Food consumption patterns have changed significantly during the last years, showing an increase in the consumption of convenience foods. Frozen foods represent an important segment of this market. The aim of the present paper was to study the influence of frozen storage on the quality of cooked organic tagliatelle. Instrumental measurements of different quality parameters (moisture content, colour, textural and rheological characteristics of frozen cooked pasta) were performed during twelve months of storage. The sensory properties of pasta were also evaluated by means of an acceptability study. In general, both instrumental and sensory analysis found that frozen storage affects negatively the acceptability of cooked pasta. The advantages obtained through fast freezing procedures are not maintained along storage, being hardness (instrumental) and consistency (sensory) the most relevant quality indices. Finally, the shelf life of the product was calculated as 3.6 and 3.8 months in the cryogenic device and air‐blast tunnel, respectively, based on simple kinetic models.  相似文献

GC/MS was used to identify compounds of essential oils from seven Ocimum taxa (O. americanum L., O. basilicum L., O. campechianum Mill., O. x citriodorum Vis., O. kilimandscharicum Baker ex Gürke and three botanical varieties and cultivars of Ocimum basilicum L.: ‘Genovese’, var. difforme and var. purpurascens). Preliminary screening of their antibacterial activity was done against a number of common pathogens (Enterococcus faecalis, Enterococus faecium, Escherichia coli 0157:H7, Listeria monocytogenes, Listeria ivanovii, Proteus vulgaris, Staphylococcus aureus, Staphylococcus epidermis) using the filter paper disc agar diffusion technique, while further analyses were done by modification of the disc diffusion method. A broad variation in the antibacterial properties of investigated essential oils was observed. E. coli 0157:H7 was inhibited by O. basilicum ‘Genovese’ essential oil, while Ocimum americanum and Ocimum x citriodorum essential oils were the most effective against Enterococcus faecalis, Enterococcus faecium, P. vulgaris, S. aureus and S. epidermis.  相似文献

The role of two common polymorphisms of enzymes involved in the metabolism of drugs and carcinogens was studied in relation to prostate cancer. The gene encoding one of these enzymes (NAT2) is located in an area where frequent allelic loss occurs in prostate cancer. Mutations at the genes CYP2D6 and NAT2 were analysed by allele-specific polymerase chain reaction and restriction mapping in DNA from 94 subjects with prostate cancer and 160 male healthy control subjects. Eleven prostate specimens were analysed for genotype and enzymatic activities NAT2, CYP2D6 and CYP3A by using the enzyme-specific substrates sulphamethazine and dextromethorphan. Enzyme activities with substrate specificities corresponding to NAT2, CYP2D6 and CYP3A are present in human prostate tissue, with mean +/-s.d. activities of 4.8+/-4.4 pmol min(-1) mg(-1) protein, 156+/-91 and 112+/-72 nmol min(-1) mg(-1) protein respectively. The Km values for the prostate CYP2D6 and CYP3A enzyme activities corresponded to that of liver CYP2D6 and CYP3A activities, and the CYP2D6 enzyme activity is related to the CYP2D6 genotype. The N-acetyltransferase, in contrast, had a higher Km than NAT2 and was independent of the NAT2 genotype. The CYP2D6 and CYP3A enzymes, and an N-acetyltransferase activity that is independent of the regulation of the NAT2 gene, are expressed in human prostate tissue. The presence of carcinogen-metabolizing enzymes in human prostate with a high interindividual variability may be involved in the regulation of local levels of carcinogens and mutagens and may underlie interindividual differences in cancer susceptibility.  相似文献

Sphingosine-1-phosphate (SPP) is a novel lipid messenger that has dual function. Intracellularly it regulates proliferation and survival, and extracellularly, it is a ligand for the G protein-coupled receptor Edg-1. Based on peptide sequences obtained from purified rat kidney sphingosine kinase, the enzyme that regulates SPP levels, we report here the cloning, identification, and characterization of the first mammalian sphingosine kinases (murine SPHK1a and SPHK1b). Sequence analysis indicates that these are novel kinases, which are not similar to other known kinases, and that they are evolutionarily conserved. Comparison with Saccharomyces cerevisiae and Caenorhabditis elegans sphingosine kinase sequences shows that several blocks are highly conserved in all of these sequences. One of these blocks contains an invariant, positively charged motif, GGKGK, which may be part of the ATP binding site. From Northern blot analysis of multiple mouse tissues, we observed that expression was highest in adult lung and spleen, with barely detectable levels in skeletal muscle and liver. Human embryonic kidney cells and NIH 3T3 fibroblasts transiently transfected with either sphingosine kinase expression vectors had marked increases (more than 100-fold) in sphingosine kinase activity. The enzyme specifically phosphorylated D-erythro-sphingosine and did not catalyze the phosphorylation of phosphatidylinositol, diacylglycerol, ceramide, D,L-threo-dihydrosphingosine or N, N-dimethylsphingosine. The latter two sphingolipids were competitive inhibitors of sphingosine kinase in the transfected cells as was previously found with the purified rat kidney enzyme. Transfected cells also had a marked increase in mass levels of SPP with a concomitant decrease in levels of sphingosine and, to a lesser extent, in ceramide levels. Our data suggest that sphingosine kinase is a prototypical member of a new class of lipid kinases. Cloning of sphingosine kinase is an important step in corroborating the intracellular role of SPP as a second messenger.  相似文献

