEBAC][PhOH] consisting of triethyl benzyl ammonium chloride (TEBAC) and phenol (PhOH), was considered as the most suitable absorbent. Systems with [TEBAC][PhOH] had lowest equilibrium pressures of the considered DES–toluene mixtures, the best thermodynamic characteristics (i.e., Henry's law constant, excess enthalpy, Gibbs free energy of solvation of toluene), and the highest self-diffusion coefficient of toluene. The molecular-level mechanism was explored by MD simulations, indicating that [TEBAC][PhOH] has the strongest interaction of DES–toluene compared to the other DESs under study. This work provides guidance to rationally design novel DESs for efficient aromatic volatile organic compounds absorption.  相似文献

The curing of an epoxy consisting of the solid hardener dicyandiamide (DICY) and the resin diglycidyl ether of bisphenol A (DGEBA) is studied in a system consisting of a tablet of DICY embedded in liquid DGEBA. Dissolution of DICY within the liquid DGEBA in combination with the transport of dissolved DICY from the tablet border into DGEBA and the chemical reaction of both reactants is studied by scanning Brillouin microscopy and infrared spectroscopy. Scanning Brillouin microscopy demonstrates the spatial and temporal evolution of the static and dynamic hypersonic properties in the course of curing in the vicinity of the DICY tablet. Infrared spectroscopy performed on epoxy pieces extracted from the final sample at different distances from the tablet surface give information about the spatial evolution of the curing process. The results achieved by both techniques are finally combined to yield a better understanding of the curing of DICY-based epoxies, which transform upon curing from strongly heterogeneous systems towards increasingly homogeneous systems.  相似文献

Particulate emissions from low-temperature biomass burning are dominated by organic matter. Here, we show that such emissions have a liquid, bead-like appearance when collected on fibrous filters, and the number of these beads are far less than expected for solid spherical particles. These shapes are in line with published drop-on-fiber theories for liquids entrained on filaments. A smoldering pine sample is yellowish, with organic carbon over 99% of the total carbon, and chars substantially in thermal-optical analysis (TOA), indicating that such liquid organic particles could affect both absorption measurements and TOA of such samples. Similar colored samples collected in the field from rice-straw burning and cook stove emissions also show a similar liquid appearance.  相似文献

Bulk heterojunction based polymer:fullerene solar cells have attracted intensive research interest both in academic and industrial communities in the last two decades, mainly related to their potential low‐cost production process. A power conversion efficiency of over 10% has been reported recently, making the commercialization of this clean and cheap solar energy convertor a realistic prospect for the near future. The intrinsic features of semitransparency and color tunability of the thin polymeric photoactive films are the greatest asset for polymer solar cells. Recently, aesthetic semitransparent polymer solar cells (ST‐PSCs) that can be integrated into transparent windows, roofs, glass and other semitransparent architectural elements have received much attention. In this perspective paper, we present the progress in achieving high performance ST‐PSCs, discuss the requirements for transparent electrodes, focusing on alternatives to tin‐doped indium oxide, and address the challenges ahead to make ST‐PSC viable for real applications. © 2013 Society of Chemical Industry  相似文献
