Gastrointestinal nematodes are a global cause of disease and death in humans, wildlife and livestock. Livestock infection has historically been controlled with anthelmintic drugs, but the development of resistance means that alternative controls are needed. The most promising alternatives are vaccination, nutritional supplementation and selective breeding, all of which act by enhancing the immune response. Currently, control planning is hampered by reliance on the faecal egg count (FEC), which suffers from low accuracy and a nonlinear and indirect relationship with infection intensity and host immune responses. We address this gap by using extensive parasitological, immunological and genetic data on the sheep–Teladorsagia circumcincta interaction to create an immunologically explicit model of infection dynamics in a sheep flock that links host genetic variation with variation in the two key immune responses to predict the observed parasitological measures. Using our model, we show that the immune responses are highly heritable and by comparing selective breeding based on low FECs versus high plasma IgA responses, we show that the immune markers are a much improved measure of host resistance. In summary, we have created a model of host–parasite infections that explicitly captures the development of the adaptive immune response and show that by integrating genetic, immunological and parasitological understanding we can identify new immune-based markers for diagnosis and control. 相似文献

A study was conducted to examine how women with spinal cord injury (SCI) perceive the stressors they encounter, and how cognitive appraisal is associated with coping and life satisfaction. Fifty women with SCI were interviewed regarding their experience with stress and coping. The interviews were then coded using a framework based on stress and coping theory (transactional model). The systematic application of quantitative methods to the coded interview data allowed for statistical analyses, which demonstrated that the context in which the women spoke about their experiences with various stressors was associated with coping strategies, time since injury, and life satisfaction. Although stress and coping are commonly seen as interacting constructs that influence quality of life outcomes, current findings suggest that appraisal of stressors in the context of loss (more common among women more recently injured) may have a direct impact on life satisfaction apart from any coping strategies put into effect. Interventions designed to facilitate coping with loss and enhance problem-solving skills, along with education about available resources, may foster a sense of empowerment after SCI and ultimately change how stressors are perceived and managed in order to dampen their negative impact on life satisfaction. The influence of different stratosphere parameters on the degradation of a multilayer film was investigated. The selected multilayer was a three polymeric layers film, a polyamide 6 film inserted between two poly(ethylene terephthalate) (PET) films. Samples were exposed for several ageing under ultraviolet radiations (filtered at 270 nm), varying the atmosphere at 55 mbar pressure (atm, atm + ozone, N2, and T = ?55 °C or +23 °C). Evolution of it mechanical properties defined by uniaxial tractions, thermo‐optical properties defined by spectrophotometry UV–vis‐NIR, chemical properties defined by FTIR‐ATR, and thermal and dielectric properties defined, respectively, by differential scanning calorimetry (DSC) and dynamical dielectric spectroscopy (DDS), were investigated. Our results showed that UV irradiation causes multilayer films degradations, that is, principally decrease of UV transmittance and stress and strain at break (?50%). An increase of the ageing temperature causes an acceleration of these degradations. Degradations principally occur on the PET side of the multilayer exposed to UV radiation. Moreover, the DDS analysis shows a plasticization effect of the primary mode in the polyamide 6 due to photo‐oxidation. Oxygen diffusion is the principal element for this plasticization, indeed it not occurs in a nonoxidative environment (nitrogen), or at low ageing temperature (?55 °C). © 2016 Wiley Periodicals, Inc. Bone injuries represent a major social and financial impairment, commonly requiring surgical intervention due to a limited healing capacity of the tissue, particularly regarding critical-sized defects and non-union fractures. Regenerative medicine with the application of bone implants has been developing in the past decades towards the manufacturing of appropriate devices. This work intended to evaluate medical 316L stainless steel (SS)-based devices covered by a polymer poly (L-lactic acid) (PLLA) coating for bone lesion mechanical and functional support. SS316L devices were subjected to a previously described silanization process, following a three-layer PLLA film coating. Devices were further characterized and evaluated towards their cytocompatibility and osteogenic potential using human dental pulp stem cells, and biocompatibility via subcutaneous implantation in a rat animal model. Results demonstrated PLLA-SS316L devices to present superior in vitro and in vivo outcomes and suggested the PLLA coating to provide osteo-inductive properties to the device. Overall, this work represents a preliminary study on PLLA-SS316L devices’ potential towards bone tissue regenerative techniques, showing promising outcomes for bone lesion support. 相似文献

Peripheral nerve injuries (PNI) can have several etiologies, such as trauma and iatrogenic interventions, that can lead to the loss of structure and/or function impairment. These changes can cause partial or complete loss of motor and sensory functions, physical disability, and neuropathic pain, which in turn can affect the quality of life. This review aims to revisit the concepts associated with the PNI and the anatomy of the peripheral nerve is detailed to explain the different types of injury. Then, some of the available therapeutic strategies are explained, including surgical methods, pharmacological therapies, and the use of cell-based therapies alone or in combination with biomaterials in the form of tube guides. Nevertheless, even with the various available treatments, it is difficult to achieve a perfect outcome with complete functional recovery. This review aims to enhance the importance of new therapies, especially in severe lesions, to overcome limitations and achieve better outcomes. The urge for new approaches and the understanding of the different methods to evaluate nerve regeneration is fundamental from a One Health perspective. In vitro models followed by in vivo models are very important to be able to translate the achievements to human medicine. 相似文献

Multiwall carbon nanotubes were dispersed in a styrene–butadiene copolymer. The effect of nanotube concentration on the tensile characteristics of the composites was examined. Electrical properties carried out under uniaxial extension show an increase in resistivity upon gradual stretching. A second stretch performed after total release of the stress was shown to lead to a flat response in resistivity. The present study had 2 major objectives. The 1st was to investigate the validity of a motivational model of couple happiness based on self-determination theory. The 2nd objective was to test the postulated simplex structure and the generalizability of the 6 forms of motivation proposed by this theory. Both members of 63 couples individually completed the Couple Motivation Questionnaire as well as measures of perceived couples' adaptive behaviors and of dyadic happiness. Results revealed that the proposed model of couple happiness was supported through significant path analyses explaining 61% and 55% of the variance of men's and women's relationship happiness, respectively. Empirical support was also provided for the postulated simplex structure. Results highlight the importance of autonomy-driven processes as opposed to controlling and amotivated processes in the development and maintenance of the quality of couples' relationships.