The aim of this research was to prove the function of the putative opine dehydrogenase from Desulfohalobium retbaense and to characterize the enzyme in terms of functional and kinetic parameters. A putative opine dehydrogenase was identified from a metagenomic library by a sequence-based technique search of the metagenomic library, and afterward was successfully heterologously produced in Escherichia coli. In order to examine its potential for applications in the synthesis of secondary amines, first the substrate specificity of the enzyme towards different amino donors and amino acceptors was determined. The highest affinity was observed towards small amino acids, preferentially L-alanine, and when it comes to α-keto acids, pyruvate proved to be a preferential amino acceptor. The highest activity was observed at pH 6.5 in the absence of salts. The enzyme showed remarkable stability in a wide range of experimental conditions, such as broad pH stability (from 6.0–11.0 after 30 min incubation in buffers at a certain pH), stability in the presence of NaCl up to 3.0 M for 24 h, it retained 80 % of the initial activity after 1 h incubation at 45 °C, and 65 % of the initial activity after 24 h incubation in 30 % dimethyl sulfoxide.  相似文献

Congestion, pollution, security, parking, noise, and many other problems derived from vehicular traffic are present every day in most cities around the world. The growing number of traffic lights that control the vehicular flow requires a complex scheduling, and hence, automatic systems are indispensable nowadays for optimally tackling this task. In this work, we propose a Swarm Intelligence approach to find successful cycle programs of traffic lights. Using a microscopic traffic simulator, the solutions obtained by our algorithm are evaluated in the context of two large and heterogeneous metropolitan areas located in the cities of Málaga and Sevilla (in Spain). In comparison with cycle programs predefined by experts (close to real ones), our proposal obtains significant profits in terms of two main indicators: the number of vehicles that reach their destinations on time and the global trip time.  相似文献

ABS is an engineering plastic that has butadiene part uniformly distributed over the acrylonitrile-styrene matrix. It possesses excellent toughness, good dimensional stability, easy processing ability, chemical resistance, and cheapness. However, it suffers from inherent shortcomings in terms of mechanical strength and vulnerability to environmental conditions. Furthermore, it is non-conducting and easily fretted. Plating on ABS can serve to enhance the strength and structural integrity as well as to improve durability and thermal resistance resulting in metallic properties on the ABS material. ABS is described as the most suitable candidate for plating because it is possible to deposit an adherent metal coating on it by only the use of chemical pretreatment process and without the use of any mechanical abrasion. This article aims to review the history of ABS plastics, properties of ABS, processes and mechanisms of plating, and studies of plating on ABS involving mainly eco-friendly methods of plating by discussing the literature published in recent years. The details of electroplating of ABS carried out in the authors’ laboratory are also presented.  相似文献

Performance and stability of five cermet-supported button-type solid oxide fuel cells featuring a bi-layered electrolyte (SSZ/SDC), an SSC cathode, and a Ni-SSZ anode, were analyzed using polarization curves, impedance spectroscopy, and post-mortem SEM observation. The cell performance degradation at 650 °C in H2/air both with and without DC bias conditions was manifested primarily as an increase in polarization resistance, approximately at a rate of 2.3 mΩ cm2 h−1 at OCV, suggesting a decrease in electrochemical kinetics as the main phenomenon responsible for the performance decay. In addition, the initial series resistance was about ten times higher than the calculated resistance corresponding to the electrolyte, reflecting a possible inter-reaction between the electrolyte layers that occurred during the sintering stage. In situ and ex situ sintered cathodes showed no obvious difference in cell performance or decay rate. The stability of the cells with and without electrical load was also investigated and no significant influence of DC bias was recorded. Based on the experimental results presented, we preliminarily attribute the performance degradation to electrochemical and microstructural degradation of the cathode.  相似文献
